Non-coding RNAs in exercise immunology:A systematic review

Regular physical exercise has been recognized as a potent modulator of immune function,with its effects including enhanced immune surveillance,reduced inflammation,and improved overall health.While strong evidence exists that physical exercise affects the specific expression and activity of non-coding RNAs(ncRNAs)also involved in immune system regulation,heterogeneity in individual study designs and analyzed exercise protocols exists,and a condensed list of functional,exercise-dependent ncRNAs with known targets in the immune system is missing from the literature.A systematic review and qualitative analysis was used to identify and categorize ncRNAs participating in immune modulation by physical exercise.Two combined approaches were used:(a)a systematic literature search for“ncRNA and exercise immunology”,(b)and a database search for microRNAs(miRNAs)(miRTarbase and DIANA-Tarbase v8)aligned with known target genes in the immune system based on the Reactome database,combined with a systematic literature search for“ncRNA and exercise”.Literature searches were based on PubMed,Web of Science,and SPORTDiscus;and miRNA databases were filtered for targets validated by in vitro experimental data.Studies were eligible if they reported on exercise-based interventions in healthy humans.After duplicate removal,95 studies were included reporting on 164 miRNAs,which were used for the qualitative synthesis.Six studies reporting on long-noncoding RNAs(lncRNAs)or circular RNAs were also identified.Results were analyzed using ordering tables that included exercise modality(endurance/resistance exercise),acute or chronic interventions,as well as the consistency in reported change between studies.Evaluation criteria were defined as“validated”with 100%of≥3 independent studies showing identical direction of regulation,“plausible”(≥80%),or“suggestive”(≥70%).For resistance exercise,upregulation of miR-206 was validated while downregulation of miR-133a appeared plausible.For endurance exercise,15 miRNAs were categorized as validated,wit

Creation of stable intrinsically anisotropic self-bound states with embedded vorticity is a challenging issue.Previously,no such states in bose−Einstein condensates(bECs)or other physical settings were known.Dipolar bEC suggests a unique possibility to predict stable two dimensional anisotropic vortex quantum droplets(2D-AVQDs).We demonstrate that they can be created with the vortex axis oriented perpendicular to the polarization of dipoles.The stability area and characteristics of the 2D-AVQDs in the parameter space are revealed by means of analytical and numerical methods.Further,the rotation of the polarizing magnetic field is considered,and the largest angular velocities,up to which spinning 2D-AVQDs can follow the rotation in clockwise and anti-clockwise directions,are found.Collisions between moving 2D-AVQDs are studied too,demonstrating formation of bound states with a vortex−antivortex−vortex structure.A stability domain for such stationary bound states is identified.Unstable dipolar states,that can be readily implemented by means of phase imprinting,quickly transform into robust 2D-AVQDs,which suggests a straightforward possibility for the creation of these states in the experiment.

The role of wettability,often characterized by contact angle(θ),in two-phase immiscible phases displacement is not well understood.In this study,the color gradient lattice boltzmann method(LbM),capable of maintaining the prescribedθ(from 0°to 180°at intervals of 10°)throughout the numerical simulations,was used to investigate the displacement patterns and displacement efficiency in a 2D porous medium.The capillary numbers(Ca)used were 0.01,1,and 100,and the viscosity ratios(M)used were 0.1,1,and 10.At M=10,the saturation(S)had a bilinear relationship withθ,while for M=0.1 and 1,the S-θrelationships were complicated by Ca.A saturation contour in the M-Ca-θspace was proposed to demonstrate the movement of a traditional 2D M-Ca phase diagram withθincrements.The value of S continued to increase after the breakthrough,and the final saturation(0.997)for the hydrophilic condition(θ=10°)was higher than that(0.673)for the hydrophobic condition(θ=170°).

Impact hydroforming(IHF),as a novel sheet metal forming technology with the advantages of high strain rate forming and flexible liquid loading,is highly suitable for efficiently manufacturing aluminum complex-shaped sheet parts.In this paper,deformation characteristics of complex sheet parts under IHF are systematically investigated.The mechanical properties of 2024 aluminum alloy under a wide range of strain rates(10-3 s-1–3.3×103 s-1)were studied.It indicated that the elongation of 2024 aluminum alloy was improved by 116.01%under strain rates of 3.306-×-103 s-1,referring to 10-3 s-1.Further,a complex-shaped part with symmetrical and asymmetrical structures was selected.The deformation characteristics of sheet and role of inertial effect under IHF were investigated with well-developed solid–liquid coupling finite element(SLC-FE)model with high accuracy.Differentiating deformation tendency is found for symmetrical structure with notably prior deformation at central zone,showing a“bulging”profile at initial forming stage.Whereas,synchronous deformation is presented for asymmetrical structure with a“flat”profile.Additionally,distinctive inertial effect was observed at different positions change for both symmetrical and asymmetrical structures,in which lower values were resulted at their central regions.Meanwhile,the inertial effect evolved with the impacting speed.Specially,larger difference of inertial effect was observed with increasing impacting speed.

