snai-1 and Epithelial-Mesenchymal Transition-Related Protein Immunoexpression in Canine Mammary Carcinomas

Epithelial-mesenchymal transition (EMT) is defined as switching of polarized epithelial cells to a migratory fibroblastoid phenotype. EMT is known to be involved in the progression and metastasis of various cancers in humans, but this specific process is still little explored in the veterinary literature. The aim of this research was to evaluate the expression of EMT-related proteins in canine mammary carcinomas (CMCs). The expression of six EMT-related proteins in 94 CMCs of female dogs was evaluated by immunohistochemistry using a tissue array method. Additionally, clinicopathological characteristics were compared with the expression of EMT-related proteins. Loss of epithelial protein and/or acquisition of the expression of mesenchymal proteins were observed in CMCs. Loss of epithelial protein and/or acquisition of the expression of mesenchymal proteins were observed, particularly in tumors with evidence of stromal invasion;however, significance was only observed between the s100A4 and vascular invasion. In addition, snai-1 nuclear immunoexpression was significantly related to E-cadherin loss. In conclusion, loss of epithelial proteins and/or the acquisition of mesenchymal proteins are associated with EMT and may have an important role in the evaluation of CMC patients. The unique immunoexpression pattern of snai-1 could help to distinguish between an adenoma and a non-metastatic carcinoma and seems to be related to conversion of myoepithelial cells to a complete mesenchymal-like phenotype. Loss of E-cadherin and cytokeratin and change of immunoexpression pattern of snai-1, N-cadherin, s100A4 and MMP-2 indicate the occurrence of EMT in canine mammary carcinomas and should result in an en bloc resection or a close follow-up.

Cathepsin D (CD), a lysosomal protease, and s100A4 protein, a calcium binding motif, are considered to be involved in metastasis in various human cancers. No data regarding such proteins are available for canine mammary carcinomas (CMCs). Accordingly, their expression in association with known factors of prognosis was investigated in this study. For that, 66 surgically resected CMCs were submitted to an immunohistochemical evaluation using anti CD, s100A4 protein, HER2, estrogen receptor α, cytokeratin 5, and p63 antibodies, further characterizing the tumors' molecular subtype. An increase in s100A4 immunoexpression by neoplastic luminal mammary cells was associated with an infiltrative tumor mode of growth, consequently leading us to conclude that s100A4 protein could be related to progression in CMCs. Additionally, the occurrence of the luminal A molecular subtype was associated with the complex histotype in CMCs. Although we have demonstrated that changes in s100A4 protein immunoexpression occurs in CMCs, further studies are needed to determine whether this represents important independent biomarkers for CMCs.

Mammary neoplasms are the most common tumors in female dogs. They are usually treated using solely surgical mastectomy—which is recognized as unsatisfactory in many cases. Given this, the benefits of chemotherapy in dogs with mammary cancer need to be further explored. some drugs that can be used for treating canines with mammary tumors may be substrates of uptake and/or efflux transporters such as the ATP-binding cassette (ABC) transporters. Unfortunately, very little is known regarding the pathobiology of such proteins in canine tumors, including mammary cancer. Accordingly, this study was designed to characterize the expression of ABC transporters P-glycoprotein, MRP1, and MRP2 and their relation with clinicopathologic factors in order to allow a better understanding of their influence in canine mammary cancer. P-glycoprotein was expressed in tumors from 55.8% of patients, while MRP1 and MRP2 were expressed in 37.2% and 39.5% of tumors, respectively. P-glycoprotein expression showed to be related with regional lymph node spread (P = 0.0038), as well as with tumor grade (P = 0.0353) and with a shorter survival (P = 0.0245). MRP1 revealed a strong association with a higher histological grade (P survival (P = 0.0002). Additionally, MRP1 was determined as prognostic indicator independent of lymph node status using Cox proportional-hazards regression multivariate analysis (P = 0.0216). No relations between MRP2 and clinicopathologic features were observed. We have found that P-glycoprotein and MRP1 are expressed in highly aggressive canine mammary tumors and are related with poor prognosis. Our results suggest that they may play a significant role in the course of canine mammary cancer progression and be promising candidate markers for a validation study on therapy outcome.

spinal cord injuries have profound detrimental effects on individuals, regardless of whether they are caused by trauma or non-traumatic events. The compromised regeneration of the spinal cord is primarily attributed to damaged neurons, inhibitory molecules, dysfunctional immune response, and glial scarring. Unfortunately, currently, there are no effective treatments available that can fully repair the spinal cord and improve functional outcomes. Nevertheless, numerous pre-clinical approaches have been studied for spinal cord injury recovery, including using biomaterials, cells, drugs, or technological-based strategies. Combinatorial treatments, which target various aspects of spinal cord injury pathophysiology, have been extensively tested in the last decade. These approaches aim to synergistically enhance repair processes by addressing various obstacles faced during spinal cord regeneration. Thus, this review intends to provide scientists and clinicians with an overview of pre-clinical combinatorial approaches that have been developed toward the solution of spinal cord regeneration as well as update the current knowledge about spinal cord injury pathophysiology with an emphasis on the current clinical management.

