What is already known on this topic?The global efforts to address the hepatitis C virus(HCV)are progressing,but there are still significant gaps in the diagnosis and treatment of HCV,leading to an increasing number of deaths related to HCV.What is added by this report?An extensive investigation was conducted to assess HCV RNA diagnosis,treatment uptake,and associated factors among individuals infected with HCV within Jiangsu Province.The study encompassed a large geographical area and utilized a substantial sample size.What are the implications for public health practice?Implementing focused interventions to improve the timely diagnosis of HCV RNA and increase the uptake of HCV treatment could effectively reduce the future burden of HCV-related health problems,deaths,and healthcare expenses.This is essential for achieving the global target of eliminating hepatitis C.

Chicken is an important food animal worldwide and plays an important role in human life by providing meat and eggs.Despite recent significant advances in gut microbiome studies,a comprehensive study of chicken gut bacterial,archaeal,and viral genomes remains unavailable.In this study,we constructed a chicken multi-kingdom microbiome catalog(CMKMC),including 18,201 bacterial,225 archaeal,and 33,411 viral genomes,and annotated over 6,076,006 protein-coding genes by integrating 135 chicken gut metagenomes and publicly available metagenome-assembled genomes(MAGs)from ten countries.We found that 812 and 240 MAGs in our dataset were putative novel species and genera,respectively,far beyond what was previously reported.The newly unclassified MAGs were predominant in Phyla Firmicutes_A(n=263),followed by Firmicutes(n=126),Bacteroidota(n=121),and Proteobacteria(n=87).Most of the classified species-level viral oper-ational taxonomic units belong to Caudovirales.Approximately,63.24%of chicken gut viromes are predicted to infect two or more hosts,including complete circular viruses.Moreover,we found that diverse auxiliary metabolic genes and antibiotic resistance genes were carried by viruses.Together,our CMKMC provides the largest integrated MAGs and viral genomes from the chicken gut to date,functional insights into the chicken gastrointestinal tract microbiota,and paves the way for microbial interventions for better chicken health and productivity.

A breathable, designable and flexible leather–heater used in wearable thermotherapy

Flexible heaters with personal thermal management capabilities have great potential in thermal therapy applications due to their excellent flexibility, low power consumption, and portability. However, manufacturing wearable heating devices that are breathable, wear resistant and conformal for long-term use is still challenging. To address these issues, we designed a leather heater using breathable, biocompatible, and tailorable leather as the substrate through a simple in-situ polymerization polypyrrole strategy. This heater exhibits excellent heating and mechanical properties（reaching 64°C at a voltage of 5 V with efficient Joule heat generation of 2286 W/m2and uniform temperature distribution, and functioning properly after 1000 cycles of bendability tests）. In addition, this heater displays better wear resistance and water vapor permeability rate(38.04 g/（m2h）). The cuttable and sewable of leather gives the strategy ability to be flexibly designed to mold the heater to the specific requirements of different body parts, providing a new approach to wearable thermal therapy.

Genome-wide identification of endogenous retrovirus elements and their active transcription in mink genome

Mammalian endogenous retroviruses(ERVs)are ancient retroviruses that have been integrated into genomes.ERVs were believed to be inactive until the discovery of ERV transcription in the mouse genome.However,the transcription level and function of ERV elements in mammalian genomes are not well understood.In this study,we performed the first genome‐wide scanning of ERV loci in the American mink(Neogale vison)genome(NeoERV)followed by transcriptomic analysis to detect actively transcribed NeoERV elements.A total of 365,791 NeoERV loci were identified,and161,205(44%)of these loci were found to be actively transcribed based on transcriptomic data from three types of tissues(amygdala,trachea and lung).More than one third of the actively transcribed NeoERV loci were tissue‐specific.Furthermore,some of the active loci were associated with host gene transcription,and the level of NeoERV transcription was positively correlated with that of host genes,specifically when active loci were located in overlapped gene regions.An in‐depth analysis of the envelope protein coding env gene showed that,in general,its transcription level was higher than that of NeoERVs,which is believed to be associated with host immunity.

