Quaternary ammonium compounds(QACs)are a class of antimicrobial disinfectants whose use in cleaning products increased during the COVID-19 ***,their low vapor pressure indicates a proclivity to persist on surfaces,and their presence suggests a level of protection against *** widespread application of QACs in response to the SARS CoV-2 virus created a need to evaluate their longevity on surfaces,for both efficacy and possible health *** are however,no standardized analytical methods for QAC surface sampling and analysis,and no published studies quantifying their concentrations on mass transportation vehicles-a high occupancy,close-contact microenvironment documented to facilitate the spread the SARS CoV-2 ***,we describe a robust liquid chromatography mass spectrometry(LC-MS)method for the analysis of QACs and simultaneous development of a direct surface sampling and extraction *** demonstrate the applicability of the method through the analysis of surface samples collected from in-service public transportation *** rapid,sensitive LC-MS method included 8 target QACs quantified on a Q-Exactive HF Hybrid Quadrupole-Orbitrap mass spectrometer using an electrospray ionization source and Dionex UltiMate 3000 UHpLC system for analyte *** standard mixtures at concentrations between 0.1 ng mL^(-1)and 2000 ng mL^(-1)were analyzed,and chromatographic separation of all analytes was achieved in less than 10 *** correlation coefficients were reported at r>0.986,and LODs ranged from 0.007 to 2.103 ng mL^(-1)for all compounds,confirming the method's sensitivity.A previously reported surface sampling and extraction protocol was modified to further simplify the procedure and expand the number of target *** new sampling protocol was optimized from 10 commercially available wipes and 4 solvent types by quantifying recovery from the ***-Aid brand small gauze pads saturated with isopropanol had

本文运用固定效应模型,分析近十年美国公立世界一流大学的经费支出数据。研究发现:美国一流公立大学中教学科研经费是主要支出;经费总支出与一流大学的排名没有显著相关性,但部分经费支出对大学排名有显著影响。对于无医院服务支出的高校,公共服务支出与大学排名显著正相关;对于有医院服务支出的高校,辅助经营支出的增加有助于大学排名的提升,公共服务支出的影响却相反。因此,提高经费使用效率比增加总额更为重要。我国“双一流”建设高校应该革新经费支出配置理念,建立科学有效的经费配置体系,增大学校各类服务支出。

Gestational heat stress alters skeletal muscle gene expression profiles and vascularity in fetal pigs in a sexually dimorphic manner

Background: There is evidence that sow heat stress(HS) during gestation affects fetal development with implications for impaired muscle growth. We have previously demonstrated that maternal HS during early to midgestation compromised muscle fibre hyperplasia in developing fetal pigs. Thus, we hypothesised these phenotypic changes are associated with a change in expression of genes regulating fetal skeletal muscle development and metabolism. To test this, at d 60 of gestation, RNA sequencing and immunohistochemistry were performed on fetal longissimus dorsi(LD) muscle biopsies collected from pregnant gilts that had experienced either thermoneutral control(CON, 20 ℃, n = 7 gilts, 18 LD samples) or controlled HS(cyclic 28 to 33 ℃, n = 8 gilts, 23 LD samples)conditions for 3 ***: A total of 282 genes were differentially expressed between the HS and CON groups in female LD muscles(false discovery rate(FDR) ≤ 0.05), whereas no differentially expressed genes were detected in male LD muscles between the two groups(FDR > 0.05). Gestational HS increased the expression of genes associated with transcription corepressor activity, adipogenesis cascades, negative regulation of angiogenesis and pro-inflammatory signalling in female LD muscles. Immunohistochemical analyses revealed a decreased muscle vascularity density in fetuses from HS group for both sexes compared to those from the CON group(p = 0.004).Conclusions: These results reveal gilt HS during early to mid-gestation altered gene expression profiles in fetal LD muscles in a sexually dimorphic manner. The molecular responses, including transcription and angiogenesis repressions and enhanced adipogenesis cascades, were exclusively observed in females. However, the associated reductions in muscle vascularity were observed independently of sexes. Collectively this may indicate female fetal pigs are more adaptive to gestational HS in terms of gene expression changes, and/or there may be sexually dimorphic differences

Tyrosine kinase receptor B isoforms alter App and BACE1 endogenous levels independently of BDNF

Brain derived neurotrophic factor (BDNF) levels and signaling via the tyrosine receptor kinase B (TrkB) have been shown to be altered in Alzheimer’s Disease. In addition, it has been reported that the isoforms of TrkB can differentially affect metabolism of amyloid precursor protein (App). Conversely, Ab, a neurotoxic cleavage product of App, has been shown to impair TrkB/ BDNF signaling. Therefore, we investigated whether the changes observed in App metabolism were due to the isoform-specific effects of TrkB on either App expression, and/or on the expression and activity of ADAM10 and BACE1. Since BDNF levels are decreased in AD, we focused on BDNF independent effects of the TrkB isoforms. We found that TrkB FL increases endogenous App levels in both HEK293 and SH-SY5Y naive cells. We did not find an increase in ADAM10 activity in HEK293 cells, but an increase in BACE1 levels. Additionally, we have found that TrkB FL is able to increase NFAT3 mediated transcriptional activity and we suggest that this causes transcriptional activation of the BACE1 promoter.

A retrospective cohort study to examine the association between the persistence of abdominal pain after cholecystectomy and ejection fraction on HIDA scan in patients with biliary dyskinesia

Background and aims:The persistence of pain in patients who underwent cholecystectomy primarily for biliary dyskinesia has been associated with gall bladder ejection fraction(GBEF)found on hepatobiliary iminodiacetic acid(HIDA)*** is a definitive treatment for cholelithiasis but is questionable in biliary *** the postoperative follow-up,gastroenterologists and surgeons often found that some patients with biliary dyskinesia continue to experience biliary pain even after *** study aims to investigate whether the value of GBEF found on the HIDA scan predicts the persistence of pain after cholecystectomy in patients with biliary dyskinesia(EFpective cohort study conducted at Kern Medical by recruiting patients from November 2019 to October 2022 in consecutive *** is a single sample study consisting of post-cholecystectomy biliary dyskinesia patients with dichotomous outcomes,i.e.,persistent typical recurrent epigastric or right upper quadrant pain with/without meals versus no *** primary outcome was the presence of pain 30 days after the *** analysis was performed with some of the bivariate indicators and continuous predictors,which contained fewer missing elements and had more ***:Univariate logistic regression suggested that HIDA GBEF as a continuous variable does not suggest a relationship with pain after surgery(p=0.3951).A ROC analysis suggested a cutoff of HIDA GBEF=16(AUC=0.60,Specificity=0.5455,Sensitivity=0.7333).Conclusion:Our study does not support the relationship between GBEF found on HIDA scan to improved outcomes after the *** larger controlled trials comparing outcomes of the patients with biliary dyskinesia who did and didn't undergo cholecystectomy are needed to look for other factors.
