Broad strategies for neutralizing SARS-CoV-2 and other human coronaviruses with monoclonal antibodies

Antibody therapeutics and vaccines for coronavirus disease 2019(COVID-19)have been approved in many countries,with most being developed based on the original strain of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2).SARS-CoV-2 has an exceptional ability to mutate under the pressure of host immunity,especially the immune-dominant spike protein of the virus,which is the target of both antibody drugs and vaccines.Given the continuous evolution of the virus and the identification of critical mutation sites,the World Health Organization(WHO)has named 5 variants of concern(VOCs):4 are previously circulating VOCs,and 1 is currently circulating(Omicron).Due to multiple mutations in the spike protein,the recently emerged Omicron and descendent lineages have been shown to have the strongest ability to evade the neutralizing antibody(NAb)effects of current antibody drugs and vaccines.The development and characterization of broadly neutralizing antibodies(bNAbs)will provide broad strategies for the control of the sophisticated virus SARS-CoV-2.In this review,we describe how the virus evolves to escape NAbs and the potential neutralization mechanisms that associated with bNAbs.We also summarize progress in the development of bNAbs against SARS-CoV-2,human coronaviruses(CoVs)and other emerging pathogens and highlight their scientific and clinical significance.

The outbreak of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),which causes coronavirus disease 2019(COVID-19),has posed a tremendous threat to global health,resulting in over 510 million infections and at least 6.23 million deaths worldwide as of early May 2022.Extensive efforts to fight against the SARS-CoV-2 pandemic have accelerated the research and clinical development of various vaccines and neutralizing antibodies(NAbs)[1,2].However,SARS-CoV-2 has an exceptional ability to mutate and evade the human immune system,which highlights the surveillance of SARS-CoV-2 evolution to anticipate and facilitate a timely response to new variants that could impact viral transmissibility and the efficacy of vaccines and antibody therapies[3].

Immunotherapy employs the immune system to combat diseases including cancer,autoimmune diseases,infectious diseases and degenerative diseases,for both treatment and prevention of the conditions.The application of immunotherapies has a long history,with the milestone of identifying cow pox as the smallpox vaccine more than 200 years ago as an example(Cook,1996).After that,in 1890,Emil Behring and Kitasato Shibasaburōpublished the first paper on serum therapy which ushered an era of antibody therapy.The progress on basic research of immunological principles and the boost of bioengineering facilitates the fast evolution of immunotherapy in recent years.Immunotherapy has changed the landscape of healthcare and promoted some advanced concepts to control diseases in medicine.In this issue,we selected the major topics on immunotherapy including antibody therapy,vaccine,and immune cell therapy for summarizing current progress in this field.

Design and experimental research of a sub-Newton N_(2)O monopropellant thruster with inner-heater

Nitrous oxide(N_(2)O)is a green propellant with excellent application prospects.A subNewton N_(2)O monopropellant thruster with inner-heater and a N_(2)O self-pressurization stable supply system with regenerative heat compensation are designed in this paper.The experimental research of the thruster is described,including measurements of preheating power,activation temperature,vacuum thrust,specific impulse,life-span and pulsed operation performance.By inserting the heater into the catalyst-bed,preheating efficiency of the heater is significantly improved compared to the thruster with outer-heater.Thus,the preheating power demand of the thruster is successfully reduced to around 10 W.The mean vacuum thrust of 322 mN is attained and the corresponding specific impulse reaches 162s at the mass flow rate of 0.2 g/s.Successful activation temperature of 523 K is achieved,and the activation performance of the thruster is affected by the loading factor.A long term hot-firing test longer than 12000 s is attained.The pulsed operation performance of the inner-preheating thruster is also studied by measuring chamber pressure.Impulses with different magnitudes are produced by adjusting the opening duration of the control valve.A minimum impulse of 81 mN·s is attained.Finally,the performance of the thruster is evaluated by comparison with other thrusters of the same type.The results indicate that the proposed thruster with inner-heater is superior in terms of preheating power,activation temperature and specific impulse performance.

