Metabolically healthy obesity refers to obese individuals who do not develop metabolic *** people store fat in subcutaneous adipose tissue(SAT)rather than in visceral adipose tissue(VAT).However,the molecules participating in this specific scenario remain ***18,a lipid droplet(LD)-associated protein,mediates the contact between the endoplasmic reticulum(ER)and LDs to facilitate LD growth and *** the present study,we show that the protein level of Rab18 is specifically upregulated in the SAT of obese people and ***18 adipocyte-specific knockout(Rab18 AKO)mice had a decreased volume ratio of SAT to VAT compared with wildtype *** subjected to high-fat diet(HFD),Rab18 AKO mice had increased ER stress and inflammation,reduced adiponectin,and decreased triacylglycerol(TAG)accumulation in *** contrast,TAG accumulation in VAT,brown adipose tissue(BAT)or liver of Rab18AKO mice had a moderate increase without ER stress ***18 AKO mice developed insulin resistance and systematic ***18 AKO mice maintained body temperature in response to acute and chronic cold induction with a thermogenic SAT,similar to the counterpart ***,Rab18-deficient 3T3-L1 adipocytes were more prone to palmitate-induced ER stress,indicating the involvement of Rab18 in alleviating lipid ***18 AKO mice provide a good animal model to investigate metabolic disorders such as impaired *** conclusion,our studies reveal that Rab18 is a key and specific regulator that maintains the proper functions of SAT by alleviating lipid-induced ER stress.

Toppling failure of rock mass/soil slope is an important geological and environmental *** its failure mechanism under different conditions has great significance in *** toppling failure of a cutting slope occurred in a hydropower station in Kyushu,Japan illustrates that the joint characteristic played a significant role in the occurrence of rock slope tipping ***,in order to consider the mechanical properties of jointed rock mass and the influence of geometric conditions,a simplified analytical approach based on the limit equilibrium method for modeling the flexural toppling of cut rock slopes is proposed to consider the influence of the mechanical properties and geometry condition of jointed rock *** theoretical solution is compared with the numerical solution taking Kyushu Hydropower Station in Japan as one case,and it is found that the theoretical solution obtained by the simplified analysis method is consistent with the numerical analytical solution,thus verifying the accuracy of the simplified ***,the Goodman-Bray approach conventionally used in engineering practice is improved according to the analytical *** results show that the allowable slope angle may be obtained by the improved Goodman-Bray approach considering the joint spacing,the joint frictional angle and the tensile strength of rock mass together.

以前的药理学研究表明,钩藤水溶性提取物C-MED-100TM不仅具有抗氧化活性,而且还具有很好的DNA修复和免疫功能.Batch-2.一种新的水溶性钩藤提取物,而它的自由基清除能力及神经保护作用还未见报道.首先检测了batch-2.六羟多巴胺诱导的SH-SY5Y细胞损伤的神经保护作用机制,然后利用红外光谱、HPLC和分光光度技术对batch-2.组成成分进行了分析.结果表明,batch-2.有清除各种自由基的能力,尤其是对羟自由基的清除(2.mg/L的batch-2.羟自由基的清除率为60%),batch-2.剂量依赖性地抑制6-羟多巴胺诱导的细胞凋亡、脂质过氧化水平、线粒体膜电位的降低和细胞内活性氧和一氧化氮的增加.同时,batch-2.制了由6-羟多巴胺诱导的SH-SY5Y细胞内iNOS和NF-κB蛋白的上调.结果表明,batch-2.六羟多巴胺诱导的SH-SY5Y细胞损伤的神经保护作用是通过清除活性氧和一氧化氮、抑制iNOS和NF-κB表达实现的.成分分析表明,batch-2.的多酚和奎宁酸含量分别为6.43%和0.0958%.上述结果显示,batch-2.抗氧化机制部分类似于EGCG.对于帕金森病的预防,batch-2.一个潜在具有很好的神经保护作用的天然抗氧化剂.

