Complement-dependent neuroinflammation in spinal cord injury:from pathology to therapeutic implications

Spinal cord injury remains a major cause of disability in young adults,and beyond acute decompression and rehabilitation,there are no pharmacological treatments to limit the progression of injury and optimize recovery in this *** the thorough investigation of the complement system in triggering and propagating cerebral neuroinflammation,a similar role for complement in spinal neuroinflammation is a focus of ongoing *** this work,we survey the current literature investigating the role of complement in spinal cord injury including the sources of complement proteins,triggers of complement activation,and role of effector functions in the *** study relevant data demonstrating the different triggers of complement activation after spinal cord injury including direct binding to cellular debris,and or activation via antibody binding to damage-associated molecular *** effector functions of complement have been implicated in spinal cord injury,and we critically evaluate recent studies on the dual role of complement anaphylatoxins in spinal cord injury while emphasizing the lack of pathophysiological understanding of the role of opsonins in spinal cord *** this pathophysiological review,we systematically review the different translational approaches used in preclinical models of spinal cord injury and discuss the challenges for future translation into human *** review emphasizes the need for future studies to dissect the roles of different complement pathways in the pathology of spinal cord injury,to evaluate the phases of involvement of opsonins and anaphylatoxins,and to study the role of complement in white matter degeneration and regeneration using translational strategies to supplement genetic models.

Profile of Maternally Derived Antibody in Broiler Chicks and In-Ovo Vaccination of Chick Embryo against Newcastle Disease

In this study, we determined the suitability of lentogenic LaSota and naturally occurring avirulent I2 vaccine strains of Newcastle disease (ND) virus for efficacious in-ovo vaccination of broiler chickens. A total of 114 embyonated eggs divided into five groups (A, B, C, D and E) consisting of 25 eggs in each of groups A and B, 20 eggs in each of groups C and D and 24 eggs in group E were used in the study. Eighteen-day-old embryonated eggs in group A were vaccinated in-ovo with ND-I2 vaccine while the same age of embryos in group B were vaccinated with ND-LaSota. Thirteen-day-old embryonated eggs in groups C and D were vaccinated with ND-I2 and ND-LaSota respectively. Group E served as unvaccinated control. There was significant difference (p 2) used for the in-ovo vaccination were pathogenic for chick embryos, however, ND-I2 vaccine was better tolerated when administered to 18-day-old chick embryo.

Immunostimulatory Effect of Levamisole on the Immune response of Goats to <i>Peste des petits ruminants </i>(PPr) Vaccination

The use of Levamisole hydrochloride as an immunostimulant has been successful in the control of certain diseases such as Derzsy’s disease in chicken as well as in increasing lymphocytes and weight gain in pigs. However, its use as immunostimulant in the prevention of Peste des petits ruminants (PPr) in goats has not been investigated. In this study, the use of Levamisole in enhancing immune response to PPr vaccination including its effects on weight gain was investigated among groups of goats (groups A, B, C and D). Virus neutralization test was used to determine the antibody profile in vaccinated goats while the total and differential leucocyte counts and body mass index (BMI) were determined using standard procedures. Goats in group A (Levamisole primed and PPr vaccinated) seroconverted to more than 3-folds of the initial pre-vaccination geometric mean titre (GMT) of neutralizing antibody beginning from second week post-vaccination and remained high throughout the period of the experiment. Similarly, there was significant increase (p 0.05) in BMI of animals in other groups throughout the period of the experiment. The results of this study further confirm the immunostimulatory effect of Levamisole when used in combination with a vaccine.

Petroleum contaminants caused great damages to environment and human ***,the port of Oran is subject of pollution mainly by PAHs(Polycyclic Aromatic Hydrocarbons)as a result of the large flow of *** bioremediation by indigenous microorganisms is an important means for their reduction and *** the present work,a hydrocarbonoclastic bacterium strain SP49F2 was considered,firstly isolated from the contaminated marine sediments and seawater at the port of Oran(Algeria),using Bushnell-Hass mineral salt medium,and identified on the basis of morphological and biochemical characteristics and molecular tools by analysis of partial 16S rrNA gene sequence,using the Basic Local Alignment Search Tool program on the data base of NCBI(National Centre for Biotechnology Information),and the EzBioCloud 16S rrNA *** of growth of this isolate on crude oil during 20 days of culture was studied at temperature 25℃,3%(w/v)of NaCl concentration and pH 7,at 140 rpm(revolutions Per Minute).Strain SP49F2 was identified molecularly as Acinetobacter johnsonii,and might support high concentrations of crude oil(up to 10%,v/v).results of growth kinetic on crude oil as sole energy and carbon source by the isolate strain showed that the stationary phase was attained at day ***,train Acinetobacter johnsonii SP49F2 could efficiently utilize crude oil as its sole carbon and energy source,and could be used as a wonderful native biological alternative for the bioremediation of the port of Oran,and marine area polluted by petroleum hydrocarbons,as an eco-friendly efficacy degrader,and may be suitable for biotechnological applications.

