YWHAZ gene contributes to the radioresistance of oral squamous cell carcinoma cells

Objective:To investigate the contribution of YWHAZ gene on the radioresistance and metastasis ability of oral squamous cell carcinoma(OSCC)***:The relationship between the expression level of YWHAZ gene and the survival of head and neck squamous cell carcinoma(HNSC)patients was analyzed using Gene Expression Profiling Interactive Analysis(GEPIA)database.A radioresistance cell line(CAL-27R)was constructed by irradiating CAL-27 cells with frac-tional *** survival was measured by colony formation *** migration and invasion were detected by transwell *** formation of yH2AX foci was detected by immunofluorescence assay,The protein expressions were detected by Westemn blot *** some experiments,CAL-27R cells were effectively transferred with siRNA YWHAZ(siYWHAZ).Results:GEPIA database showed that the expression level of YWHAZ in HNSC tumors was higher than that in adjacent normal tissues,and the HNSC patients with higher level of YWHAZ had a shorter *** vitro experiments demonstrated that the expression of YWHAZ protein in CAL-27 cells was lower than HSC-3 cells(t=18.89,P<0.01)and radioresistant CAL-27R cells(t=25.70,P<0.01).Knockdown of YWHAZ gene significantly increased radiation-induced cell killing effect,apoptosis induction,and yH2AX foci formation inCAL-27R and HSC-3 ***,siRNA YWHAZ transfection also reduced the invasion and migration abilities of the irradiated CAL 27R[t=21.09,P<0.01(migration);t=6.16,P<0.05(invasion)]and HSC-3 cells[t=34.53,P<0.001(migration);t=4.92,P<0.05(invasion)]and attenuated radiation-induced expressions of metastasis-related proteins,Conclusion:YWHAZ contributes to the radioresistance of oral squamous cells and thus it may applicable to be a potential target for OSOC radiotherapy.

miR-125b reverses cisplatin resistance by regulating autophagy via targeting RORA/BNIP3L axis in lung adenocarcinoma

The platinum-based chemotherapy is one of the most frequently used treatment protocols for lung adenocarcinoma(LUAD),and chemoresistance,however,usually results in treatment failure and limits its application in the *** has been shown that microRNAs(miRNAs)play a significant role in tumor *** this study,miR-125b was identified as a specific cisplatin(DDP)-resistant gene in LUAD,as indicated by the bioinformatics analysis and the real-time quantitative PCR *** decreased serum level of miR-125b in LUAD patients was correlated with the poor treatment response rate and short survival ***-125b decreased the A549/DDP proliferation,and the multiple drug resistance-and autophagy-related protein expression levels,which were all reversed by the inhibition of *** addition,xenografts of human tumors in nude mice were suppressed by miR-125b,demonstrating that through autophagy regulation,miR-125b could reverse the DDP resistance in LUAD cells,both in vitro and in *** mechanistic studies indicated that miR-125b directly repressed the expression levels of RORA and its downstream BNIP3L,which in turn inhibited autophagy and reversed *** on these findings,miR-125b in combination with DDP might be an effective treatment option to overcome DDP resistance in LUAD.

A novel efficient energy absorber with free inversion of a metal foam-filled circular tube

In this paper, a novel efficient energy absorber with free inversion of a metal foam-filled circular tube(MFFCT) is designed, and the axial compressive behavior of the MFFCT under free inversion is studied analytically and numerically. The theoretical analysis reveals that the energy is mainly dissipated through the radial bending of the metal circular tube, the circumferential expansion of the metal circular tube, and the metal filled-foam compression. The principle of energy conservation is used to derive the theoretical formula for the minimum compressive force of the MFFCT over free inversion under axial loading. Furthermore, the free inversion deformation characteristics of the MFFCT are analyzed numerically. The theoretical steady values are found to be in good agreement with the results of the finite element(FE) analysis. The effects of the average diameter of the metal tube, the wall thickness of the metal tube, and the filled-foam strength on the free inversion deformation of the MFFCT are considered. It is observed that in the steady deformation stage, the load-carrying and energy-absorbing capacities of the MFFCT increase with the increase in the average diameter of the metal tube, the wall thickness of the metal tube, or the filled-foam strength. The specific energy absorption(SEA) of free inversion of the MFFCT is significantly higher than that of the metal tube alone.

We report on using synthetic silicon for a high-precision X-ray polarimeter comprising a polarizer and an analyzer,each based on a monolithic channel-cut crystal used at multiple Brewster reflections with a Bragg angle very close to 45°.Experiments were performed at the BL09B bending magnet beamline of the Shanghai Synchrotron Radiation Facility using a Si(800)crystal at an X-ray energy of 12.914 keV.A polarization purity of 8.4×10^(-9)was *** result is encouraging,as the measured polarization purity is the best-reported value for the bending magnet ***,this is the firstly systematic study on the hard X-ray polarimeter in China,which is crucial for exploring new physics,such as verifying vacuum birefringence.