Numerical modeling of nutrient transport to assess the agricultural impact on the trophic state of reservoirs

Estimating the Lost Economic Production Caused by Internal Displacement Because of Disasters

Disasters are a large cause of internal displacement at the global level, in 2017 alone affecting more than19 million people. This dimension of disaster risk is usually overlooked in many disaster risk management strategies and assessments. It is only very recently that efforts to quantify displacement risk and integrate that dimension into existing physical risk models have been made. A key component of internal displacement assessment is the quantification of its economic impacts, both direct and indirect, not only to make a figure available but to raise awareness and increase accountability among governments, stakeholders, policymakers, and decision *** article presents a novel and peril-agnostic methodology with which to estimate, using a scenario-based approach, the economic production lost because of internal population displacement. The methodology is applied by using as a triggering event the April 2015 Gorkha, Nepal M7.8 earthquake. The method quantifies the indirect cost caused by internal population displacement because of that event in terms of lost economic production and estimates the loss to be between UsD 400 and 850 million.

Elucidating woody community diversity and structure change along environmental gradients is still a central issue of tropical forest ecology. We have evaluated changes in alpha and beta diversity, community composition and structure of woody communities along an elevational gradient in Dwarf Cloud Forests, southeast Brazil. We selected five areas along an elevational gradient(1,300-1,600 m asl), and randomly allocated 10 plots(10 m × 20 m) in each area in the Ibitipoca state Park(IsP), serra da Mantiqueira, southeastern Brazil. All woody species(diameter at breast height ≥ 5.0 cm) were collected for taxonomic identification. Thus, we analysed the woody communities based on a phytosociological approach. Overall, 147 species and 37 families were recorded, distributed in 2,303 individuals. No differences in the species richness pattern were observed between areas using the rarefaction and extrapolation curves. significant differences in species composition and structure between areas were observed. The high beta diversity observed, corroborated by the Jaccard coefficient, increases with decreasing altitude. Our results showed that woody communities in the studied Dwarf Cloud Forests have a higher diversity and structural variability along elevational gradient.

An Input–Output Ex Ante Regional Model to Assess the short-Term Net Effects of the 16 April 2016 Earthquake in Ecuador

The 16 April 2016 earthquake in Ecuador exposed the significant weaknesses concerning the methodological designs to compute—from an economic standpoint—the consequences of a natural hazard-related disaster for productive exchanges and the accumulation of capital in *** study addressed one of these challenges with an innovative ex ante model to measure the partial and net short-term effects of a natural hazard-related catastrophe from an interregional perspective,with the 16 April 2016 earthquake serving as a case *** general,the specified and estimated model follows the approach of the extended Miyazawa model,which endogenizes consumption demand in a standard input–output model with the subnational interrelations and resulting *** to the country’s limitations in its regional account records the input–output matrices for each province of Ecuador had to be estimated,which then allowed transactions carried out between any two sectors within or outside a given province to be identified by means of the RAs *** estimations provide evidence that the net short-term impact on the national accounts was not significant,and under some of the simulated scenarios,based on the official information with respect to earthquake management,the impact may even have had a positive effect on the growth of the national product during 2016.

seasonal influence on cone penetration test: An unsaturated soil site example

Interpretation of electric cone penetration test(CPT) based pore water pressure measurement(CPTu) is well established for soils with behavior that follows classical soil mechanics. The literature on the interpretation of these tests performed on unsaturated tropical soils is limited, and little is known about the influence of soil suction on in situ test data. In this context, the CPT data are presented and discussed to illustrate the seasonal variability in an unsaturated tropical soil site. The test data show that soil suction significantly influenced CPT data up to a depth of 4 m at the study site. It shows the importance of considering seasonal variability in unsaturated soil sites caused by soil suction, which was related to water content through a soil-water retention curve(sWRC). It is also important to consider this aspect in the interpretation of CPT data from these soils.
