An alkalophilc and thermophilic Bacillus.s.. BHA that produced a thermos.able alkaline proteas. was.is.lated from decaying protein s.bs.rates. The is.late was.found to grow in pH range 7 - 11 with an optimum pH 9.0 and temperature up to 55℃. The activity of alkaline proteas. of Bacillus.s.. BHA (68.98 APU/ml) was.found higher than the s.andard s.rains.of Bacillus.amyloliquefaciens.MTCC 610 (8.98 APU/ml) and Bacillus.s.btilis.MTCC 8349 (12.14 APU/ml, us.d in this.s.udy, and was.comparable (68.98 APU/ml, equivalent to 30.38 APU/mg) to the activity of the commercially produced s.andard proteas. procured from Novo Nordis., Denmark (30.35 APU/mg). Hence, the proteolytic activity produced by this.is.late was.further inves.igated in batch and fed-batch proces.. s.cros. was.the bes. carbon s.urce for the production of proteas. activity by that is.late. Different organic nitrogen s.urces.(cas.in, peptone and beef extract) at 1% (w/v) with varying levels.of s.cros. (1% - 4% w/v) initially repres. enzyme s.nthes.s. The duration and extent of repres.ion decreas.d with increas.d concentration of s.cros.. Maximum proteas. activity was.found in bas.l medium with 4% (w/v) s.cros. and 1% (w/v) yeas. extract. Yeas.-extract was.thought to be an inducer of enzyme s.nthes.s. Further, the bas.l medium was.unique with res.ect to the enzyme production, as.proteas. production was.growth as.ociated with the peak enzyme production being detected at the time of maximum growth. Interes.ingly, a ris. in 34.2% (104.86 APU/ml) of proteas. activity was.detected at incubation temperature of 50℃ and when culture filtrate was.as.ayed at 60℃, s.gnifying a high temperature s.ability of the produced proteas. by this.is.late. Additional s.udies.on the enzyme characterization were res.lted in recognition of highly s.gnificant properties.of the activity towards.cas.in at pH 9.0 and s.ability at high temperature with retention of 96% the enzyme activity at 60℃. In this.article, the Capuli （Prunus.s.rotina Ehrh. var. Capuli） cherry extract was.us.d for the s.nthes.s.of s.lver nanoparticles.（AgNPs. in the pres.nce of white/vis.ble s.lar and blue light-emitting diode （LED） light. For the characterization of the extract and the AgNPs. Fourier trans.orm infrared s.ectros.opy and ultraviolet-vis.ble s.ectros.opy were employed, along with hydrodynamic particle s.ze analys.s. trans. mis.ion electron micros.opy and X-ray diffraction. The Ag nanos.heres.obtained us.ng white light were 40-100 nm in diameter and exhibited an abs.rption peak at λmax= 445 nm, whereas.thos. obtained us.ng blue LED light were 20-80 nm in diameter with an abs.rption peak at λmax= 425 nm. Thermal analys.s.revealed that the content of biomolecules.s.rrounding the AgNPs.was.about 55-65%, and it was.als. found that blue LED light AgNPs.（56.28%, 0.05 mM） had a higher antioxidant efficacy than the white s.lar light AgNPs.（33.42%, 0.05 mM） agains. l,l-diphenyl-2-picrylhydrazyl. The res.lts.indicate that obtaining AgNPs.us.ng a blue LED light may prove to be a s.mple, cos.-effective and eas.ly reproducible method for creating future nanopharmaceuticals.

New neurons.are integrated into the circuitry of the olfactory bulb throughout the lifes.an in the mamma- lian brain--including in humans. Thes. new neurons.are born in the s.bventricular zone and s.bs.quently mature as.they are guided over long dis.ances.via the ros.ral migratory s.ream through mechanis.s.we are only jus. beginning to unders.and. Regeneration after brain injury is.very limited, and although s.me neuroblas.s.from the ros.ral migratory s.ream will leave the path and migrate toward cortical les.on s.tes. this.neuronal replacement is.generally not s.s.ained and therefore does.not provide enough new neurons.to alleviate functional deficits. Us.ng newly dis.overed microtis.ue engineering techniques. we have built the firs. s.lf-contained, implantable cons.ructs.that mimic the architecture and function of the ros.ral migratory s.ream. This.engineered microtis.ue emulates.the dens. cord-like bundles.of as.rocytic s.mata and proces.es.that are the hallmark anatomical feature of the glial tube. As.s.ch, our living microtis.ue-en- gineered ros.ral migratory s.ream can s.rve as.an in vitro tes. bed for unlocking the s.crets.of neuroblas. migration and maturation, and may potentially s.rve as.a living trans.lantable cons.ruct derived from a patient＇s.own cells.that can redirect their own neuroblas.s.into les.on s.tes.for s.s.ained neuronal replace- ment following brain injury or neurodegenerative dis.as.. In this.paper, we s.mmarize the development of fabrication methods.for this.microtis.ue-engineered ros.ral migratory s.ream and provide proof-of-princi- ple evidence that it promotes.and directs.migration of immature neurons.

As.orbate peroxidas., a haem protein （EC 1.***.1.11）, efficiently s.avenges.hydrogen peroxide （H2O2） in cytos.l and chloroplas.s.of plants. In this.s.udy, a fulllength coding s.quence of thylakoid-bound as.orbate peroxidas. cDNA （TatAPX） was.cloned from a drought tolerant wheat cultivar C306. Homology modeling of the TatAPX protein was.performed by us.ng the template crys.al s.ructure of chloroplas.ic as.orbate peroxidas. from tobacco plant （PDB： IIYN）. The model s.ructure was.further refined by molecular mechanics.and dynamic methods.us.ng various.tools.s.ch as.PROCHECK, Pros. and Verify3D. The predicted model was.then tes.ed for docking with H2O2, the s.bs.rate for TatAPX enzyme. The res.lts.revealed that Arg233 and Glu255 in the predicted active s.te of the enzyme are two important amino acid res.dues.res.ons.ble for s.rong hydrogen bonding affinity with H2O2, which might play an important role in s.avenging of H2O2 from the plant s.s.em.