Over the years,numerous geotechnical approaches have been implemented to mitigate the adverse effects of climate warming on various infrastructures in the permafrost region of the Qinghai-Tibet Plateau(QTP),such as the Qinghai-Tibet Highway and Railway,and achieved the expected engineering ***,little attention has been given to whether the performance of these geotechnical approaches has changed during the ongoing process of climate *** investigate the performance variation of one of these geotechnical approaches,which is two-phase closed thermosyphon(TPCT),during sustained climate warming,we conducted a statistical analysis of soil temperature monitoring data in 2003-2020 from eight regular embankments and six TPCT embankments in our permafrost monitoring *** results indicate that TPCT undeniably has a cooling effect on the permafrost beneath embankments,even rapidly eliminated previously formed taliks beneath ***,further analysis reveals that the performance of TPCT has been weakening during sustained climate warming,which has confirmed by the re-forming of the taliks beneath embankment where they had been previously *** on the current understanding,we attributed the weakening of thermosyphon performance to a significant reduction in the air temperature freezing index caused by ongoing climate *** this study,we aimed to draw attention to the evolving performance of geotechnical approaches in permafrost regions amid climate warming,prompting necessary engineering innovations to address this situation and ensure the sustainable development of the permafrost region on the QTP.

Metallic zinc is an excellent anode material for Zn-ion batteries,but the growth of Zn dendrite severely hinders its practical ***,an efficient and economical cationic additive,poly dimethyl diallyl ammonium(PDDA) was reported,used in aqueous Zn-ion batteries electrolyte for stabilizing Zn *** growth of zinc dendrites can be significantly restrained by benefiting from the pronounced electrostatic shielding effect from PDDA on the Zn metal ***,the PDDA is preferentially absorbed on Zn(002) plane,thus preventing unwanted side reactions on Zn *** to the introduction of a certain amount of PDDA additive into the common ZnSO_(4)-based electrolyte,the cycle life of assembled Zn‖Zn cells(1 mA·cm^(-2) and 1 mAh·cm^(-2)) is prolonged to more than 1100 *** response to the perforation issue of Zn electrodes caused by PDDA additives,the problem can be solved by combining foamy copper with zinc *** real application,Zn-ion hybrid supercapacitors and MnO_(2)‖Zn cells were assembled,which exhibited excellent cycling stability with PDDA *** work provides a new solution and perspective to cope with the dendrite growth problem of Zn anode.

Organic matter is the basis for oil and gas generation,and the depositional environment controls its *** first member of the Qingshankou Formation(K_(2)qn^(1))in Songliao Basin has a thick organic-rich shale and so is an important target section for shale oil exploration and *** the Gulong Sag,shale samples from this unit were collected over the full length of the *** characterization of the environments of deposition(EOD)of K_(2)qn^(1)was improved by utilizing lithological characteristics,thin section observations,elemental compositions,and organic carbon *** with the normalization coefficients proposed in this paper,an organic matter correlation model was established to elucidate the factors that influence organic matter *** the bottom to the top of K2qn1,the lake depth gradually becomes shallower,the primary productivity first decreases and then increases,the reducing conditions become stronger and then weaker,the water salinity gradually decreases,the climate first becomes semi-humid and then warm and humid,and the input of terrigenous debris first decreases and then increases.A major marine transgression at the base of the K_(2)qn^(1)᾽s brought in nutrients to increase primary productivity,and the density-stratified reducing environment preserved and enriched organic *** primary productivity occurred during the middle of the deposition of the K_(2)qn^(1),while terrigenous input is *** matter is preserved in reduced deep lake environments,resulting in organic matter-rich black *** lake became shallower,and the salinity decreased in the upper part of K_(2)qn^(1).Benthic organisms rapidly multiplied,consuming large amounts of oxygen and destroying the previously depositional environment,resulting in a reducing environment disturbed by benthic organisms with poor preservation conditions and the lowest organic matter content.

Magnetic and electrical transport study of the intrinsic magnetic topological insulator MnBi_(2)Te_(4)with Ge doping

As an intrinsic magnetic topological insulator with magnetic order and non-trivial topological structure,MnBi_(2)Te_(4)is an ideal material for studying exotic topological states such as quantum anomalous Hall effect and topological axion insulating ***,we carry out magnetic and electrical transport measurements on(Mn1–xGex)Bi_(2)Te_(4)(x=0,0.15,0.30,0.45,0.60,and 0.75)single *** is found that with increasing x,the dilution of magnetic moments gradually weakens the antiferromagnetic exchange ***,Ge doping reduces the critical field of ferromagnetic ordering,which may provide a possible way to implement the quantum anomalous Hall effect at lower magnetic *** transport measurements suggest that electrons are the dominant charge carriers,and the carrier density increases with the Ge doping ***,the Kondo effect is observed in the samples with x=0.45,0.60,and *** results suggest that doping germanium is a viable way to tune the magnetic and electrical transport properties of MnBi_(2)Te_(4),opening up the possibility of future applications in magnetic topological insulators.