Field Trial of a Thermostable Peste des petits ruminants (PPr) Vaccine in a Semi-Arid Zone of Nigeria

The field trial of a candidate thermostable Peste des petits ruminants (PPr) vaccine was carried out in flocks of sheep and goats under the extensive system of management. The immune response of vaccinated animals was determined using the neutralisation test to detect PPr virus specific antibody. Vaccinated animals seroconverted and a four-fold or more rise in antibody titre were observed between pre-vaccination and post-vaccination antibodies. The vaccine elicited significant antibody response in goats through the different routes of administration (intramuscular, intranasal, intraocular, subcutaneous and orally), but was poorly transmitted between the vaccinees and in-contact animals. The sheep responded poorly to the vaccine administered through most of the routes, except for those vaccinated through intramuscular and subcutaneous routes that seroconverted significantly (≥4 fold rise). The vaccine retained a potent titre of 3.1 log10 TCID50 for more than 8 hours after reconstitution in PBS at room temperature. Based on the response of goats to oral vaccination, it is suggested that the vaccine could be administered on the field through the oral routes and has the potential to be adapted to a feed-based administration for wider application to the scattered livestock populations under the extensive system of management.

One hundred twenty-five endophytic microorganisms were isolated from the roots,stems,and leaves of four prominent rice cultivars growing in temperate *** potential to combat rice blast disease and promote plant growth was *** dual culture tests highlighted the strong antagonistic activity of five fungal(ranging from 89%–70%)and five bacterial(72%–61%)*** examination focused on volatile compounds produced by selected isolates to counter the blast *** these,the highest chitinase(13.76μg mL−1)and siderophore(56.64%),was exhibited by Aspergillus flavus,and the highest HCN production was shown by Stenotrophomonas maltophilia(36.15μM mL−1).In terms of growth promotion traits,Aspergillus flavus and Enterobacter cloacae excelled in activities viz,phosphorous solubilization,ammonia production,auxin and gibberellic acid production,and nitrogen *** Identity of these endophytes was confirmed through molecular analysis as Trichoderma afroharzianum,Trichoderma harzianum,Penicillium rubens,Aspergillus flavus,Stenotrophomonas rhizophila,Stenotrophomonas maltophilia,Bacillus cereus,Enterobacter cloacae,and Bacillus *** greenhouse conditions,the highest disease control was shown by isolate Bacillus licheniformis and *** with an inhibition of 79%,followed by ***(77%)and ***(73%).The overall results of this study showed that Bacillus licheniformis and Stenotrophomonas rhizophila have great potential to be used as bio-stimulant and biocontrol agents to manage rice blast disease.

A Novel Approach in Hydrogeochemical Exploration for Uranium Mineralization:an Example from West Central Sinai,Egypt

Groundwater in contact with ore deposits may acquire a chemical composition that could be used as a guide for exploration. Eight well-water samples are collected from a known uraniummineralized area near Abu Zenima, west central Sinai to examine the applicability of using the hydrogeochemical technique in the search for uranium mineralization in similar arid areas. The analytical chemical data of the ground water is compared with ground radiometric measurements. The obtained results indicate that groundwater affected by uranium mineralization has a specific relativity of major anions expressed essentially as SO_4〉Cl〉HCO_3 and to a lesser extent as Cl〉SO_4〉HCO_3, associated as a rule with low magnesium content. This association constitutes a signature of uranium mineralization on the composition of groundwater in west central Sinai and could be used as an important exploration guide in the search for uranium deposits in similar areas. Anomalies in Ni, Fe, Zn and Cr and other pathfinder elements in groundwater can furnish geochemical guides to uranium ores. The immobile trace element anomalies, including Zn, Ni and Fe are strongly distributed near the orebody; whereas the relatively mobile trace elements, including Co, U, V and Cr, constitute the dispersion haloes away from the orebody. A new hydrogeochemical discrimination diagram is constructed to be used as a quick and cost effective exploration tool in the search for uranium occurrences in environmentally similar arid areas. Based on the obtained results, a new site for uranium occurrence, west of W. baba, is delineated and recommended for future detailed geological and geochemical surveying.