The retinoblastoma gene product(pRb)is a chromatin-associated protein that can either suppress or promote activity of key regulators of tissue-specific differentiation.We found that twelve weeks after transfection of the exogenous active(Δb/X andΔр34)or inactive(ΔS/N)forms of Rb into the 10T1/2 mesenchymal stem cells and clonal selection not a single cell line did contain exogenous Rb,despite being G-418 resistant.However,the consequences of the transient production of exogenous Rb had different effects on the cell fate.TheΔb/X andΔр34 cells transfected with active form of Rb showed elevated levels of inducible adipocyte differentiation(AD).On the contrary,theΔS/N cells transfected with inactive Rb mutant were insensitive to induction of AD associated with abolishing of expression of the PPARγ2.Additionally,the PPARγ2 promoter in undifferentiatedΔS/N cells was hypermethylated,but all except−60 position CpG became mostly demethylated after cells exposure to AD.We conclude that while transient expression of inactive exogenous Rb induces long term epigenetic alterations that prevent adipogenesis,production of active exogenous Rbs results in an AD-promoting epigenetic state.These results indicate that pRb is involved in the establishment of hereditary epigenetic memory at least by creating a methylation pattern of PPARγ2.

Monitoring lime and cement improvement using spectral induced polarization and bender element techniques

Spectral induced polarization(SIP)and bender element(bE)techniques show a high sensitivity to particle size,particle distribution and content of generated hydration products,which essentially govern the efficiency of ground improvement.In this context,both SIP and bE were integrated on a column setup to monitor the processes of lime and cement stabilization.A 5 mmol/L Na2CO3 solution was injected into the sand-lime mixture to produce CaCO3 precipitation,while deionized water was injected into the sandcement mixture to induce the hydration of cement.The average diameters of the precipitated particles or clusters were calculated from the relaxation time,which was a significant parameter of SIP signals,via the Schwarz equation.Two pairs of bE were used to demonstrate the heterogeneity of the product precipitation,which was probably caused by the location of the inflow and outflow on the SIP-bE column.SIP and bE showed the capability of nondestructively monitoring the spatiotemporal chemical evolution processes,which could be helpful for engineering applications.

The genes referred to as ontogenes are responsible for conditional mutations. based on the results of the research of conditional mutations in D. melanogaster, we attempt to figure out the biological role of ontogenes. We conclude that ontogenes in the process of individual development control the construction of the living organisms of cells (cellular construction), which comprises the induction of cell division, determination of division plane, and the location of daughter cells after the division is completed. The process of morphogenesis consists of cellular construction and protein synthesis. Protein synthesis is controlled by protein-coding (Mendelian) genes. Mendelian genes are switched on by ontogenes. In terms of the two-component genome composed of Mendelian genes and ontogenes, we consider 1) the concept of biological character;2) interspecific incompatibility;3) ontogenesis;4) phylogenesis;and 5) mutagenesis. Ontogenes, which control cellular construction, possess the specific features unusual for Mendelian genes, namely, 1) the activity in germ line tissue;2) remote interaction;and 3) activity in a compacted state (heterochromatization). These specific features of ontogenes suggest that unlike the Mendelian genes with their chemical activity, ontogenes possess another type of activity (biophysical) involving induction of an electromagnetic field.

The conditional mutations in drosophila were obtained by γ-irradiation and selected using the test for dominant lethality. The conditional mutations survive under permissive genetic conditions and, additionally, commence to display novel properties. One of such properties is a recessive lethality. Ten conditional mutations that displayed recessive lethality were mapped with the help of a standard set of deletions. Half mutations contained two and more lethal defects. The fact that a large number of the lethal defects are associated with one mutation suggests that γ-irradiation is the most unlikely cause of the defects. One of the conditional mutations carried four lethal regions and had a Small barrel (Smba) visual phenotype. The Smba phenotype in the Smba/ In(2LR) Cy strain is inherited according to a parental type and disappears in the Smba/In(2LR) Pm strain. Lethality in two of the four lethal regions also disappears in this strain. A separate experiment was conducted to clarify how these regions lost a lethal manifestation after the In(2LR) Cy chromosome in the Smba/In(2LR) Cy strain was replaced with the In(2LR) Pm chromosome. The process of disappearance of the Smba phenotype was also observed in three Smba/In(2LR) Cy substocks. These data suggest that the regions of multiple recessive lethality emerge in a secondary manner under the effect of the earlier formed radiation-induced mutation in ontogene. It is assumed that the recessive lethal regions are the ontogenes with an altered DNA conformation. The conformation in ontogenes is changed in the germline cells during a regular “editing” of the individual development program.

A mutation in an ontogene acts as a conditional dominant lethal: it is lethal in a certain genotype but not lethal in another. In total, 30 mutations of this type residing in the Drosophila melanogaster X chromosome have been assayed for their ability to cause meiotic nondisjunction. The level of X nondisjunction in the females heterozygous for the mutation in ontogene appears to be very high. The share of matroclinous daughters reaches 24.7% of the overall offspring and of patroclinous males, 24.9%. Neither inversion in the opposite X chromosome nor additional Y chromosome has any effect on the X nondisjunction. The balance of the XX and X0 egg cells is disturbed: exceptional daughters are prevalent in the offspring of the females with a normal opposite X chromosome and exceptional sons, in the offspring of the females with an inverted X chromosome. In addition, 12% of the matroclinous daughters of the females with a normal opposite X chromosome are homozygous for the marker of one of the maternal X chromosomes (“equational” nondisjunction). A “fading” parental effect of the mutation in ontogene on the X chromosome nondisjunction is also observed. Under experimental conditions, the mutant ontogenes reside in meiotic densely compacted X chromosomes. We infer that the ontogenes are DNA regions with controlled compaction. It is postulated that the genetic activity of ontogenes is determined by this compaction and has a biophysical (electromagnetic) nature. In a meiotic cell, ontogenes induce physical fields providing the operation of meiotic proteins. The structure of these fields is distorted in the mutants for ontogenes, thereby decreasing the efficiency of proteins and, as a consequence, causing meiotic defects.