Current influenza vaccines need to be updated annually owing to constant antigenic drift in the globular head of the viral surface hemagglutinin(HA)glycoprotein.The immunogenic subdominant stem domain of HA is highly conserved and can be recognized by antibodies capable of binding multiple HA subtypes.Therefore,the HA stem antigen is a promising target for the design of universal influenza vaccines.On the basis of an established lipid nanoparticle-encapsulated mRNA vaccine platform,we designed and developed a novel universal influenza mRNA vaccine(mHAs)encoding the HA stem antigen of the influenza A(H1N1)virus.We tested the efficacy of the mHAs vaccine using a mouse model.The vaccine induced robust humoral and specific cellular immune responses against the stem region of HA.Importantly,two doses of the mHAs vaccine fully protected mice from lethal challenges of the heterologous H1N1 and heterosubtypic H5N8 influenza viruses.Vacci-nated mice had less pathological lung damage and lower viral titers than control mice.These results suggest that an mRNA vaccine using the conserved stem region of HA may provide effective protection against seasonal and other possible influenza variants.

Human gut microbiome: the second genome of human body

The human body is actually a super-organism that is composed of 10 times more microbial cells than our body cells.Metagenomic study of the human microbiome has demonstrated that there are 3.3 million unique genes in human gut,150 times more genes than our own genome,and the bacterial diversity analysis showed that about 1000 bacterial species are living in our gut and a majority of them belongs to the divisions of Firmicutes and Bacteriodetes.In addition,most people share a core microbiota that comprises 50–100 bacterial species when the frequency of abundance at phylotype level is not considered,and a core microbiome harboring more than 6000 functional gene groups is present in the majority of human gut surveyed till now.Gut bacteria are not only critical for regulating gut metabolism,but also important for host immune system as revealed by animal studies.

The antibiotic resistance is natural in bacteria and predates the human use of antibiotics. Numerous antibiotic resistance genes （ARGs） have been discovered to confer resistance to a wide range of antibiotics. The ARGs in natural environments are highly integrated and tightly regulated in specific bacterial metabolic networks. However, the antibiotic selection pressure conferred by the use of antibiotics in both human medicine and agriculture practice leads to a significant increase of antibiotic resistance and a steady accumulation of ARGs in bacteria. In this review, we summarized, with an emphasis on an ecological point of view, the important research progress regarding the collective ARGs （antibiotic resistome） in bacterial communities of natural environments, human and animals, i.e., in the one health settings. We propose that the resistance gene flow in nature is ＂from the natural environments＂ and ＂to the natural environments＂; human and animals, as intermediate recipients and disseminators, contribute greatly to such a resistance gene ＂circulation.＂

Analyses of N‑Terminal Pro‑Brain Natriuretic Peptide,Cardiac Troponin T,and Creatine Kinase MB in Pericardial Fluid in Sudden Cardiac Death Caused by Ischemic Heart Disease

Background:Biochemical analyses of N‑terminal pro‑brain natriuretic peptide(NT‑proBNP),cardiac troponin T(cTnT),and creatine kinase MB(CK‑MB)have been reported to be valuable for the auxiliary diagnosis of sudden cardiac death(SCD)in previous forensic studies.Aims and Objectives:The present study aimed to evaluate the diagnostic efficiency of combined analyses of NT‑proBNP,cTnT and CK‑MB in the pericardial fluid for forensic diagnosis of SCD caused by ischemic heart disease.Materials and Methods:Levels of NT‑proBNP,cTnT,and CK‑MB in the pericardial fluid of 132 medicolegal autopsy cases were obtained through electrochemiluminescence method.Results:NT‑proBNP,cTnT,and CK‑MB levels were significantly elevated in SCD cases(P<0.05).Receiver‑operating characteristics(ROC)analysis showed that NT‑proBNP,cTnT,and CK‑MB have diagnostic value for the diagnosis of SCD:NT‑proBNP,cutoff value of 2236 pg/ml;cTnT,cutoff value of 199.51 ng/ml;CK‑MB:cutoff value of 2742.5 ng/ml,and the combined analyses of these three biomarkers have better diagnostic efficiency than each single biomarker alone.Moreover,the causes of SCD were sub‑divided into acute ischemic heart disease,acute myocardial infarction(AMI),and recurrent myocardial infarction subgroups for further analysis,which revealed that the ratio of cTnT/CK‑MB could be used to distinguish AMI with the cutoff value of 0.1085 estimated by ROC analysis.Conclusion:These observations suggested that the postmortem biochemical analyses of NT‑proBNP,cTnT,and CK‑MB in the pericardial fluid may assist to diagnose SCD in forensic practice,and the combined analyses of multiple biomarkers have better diagnostic efficiency than each single biomarker alone.On the basis of the postmortem biochemical analyses of NT‑proBNP,cTnT and CK‑MB,combining the ratio of cTnT/CK‑MB could be used to distinguish AMI.
