Review:DNA-damage response network at the crossroads of cell-cycle checkpoints, cellular senescence and apoptosis

Tissue homeostasis requires a carefully-orchestrated balance between cell proliferation, cellular senescence and cell death. Cells proliferate through a cell cycle that is tightly regulated by cyclin-dependent kinase activities. Cellular senescence is a safeguard program limiting the proliferative competence of cells in living organisms. Apoptosis eliminates unwanted cells by the coordinated activity of gene products that regulate and effect cell death. The intimate link between the cell cycle, cellular senes- cence, apoptosis regulation, cancer development and tumor responses to cancer treatment has become eminently apparent. Extensive research on tumor suppressor genes, oncogenes, the cell cycle and apoptosis regulatory genes has revealed how the DNA damage-sensing and -signaling pathways, referred to as the DNA-damage response network, are tied to cell proliferation, cell-cycle arrest, cellular senescence and apoptosis. DNA-damage responses are complex, involving “sensor” proteins that sense the damage, and transmit signals to “transducer” proteins, which, in turn, convey the signals to numerous “effector” proteins implicated in specific cellular pathways, including DNA repair mechanisms, cell-cycle checkpoints, cellular senescence and apoptosis. The Bcl-2 family of proteins stands among the most crucial regulators of apoptosis and performs vital functions in deciding whether a cell will live or die after cancer chemotherapy and irradiation. In addition, several studies have now revealed that members of the Bcl-2 family also interface with the cell cycle, DNA repair/recombination and cellular senescence, effects that are generally distinct from their function in apoptosis. In this review, we report progress in understanding the molecular networks that regulate cell-cycle checkpoints, cellular senescence and apoptosis after DNA damage, and discuss the influence of some Bcl-2 family members on cell-cycle checkpoint regulation.

PRCl Marks the Difference in Plant PcG Repression

From mammals to plants, the Polycomb Group （PcG） machinery plays a crucial role in maintaining the repres- sion of genes that are not required in a specific differentiation status. However, the mechanism by which PeG machinery mediates gene repression is still largely unknown in plants. Compared to animals, few PcG proteins have been identi- fied in plants, not only because just some of these proteins are clearly conserved to their animal counterparts, but also because some PcG functions are carried out by plant-specific proteins, most of them as yet uncharacterized. For a long time, the apparent lack of Polycomb Repressive Complex （PRC）I components in plants was interpreted according to the idea that plants, as sessile organisms, do not need a long-term repression, as they must be able to respond rapidly to environmental signals; however, some PRC1 components have been recently identified, indicating that this may not be the case. Furthermore, new data regarding the recruitment of PcG complexes and maintenance of PcG repression in plants have revealed important differences to what has been reported so far. This review highlights recent progress in plant PcG function, focusing on the role of the putative PRC1 components.

Colitis-associated colon cancer:Is it in your genes?

Colitis-associated colorectal cancer(CA-CRC) is the cause of death in 10%-15% of inflammatory bowel disease(IBD) patients. CA-CRC results from the accumulation of mutations in intestinal epithelial cells and progresses through a well-characterized inflammation to dysplasia to carcinoma sequence. Quantitative estimates of overall CA-CRC risks are highly variable ranging from 2% to 40% depending on IBD severity, duration and location, with IBD duration being the most significant risk factor associated with CA-CRC development. Recently, studies have identified IBD patients with similar patterns of colonic inflammation, but that differ with respect to CA-CRC development, suggesting a role for additional non-inflammatory risk factors in CA-CRC development. One suggestion is that select IBD patients carry polymorphisms in various low penetrance disease susceptibility genes, which predispose them to CA-CRC development, although these loci have proven difficult to identify in human genomewide association studies. Mouse models of CA-CRC have provided a viable alternative for the discovery, validation and study of individual genes in CA-CRC pathology. In this review, we summarize the current CA-CRC literature with a strong focus on genetic predisposition and highlight an emerging role for mouse models in the search for CA-CRC risk alleles.

Parentage analysis in Gabonese colonies of soil-feeding termites belonging to the Cubitermes sp. affinis subarquatus complex of species （Termitidae＂ Termitinae）

Cubitermes spp. are widely distributed soil-feeding termite species in sub- Saharan Africa which play a fundamental role in soil structure and fertility. A complex of at least four cryptic species （i.e., Cubitermes sp. affunis subarquatus complex of species） has been recently described using molecular markers. In order to investigate the breeding system of these species, five microsatellite markers were used to carry out parentage and re- latedness analyses in 15 Gabonese colonies. Monogamy was confirmed as the predominant reproductive organization in Cubitermes spp. （76% of the colonies）. Within 30% of these monogamous colonies, a high relatedness between reproductives was shown, suggesting that mating between related individuals occurs. However, Cubitermes colonies can deviate from monogamy. Indeed, parental contributions by at least two related reproductives of the same sex were revealed in four colonies and polyandry was demonstrated in two of them. Infiltration of reproductives in the colony is the most plausible explanation for such cases of polygamy in Cubitermes spp.

