Noninvasive diagnosis of bone density and mechanical properties using non-radiation imaging modality is an emerging area with promising in early prediction of osteopenia and treatment effectiveness in the clinic and functional disuse,i.e.,long-term bedrest and space *** in quantitative ultrasound have shown advantages in measuring both bone density and mechanical strength,non-radiation,imaging capability,and easy to *** challenge that remained is the poor penetration of ultrasound signals passing through trabecular and cortical bones and acoustic energy scattering.A new scanning confocal ultrasound technology is developed in this lab to detect the alteration of bone to provide diagnostic results in bone density and structure properties.A software-controlled flexible ultrasound system with 2-D dual array transducer is developed and proposed for the purpose of noninvasive bone density diagnosis and assessment of bone ***(Tx)transducer elements are divided into sub-blocks to excite the ultrasound signals in sequence to decrease the system complexity while maintaining beam pattern properties by the signal processing procedure at receiving(Rx)*** is also applied to reduce acoustic side lobes and to make the resolution in the ultrasound field of view(FOV)more *** study may provide basic understanding of modulated confocal ultrasound beam forming for tissue characterization,such as trabecular bone structual and strength properties.

Drought Stress and Preharvest Aflatoxin Contamination in Agricultural Commodity: Genetics, Genomics and Proteomics

Throughout the world, aflatoxin contamination is considered one of the most serious food safety issues concerning health. Chronic problems with preharvest afiatoxin contamination occur in the southern US, and are particularly troublesome in corn, peanut, cottonseed, and tree nuts. Drought stress is a major factor to contribute to preharvest afiatoxin contamination. Recent studies have demonstrated higher concentration of defense or stress-related proteins in corn kerners of resistant genotypes compared with susceptible genotypes, suggesting that preharvest field condition （drought or not drought） influences gene expression differently in different genotypes resulting in different levels of ＂end products＂： PR（pathogenesis-related） proteins in the mature kernels. Because of the complexity of Aspergillus-plant interactions, better understanding of the mechanisms of genetic resistance will be needed using genomics and proteomics for crop improvement. Genetic improvement of crop resistance to drought stress is one component and will provide a good perspective on the efficacy of control strategy. Proteomic comparisons of corn kernel proteins between resistant or susceptible genotypes to Aspergillus flavus infection have identified stress-related proteins along with antifungal proteins as associated with kernel resistance. Gene expression studies in developing corn kernels are in agreement with the proteomic studies that defense-related genes could be upregulated or downregulated by abiotic stresses.

Monitoring the Expression of Maize Genes in Developing Kernels under Droug ht Stress usingOligo-microarray

Preharvest aflatoxin contamination of grain grown on the US southeastern Coast Plain is provoked and aggravated by abiotic stress. The primary abiotic stress is drought along with high temperatures. The objectives of the present study were to monitor gene expression in developing kernels in response to drought stress and to identify drought-responsive genes for possible use in germplasm assessment. The maize breeding line Tex6 was used, and gene expression profiles were analyzed in developing kernels under drought stress verses well-watered conditions at the stages of 25, 30, 35, 40, 45 d after pollination （DAP） using the 70 mer maize oligo-arrays. A total of 9 573 positive array spots were detected with unique gene IDs, and 7 988 were common in both stressed and well-watered samples. Expression patterns of some genes in several stress response-associated pathways, including abscisic acid, jasmonic acid and phenylalanine ammonia-lyase, were examined, and these specific genes were responsive to drought stress positively. Real-time quantitative polymerase chain reaction validated microarray expression data. The comparison between Tex6 and B73 revealed that there were significant differences in specific gene expression, patterns and levels. Several defense-related genes had been downregulated, even though some defense-related or drought responsive genes were upregulated at the later stages.

