AIM To screen clinically relevant micro RnAs (mi RnAs) silenced by DnA methylation in human hepatocellular carcinoma(HCC).METHODS Knockdown of DnA methyltransferases (DnMTs) using si RnAs and mi RnA profiling in HCC cell lines were performed to identify DnA hypermethylation-mediated mi RnA downregulation. Confirmation using individual quantitative real-time PCR (qR T-PCR) assays was thenperformed followed by DnA methylation quantification at the promoter of the mi RnA genes. Quantification of DnA methylation and mi RnA expression was then performed in primary HCC tumor samples and related with clinicopathological *** mi RnA profiling after DnMT knockdown in HCC cell lines revealed upregulation of mi R-23, mi R-25 and mi R-183. After q RT-PCR confirmation and Cp G island methylation quantification of these miR nAs in cell lines, further analysis in primary HCC specimens showed that hsa-mi R-183 is hypermethylated in 30% of HCC (n = 40). Expression of mature miR-183 showed an inverse correlation with DnA methylation levels. In HCC cells, DnMT knockdown and 5-aza-2'-deoxycytidine treatment reduced methylation and stimulated expression of mi R-183. In HCC patients, hypermethylation at hsami R-183 promoter significantly correlates with poor survival (log-rank test P = 0.03). DnA methylation analysis in healthy liver, benign liver tumors (hepatocellular adenoma and focal nodular hyperplasia) and their corresponding adjacent tissues showed absence of hypermethylation supporting the notion that aberrant methylation at hsa-miR-183 is specific for the malignant transformation of *** Our data indicate that hypermethylation of hsa-miR-183 is a frequent event in HCC and potentially useful as a novel surrogate diagnostic and prognostic marker.

Calcium-dependent protein kinases （CDPKs） comprise a family of plant serine/threonine protein kinases in which the calcium sensing domain and the kinase effector domain are combined within one molecule. So far, a biological function in abiotic stress signaling has only been reported for few CDPK isoforms, whereas the underlying biochemical mechanism for these CDPKs is still mainly unknown. Here, we show that CPK21 from Arabidopsis thaliana is biochemically activated in vivo in response to hyperosmotic stress. Loss-of-function seedlings of cpk21 are more tolerant to hyperosmotic stress and mutant plants show increased stress responses with respect to marker gene expression and metabolite accumulation. In transgenic Arabidopsis complementation lines in the cpk21 mutant background, in which either CPK21 wildtype, or a full-length enzyme variant carrying an amino-acid substitution were stably expressed, stress responsitivity was restored by CPK21 but not with the kinase inactive variant. The biochemical characterization of in planta synthesized and purified CPK21 protein revealed that within the calcium-binding domain, n-terminal EF1- and EF2-motifs compared to C-terminal EF3- and EF4-motifs differ in their contribution to calcium-regulated kinase activity, suggesting a crucial role for the n-terminal EF-hand pair. Our data provide evidence for CPK21 contributing in abiotic stress signaling and suggest that the n-terminal EF-hand pair is a calcium-sensing determinant controlling specificity of CPK21 function.

Loss of Aβ-nerve endings associated with the Merkel cell-neurite complex in the lesional oral mucosa epithelium of lichen planus and hyperkeratosis

The Merkel cell-neurite complex initiates the perception of touch and mediates Ap slowly adapting type I responses. Lichen planus is a chronic inflammatory autoimmune disease with T-cell-mediated inflammation, whereas hyperkeratosis is characterized with or without epithelial dysplasia in the oral mucosa. To determine the effects of lichen planus and hyperkeratosis on the Merkel cell-neurite complex, healthy oral mucosal epithelium and lesional oral mucosal epithelium of lichen planus and hyperkeratosis patients were stained by immunohistochemistry （the avidin-biotin-peroxidase complex and double immunofluorescence methods）using pan cytokeratin, cytokeratin 20 （K20, a Merkel cell marker）, and neurofilament 200 （nF200, a myelinated Aβ- and Aδ-nerve fibre marker） antibodies. n F200-immunoreactive （ir） nerve fibres in healthy tissues and in the lesional oral mucosa epithelium of lichen planus and hyperkeratosis were counted and statistically analysed. In the healthy oral mucosa, K20-positive Merkel cells with and without close association to the intraepithelial nF200-ir nerve fibres were detected. In the lesional oral mucosa of lichen planus and hyperkeratosis patients, extremely rare nF200-ir nerve fibres were detected only in the lamina propria. Compared with healthy tissues, lichen planus and hyperkeratosis tissues had significantly decreased numbers of nF200-ir nerve fibres in the oral mucosal epithelium. Lichen planus and hyperkeratosis were associated with the absence of Aβ-nerve endings in the oral mucosal epithelium. Thus, we conclude that mechanosensation mediated by the Merkel cell-neurite complex in the oral mucosal epithelium is impaired in lichen planus and hvperkeratosis.