Quality audit of colonoscopy reports amongst patients screened or surveilled for colorectal neoplasia

AIM： To complete a quality audit using recently pub- lished criteria from the Quality Assurance Task Group of the National Colorectal Cancer Roundtable. METHODS： Consecutive colonoscopy reports of patients at average/high risk screening, or with a prior col6rectal neoplasia （CRN） by endoscopists who perform 11 000 procedures yearly, using a commercial computerized endoscopic report generator. A separate institutional da- tabase providing pathological results. Required documen- tation included patient demographics, history, procedure indications, technical descriptions, colonoscopy findings, interventions, unplanned events, follow-up plans, and pathology results. Reports abstraction employed a stan- dardized glossary with 10% independent data validation. Sample size calculations determined the number of re- ports ***： Two hundreds and fifty patients （63.2± 10.5 years, female： 42.8%, average risk： 38.5%, per- sonal/family history of CRN： 43.3%/20.2%） were scoped in June 2009 by 8 gastroenterologists and 3 surgeons （mean practice： 17.1 ± 8.5 years）. Procedural indica- tion and informed consent were always documented. 14% provided a previous colonoscopy date （past polyp removal information in 25%, but insufficient in most to determine surveillance intervals appropriateness）. Most procedural indicators were recorded （exam date： 98.4%, medications： 99.2%, difficulty level： 98.8%, prep quality： 99.6%）. All reports noted extent of visualization （cecum： 94.4%, with landmarks noted in 78.8% - photodocu- mentation： 67.2%）. No procedural times were recorded. One hundred and eleven had polyps （44.4%） with ana- tomic location noted in 99.1%, size in 65.8%, morphol- ogy in 62.2%; removal was by cold biopsy in 25.2% （cold snare： 18%, snare cautery： 31.5%, unrecorded： 20.7%）, 84.7% were retrieved. Adenomas were noted in 24.8% （advanced adenomas： 7.6%, cancer： 0.4%） in this population with varying previous colonic investigations. CONCLUSION： This audit reveals lacking repo

Femtosecond Laser-Induced Breakdown Spectroscopy for Detection of Trace Elements in Sophora Leaves

Sophora leaves from several areas in Beijing are analysed by femtosecond laser-induced breakdown spectroscopy （femto-LIBS）. Although the used spectral detection system is not time-resolved, the spectral lines of trace mineral elements are detected and analysed. It is proven that the femto-LIBS can be an efficient method to detect mineral trace elements contained in tree leaves which is a biomonitor for atmospheric pollution assessment or botanic studies. An interesting case of correlation between trace elements detected in tree leaves and the pollution of the concerned area is presented, for the first time to our knowledge.

Salinity effect on the compaction behaviour,matric suction,stiffness and microstructure of a silty soil

To better understand the salinity effect on the compaction behaviour of soil,standard Proctor compaction test was conducted on soil samples with different *** suction and small-strain shear modulus,G_(max),were determined and pore size distribution was also investigated on samples statically compacted at different water *** showed that with the decrease of soil salinity from initial value of 2.1‰(g of salt/kg of dry soil)to zero,the maximum dry density increased and the optimum water content decreased,whereas there was no significant change with the increase of soil salinity from 2.1‰ to 6.76‰.Interestingly,it was observed that G_(max) also decreased when the soil salinity decreased from initial value of 2.1‰ to zero and kept almost constant when the soil salinity increased from 2.1‰ to 6.76‰,for dry samples with similar matric suction and also for samples compacted at optimum state and on wet side whose matric suctions were slightly different due to the difference in remoulded water ***,the effect of salinity on compaction behaviour and G_(max) decreased for samples compacted from dry side to wet *** pore size distribution exhibited bi-modal characteristics with two populations of micro-and macro-pores not only for samples compacted on dry side and at optimum state,but also for those compacted on wet *** examination showed that the modal size of micro-pores shifted to lower values and that of macro-pores shifted to higher values for saline soil compared to the soil without salt.

Determining osmotic suction through electrical conductivity for unsaturated low-plasticity soils

Determining osmotic suction from the electrical conductivity(EC)of soil pore water was widely reported in the ***,while dealing with unsaturated soils,they do not have enough soil pore water to be extracted for a reliable measurement of *** this paper,the chilled-mirror dew-point hygrometer and contact filter paper method were used to determine the total and matric suctions for low-plasticity soils with different salinities(0.05‰,2.1‰,and 6.76‰).A new piecewise function was proposed to calculate the osmotic suction,with the piecewise point corresponding to the first occurrence of precipitated salt in mixed salt solutions(synthetic seawater).EC,ion and salt concentrations used for osmotic suction calculation were transformed from the established relationships of mixed salt solution instead of experimental *** calculated osmotic suction by the proposed equation and the equations in the literature was compared with the indirectly measured one(the difference between the measured total and matric suctions).Results showed that the calculated osmotic suction,especially the one calculated using the proposed function,was in fair agreement with the indirectly measured data(especially for specimens with higher salinity of 6.76‰),suggesting that the transformation of EC and concentrations from the established relationship is a good alternative to direct measurement for lowplasticity *** particular,the proposed method could be applied to unsaturated low-plasticity soils which do not have enough soil pore water for a proper EC measurement.

Roles of Polycomb complexes in regulating gene expression and chromatin structure in plants

The evolutionary conserved Polycomb Group(PcG)repressive system comprises two central protein complexes,PcG repressive complex 1(PRC1)and *** complexes,through the incorporation of histone modifications on chromatin,have an essential role in the normal development of *** recent years,a significant effort has been made to characterize these complexes in the different kingdoms,and despite there being remarkable functional and mechanistic conservation,some key molecular principles have *** this review,we discuss current views on the function of plant PcG *** compare the composition of PcG complexes between animals and plants,highlight the role of recently identified plant PcG accessory proteins,and discuss newly revealed roles of known PcG *** also examine the mechanisms by which the repression is achieved and how these complexes are recruited to target ***,we consider the possible role of some plant PcG proteins in mediating local and long-range chromatin interactions and,thus,shaping chromatin 3D architecture.