Survey of Cereal Consumption Habits in the Community of Djougou, Benin

The cereal group occupies a prominent place in the dietary habits of people in northern Benin and there is little recent information on cereal consumption. This study aims to assess the consumption, acquisition and supply of cereals to households in the community of Djougou. A semi-directive survey with KoBoCollect was conducted among 369 households to collect individual cereal food consumption data. The survey data processed by statistical tools showed that the most consumed cereals are maize (95%, p = 0.887), millet (58%, p = 0.755), rice (55%, p = 0.753), sorghum (15%, p = 0.635), wheat (5%, p = 0.920) and fonio barely 5%. The most common mode of acquisition in Djougou is purchase (50%, p = 0.947) but donation is also observed (25%, p = 0.988) as well as production observed in 20.6% of households. Purchases are made from retailers in local markets (45%, p = 0.920) but also in streets and alleys (30%, p = 0.765). The most widely used preservation technique is drying at room temperature (70%, p = 0.995). Households most often dry in the areas provided in the field (50%, p = 0.783) and at home (40%, p = 0.643). The preferred storage location is the kitchen (60%, p = 0.790). The bedroom (20%, 0.669) and the store (15%, 0.522) are the alternative places for storing cereals. In addition, the supply costs of cereals increased between 2020 and 2021. This vertiginous rise in prices is due, among other things, to the covid19 pandemic. The various data generated not only make it possible to have fresh data but also to invest them in the assessment of health risks for the achievement of a high level of protection of the health and life of consumers.

One of the most devastating diseases of rice worldwide is bacterial blight (BLB) caused by Xanthomonas oryzae pv. Oryzae (Xoo). In Benin, Xoo was first described in 2013 on wild rice Oryzae longistaminata. So far, no study has been done on Beninese Xoo strains. We do not know whether the pathogen has already passed into the rice varieties grown, or if they are exposed to other bacteria. Whereas the use of resistant varieties, carrying resistance genes, is the only highly effective and environmentally friendly way to control this disease, no information is available on these Xoo resistance genes in rice varieties grown in Benin apart from the one we recently. This study aims to identify Beninese Xoo strains, causing BLB and screen rice varieties grown in Benin for the main resistance genes. Diseased rice leaves showing typical symptoms of fire blight collected from different rice fields in the three phytogeographic areas of Benin were analyzed by PCr for Xoo-specific sequence identification. Furthermore, seventy-five collected rice accessions were screened to identify xa5, Xa7, xa13, and Xa21 resistance genes to Xoo. The results reveal that Xanthomonas oryzae was identified in two fields in Banikouara and one in Malanville. On the other hand, Sphingomonas sp. has been identified in several other rice fields in Benin. Forty-seven of seventy-five rice accessions examined (62.66%) carried Xoo resistance genes with 3 (4%) and 40 (53.33%) of xa5 and Xa21 respectively. None of the accessions had either Xa7 or xa13 resistance genes. Three accessions possess both xa5 and Xa21 genes. Isogenic lines IrBB60 and IrBB21, supposed to be a positive control, presented a Xoo sensitivity allele. These results indicate that Xoo has moved from the wild rice variety to the cultivated variety in northern Benin and varietal improvement programs must be implemented with varieties having several resistance genes for the efficient response against a possible BLB pandemic in Benin.

The serological prevalence of foot and mouth disease virus (FMDV) among the cattle population in the North region of Cameroon was determined using elISA (Enzyme Linked Immunosorbent Assays) serological tests for structural as well as non-structural proteins. In these cattle, FMDV rNA was identified, amplified, sequenced and the sequences were used to construct a phylogenetic tree. A sedentary cattle population randomly selected from six veterinary centres in the North region was sampled twice, six months apart. High prevalence of FMDV antibody was recorded in the first (402/466 (85.84%)) and second (358/411 (86.90%)) sampling periods. There was no significant difference (P > 0.05) in prevalence of FMDV antibody between the two sampling periods. Goudali and Peulh breeds of cattle and animals of three to five years old were the most infected with FMDV and mostly in the months of May and August. A seroprevalence of 100% (n = 14) of FMDV against serotypes A and O was observed in sera from convalescent animals in the study area. FMDV antigen detection elISA showed a prevalence of 18/37 (48.65%) for serotypes SAT1 (8.1%), SAT2 (35.1%), A (10.8%) and O (2.7%) among the clinically infected animals. There was no significant difference (P > 0.05) in prevalence of FMDV rNA between the sampling periods. A prevalence of FMDV rNA (17.5% (n = 120) and 16.7% (n = 240)) was observed among the sedentary animals that were sampled four to five months apart. FMDV rNA prevalence of 28/37 (75.6%) among clinically infected animals was also observed, thus confirming all the 12 outbreaks investigated. Sequence analysis of VP1 coding gene of the SAT2 serotype showed that it was homologous to the Libyan isolates (that caused epidemics in northern Africa in 2012) and also clustered with the serotypes isolated from both Nigeria and Sudan in 2007.