Western Kenya suffers a highly endemic and also very heterogeneous epidemic of human immunodeficiency virus(HIV).Although female sex workers(FSW)and their male clients are known to be at high risk for HIV,HIV prevalence across regions inWestern Kenya is not strongly correlated with the fraction of women engaged in commercial *** agentbased network model of HIV transmission,geographically stratified at the county level,was fit to the HIV epidemic,scale-up of interventions,and populations of FSW in Western Kenya under two assumptions about the potential mobility of FSW *** the first,all clients were assumed to be resident in the same geographies as their interactions with *** the second,some clients were considered non-resident and engaged only in interactions with FSW,but not in longer-term non-FSW partnerships in these *** both assumptions,the model successfully reconciled disparate geographic patterns of FSW and HIV *** patterns in the model suggest a greater role for FSW in local transmission when clients were resident to the counties,with 30.0%of local HIV transmissions attributable to current and former FSW and clients,compared to 21.9%when mobility of clients was ***,the overall epidemic drivers remained similar,with risky behavior in the general population dominating transmission in highprevalence *** modeling suggests that co-location of high-risk populations and generalized epidemics can further amplify the spread of HIV,but that large numbers of formal FSW and clients are not required to observe or mechanistically explain high HIV prevalence in the general population.

BACKGROUnD Malnutrition is prevalent in inflammatory bowel disease (IBD). Multiple nutrition screening (nST) and assessment tools (nAT) have been developed for general populations, but the evidence in patients with IBD remains unclear. AIM To systematically review the prevalence of abnormalities on nSTs and nATs, whether nSTs are associated with nATs, and whether they predict clinical outcomes in patients with IBD. METHODS Comprehensive searches performed in Medline, CInAHL Plus and PubMed. Included: English language studies correlating nSTs with nATs or nSTs/nATs with clinical outcomes in IBD. Excluded: Review articles/case studies;use of body mass index/laboratory values as sole nST/nAT;agend 1618 patients were included, 72% Crohn’s disease and 28% ulcerative colitis. Four nSTs (the Malnutrition Universal Screening Tool, Malnutrition Inflammation Risk Tool (MIRT), Saskatchewan Inflammatory Bowel Disease nutrition Risk Tool (SaskIBD-nRT) and nutrition Risk Screening 2002 (nRS-2002) were significantly associated with nutritional assessment measures of sarcopenia and the Subjective Global Assessment (SGA). Three nSTs (MIRT, nRS-2002 and nutritional Risk Index) were associated with clinical outcomes including hospitalizations, need for surgery, disease flares, and length of stay (LOS). Sarcopenia was the most commonly evaluated nAT associated with outcomes including the need for surgery and post-operative complications. The SGA was not associated with clinical outcomes aside from LOS. COnCLUSIOn There is limited evidence correlating nSTs, nATs and clinical outcomes in IBD. Although studies support the association of nSTs/nATs with relevant outcomes, the heterogeneity calls for further studies before an optimal tool can be recommended. The nRS-2002, measures of sarcopenia and developments of novel nSTs/nATs, such as the MIRT, represent key, clinically-relevant areas for future exploration.

Third-order non-linearities are important because they allow control over light pulses in ubiquitous high-quality centro-symmetric materials like silicon and *** four-wave mixing provides a direct measure of the third-order non-linear sheet susceptibility x^(3)(where L represents the material thickness)as well as technological possibilities such as optically gated detection and emission of *** picosecond pulses from a free electron laser,we show that silicon doped with P or Bi has a value of x^(3)Lin the THz domain that is higher than that reported for any other material in any wavelength *** immediate implication of our results is the efficient generation of intense coherent THz light via upconversion(also a x^(3)process),and they open the door to exploitation of nondegenerate mixing and optical nonlinearities beyond the perturbative regime.