A method for rapid identification and quantification of phthalate plasticizers in beverages was developed.A number of 15 phthalate plasticizers which covered all the phthalates concerned in the US Consumer Product Safety Improvement Act(CPSIA),European Union legislations and Chinese national standards(GB)were *** a combined solid-phase micro-extraction(SPME)and direct analysis in real time mass spectrometry(DART-MS)approach,phthalates at sub-ng•mL^(−1)levels can be qualitatively and quantitatively analyzed in a short *** use of ultrahigh-resolving power and the accurate mass measurement capacity naturally provided by Fourier transform ion cyclotron resonance mass spectrometry(FT-ICR-MS)minimizes the matrix interferences and thus enables the evaluation of phthalates in a complex matrix without extensive sample handlings or *** limits of quantification(LOQs)were estimated to be at 0.3-5.0 ng•mL^(−1),lower than the Maximum Residue Limit(MRL)regulated by the European Union legislations(2007/19/EC)in foods,beverages,food packaging and toys(0.3-30 ng•mL^(−1)).This rapid and easy-to-use SPME-DART-FT-ICR-MS method provided a relatively high-throughput and powerful analytical approach for quick testing and screening phthalates in beverages and water samples to ensure food safety.

Effects of different brush border membrane vesicle isolation protocols on proteomic analysis of Cry1Ac binding proteins from the midgut of Helicoverpa armigera

Brush border membrane vesicles （BBMV） isolated from insect midguts have been widely used to study CrylA binding proteins. Sample preparation is important in two- dimensional electrophoresis （2-DE）, so to determine a suitable BBMV preparation method in Helicoverpa armigera for 2-DE, we compared three published BBMV preparation methods mostly used in sodium dodecyl sulfate-polyacrylamide gel electrophoresis （SDS- PAGE）. All methods yielded similar types and numbers of binding proteins, but in different quantities. The Abdul-Rauf and Ellar protocol was the best of the three, but had limitations. Sufficient protein quantity is important for research involving limited numbers of insects, such as studies of insect resistance to Bacillus thuringiensis in the field. Consequently, we integrated the three BBMV isolation methods into a single protocol that yielded high quantities of BBMV proteins from H. armigera larval midguts, which proved suitable for 2- DE analysis.

A review of spatial targeting methods of payment for ecosystem services

Payments for Ecosystem Services(PES)have been studied extensively over the past decade as an important policy tool for coordinating ecological protection and regional socioeconomic *** of the greatest challenges of PES implementation is to understand where to pay,i.e.,spatial targeting,which can directly impact PES effectiveness and *** this study,we conducted a systematic review of spatial targeting methods based on literature analysis using ***,peer-reviewed articles related to spatial targeting of PES were selected from the Web of Science database based on *** applying PES spatial targeting methods were then chosen and analyzed after all articles were *** total,70%of the chosen cases focused on improving the compensation efficiency of biodiversity or another single environmental objective,whereas the remaining cases focused on coordinating trade-offs between equity and efficiency or multiple environmental *** main PES spatial targeting approaches included cost-benefit analysis,multi-objective optimization,data envelope analysis and other methods aimed at specific *** these,cost-benefit analysis has been most widely applied at different scales,including county,regional and watershed *** differences among the different PES spatial targeting methods were found,including in PES spatial targeting dimensions,efficiency optimization approaches and method application *** practice of PES spatial targeting requires the selection of appropriate methods based on contextual biophysical and socioeconomic conditions as well as relevant environmental *** combined application of PES spatial targeting methods,compensation willingness of stakeholders and dynamic implementation of PES spatial targeting should be considered in future research.

Wnt signaling has emerged as a major regulator of tissue development by governing the self-renewal and maintenance of stem cells in most tissue types. As a key upstream regulator of the Wnt pathway, the transmem- brane E3 ligase ZNRF3 has recently been established to play a role in negative regulation of Wnt signaling by targeting Frizzled （FZD） receptor for ubiquitination and degradation. However, the upstream regulation of ZNRF3, in particular the turnover of ZNRF3, is still unclear. Here we report that ZNRF3 is accumulated in the presence of proteasome inhibitor treatment independent of its E3-ubiquitin ligase activity. Furthermore, the Cullin f-specific SCF complex containing β-TRCP has been identified to directly interact with and ubiqui- tinate ZNRF3 thereby regulating its protein stability. Similar with the degradation of β-catenin by β-TRCP, ZNRF3 is ubiquitinated by β-TRCP in both CKI-phos- phorylation- and degron-dependent manners. Thus, our findings not only identify a novel substrate for β-TRCP oncogenic regulation, but also highlight the dual regu- lation of Wnt signaling by β-TRCP in a context-dependent manner where β-TRCP negatively regulates Wnt signaling by targeting β-catenin, and positively regulates Wnt signaling by targeting ZNRF3.