Damage localization effects of the regeneratively-cooled thrust chamber wall in LOX/methane rocket engines

To investigate the damage localization effects of the thrust chamber wall caused by combustions in LOX/methane rocket engines, a fluid-structural coupling computational methodology with a multi-channel model is developed to obtain 3-demensioanl thermal and structural responses.Heat and mechanical loads are calculated by a validated finite volume fluid-thermal coupling numerical method considering non-premixed combustion processes of propellants. The methodology is subsequently performed on an LOX/methane thrust chamber under cyclic operation. Results show that the heat loads of the thrust chamber wall are apparently non-uniform in the circumferential direction. There are noticeable disparities between different cooling channels in terms of temperature and strain distributions at the end of the hot run phase, which in turn leads to different temperature ranges, strain ranges, and residual strains during one cycle. With the work cycle proceeding, the circumferential localization effect of the residual strain would be significantly enhanced. A post-processing damage analysis reveals that the low-cycle fatigue damage accumulated in each cycle is almost unchanged, while the quasi static damage accumulated in a considered cycle declines until stabilized after several cycles. The maximum discrepancy of the predicted lives between different cooling channels is about 30%.

High mortality rates of 25%to,50%were reported in patients with extubation failure.liExtubation failure or re-intubation was an independent risk factor for death after extubation,not the precise reason.Common reasons included primary respiratory failure,cardiac dysfunction,ineffective cough,excess secretions,upper airway obstruction,etc.Therefore,death after extubation should be related to these reasons above in theory.

Thermal-structural analysis of regeneratively-cooled thrust chamber wall in reusable LOX/Methane rocket engines

To predict the thermal and structural responses of the thrust chamber wall under cyclic work,a 3-D fluid-structural coupling computational methodology is developed.The thermal and mechanical loads are determined by a validated 3-D finite volume fluid-thermal coupling computational method.With the specified loads,the nonlinear thermal-structural finite element analysis is applied to obtaining the 3-D thermal and structural responses.The Chaboche nonlinear kinematic hardening model calibrated by experimental data is adopted to predict the cyclic plastic behavior of the inner wall.The methodology is further applied to the thrust chamber of LOX/Methane rocket engines.The results show that both the maximum temperature at hot run phase and the maximum circumferential residual strain of the inner wall appear at the convergent part of the chamber.Structural analysis for multiple work cycles reveals that the failure of the inner wall may be controlled by the low-cycle fatigue when the Chaboche model parameter c3= 0,and the damage caused by the thermal-mechanical ratcheting of the inner wall cannot be ignored when c3〉 0.The results of sensitivity analysis indicate that mechanical loads have a strong influence on the strains in the inner wall.

Data Processing Methods of Flow Field Based on Artificial Lateral Line Pressure Sensors

The estimation of the type and parameter of flow field is important for robotic fish.Recent estimation methods cannot meet the requirements of the robotic fish due to the lack of prior knowledge or the under-fitting of the model.A processing method including data preprocessing,feature extraction,feature selection,flow type classification and flow field parameters estimation,is proposed based on the data of the pressure sensors in an artificial lateral line.Probabilistic Neural Network(PNN)is used to classify the flow field type and the Generalized Regressive Neural Network(GRNN)is the best choice for estimating the flow field parameters.Also,a few filtering methods for data preprocessing,three methods for feature selection and nine parameters estimation methods are analysis for choosing better method.The proposed method is verified by the experiments with both simulation and real data.

Sepsis remains one of the leading causes of death globally,in spite of advanced developments in intensive care and better understandings of pathophysiology related to sepsis.There is no special treatment or drug available for sepsis,currently.Under normal circumstances,neutrophil is a major player in acute infection control.However,during sepsis,the migration abilities and antimicrobial functions of neutrophils are impaired,resulting in a dysregulated immune response.Recent studies have indeed demonstrated that blocking or reversing neutrophil migration and impaired antibacterial function can improve the outcomes in septic animal models.This article systemically synthesized information regarding related factors and signaling involved in the functions of neutrophils in sepsis.This review also discussed the possibility that neutrophils be used as a marker for specific diagnosis and/or prediction of the outcomes of sepsis.