BACKGROUND Acute-on-chronic liver disease(AoCLD)accounts for the majority of patients hospitalized in the Department of Hepatology or Infectious *** To explore the characterization of AoCLD to provide theoretical guidance for the accurate diagnosis and prognosis of *** Patients with AoCLD from the Chinese Acute-on-Chronic Liver Failure(ACLF)study cohort were included in this *** clinical characteristics and outcomes,and the 90-d survival rate associated with each clinical type of AoCLD were analyzed,using the Kaplan-Meier method and the log-rank *** A total of 3375 patients with AoCLD were enrolled,including 1679(49.7%)patients with liver cirrhosis acute decompensation(LC-AD),850(2..2.)patients with ACLF,577(17.1%)patients with chronic hepatitis acute exacer-bation(CHAE),and 2.9(8.0%)patients with liver cirrhosis active phase(LC-A).The most common cause of chronic liver disease(CLD)was HBV infection(71.4%).The most common precipitants of AoCLD was bacterial infection(2..8%).The 90-d mortality rates of each clinical subtype of AoCLD were 43.4%(2.2.535)for type-C ACLF,36.0%(36/100)for type-B ACLF,2..0%(58/2.5)for type-A ACLF,9.0%(151/1679)for LC-AD,3.0%(8/2.9)for LC-A,and 1.2.(7/577)for *** HBV infection is the main cause of CLD,and bacterial infection is the main precipitant of *** most common clinical type of AoCLD is *** diagnosis and timely intervention are needed to reduce the mortality of patients with LC-AD or ACLF.

Traditional Chinese medicines (TCMs) have a long history of playing a vital role in disease prevention, symptom alleviation, and health improvement. However, their complex ingredients and as-yetunknown mechanisms restrict their application. With increasing evidence indicating that the gut microbiota is important in host health and may be associated with the therapeutic activity of TCM components, it may now be possible to assess the effects of TCMs from the perspective of the gut microbiota. The gut microbiota functions within four major physiological pathways as follows: It participates in host metabolism, forms global immunity, maintains homeostasis of the gastrointestinal tract, and affects brain function and host behavior. This article reviews the reported correlations between TCMs and certain diseases, such as chronic liver disease, ulcerative colitis, obesity, and type 2.diabetes, and elucidates the underlying mechanisms, with a focus on changes in the gut microbiota. In future, further studies are required with more advanced experimental design in order to reveal the interactions between TCMs and the gut microbiota, and provide new insight into and guidance for TCM-based drug discovery.

本研究以分离自青藏高原植物内生真菌(3株青霉和1株木霉)浸种燕麦,以未接种培养基为无菌株对照(CK),探究轻度、中度和重度干旱胁迫下其对燕麦生长、生理及根系内生真菌群落结构的影响。浸种后的种子置于培养皿中发芽,在正常条件下生长30 d后,在不同渗透胁迫处理下生长10 d,添加50 mL的聚乙二醇溶液模拟不同梯度干旱胁迫:5%、10%、15%(轻度胁迫);2.%、2.%(中度胁迫);30%、35%(重度胁迫),以无菌水作为未胁迫对照(0%)。结果表明,随着胁迫的加剧,燕麦幼苗的株高、鲜重、干重和叶绿素含量普遍下降,而过氧化物酶活性和丙二醛、脯氨酸含量随着内生真菌处理的不同而变化很大。真菌浸种燕麦,盆栽40 d根系Illumina Miseq结果共获得192.44条有效序列,包括1140个OTU,隶属于9门-15纲-34目-56科-148属;与对照相比,浸种改变了燕麦根系内生真菌群落结构;真菌浸种提高了担子菌门红菇科的相对丰度,优势属为乳菇属,而对照的优势属为链格孢属。主成分分析将浸种所用的青霉属和木霉属菌株分为3类,分别为促生菌株、增强抗逆性菌株和增加微生物多样性的菌株;白木霉增加了根系内生真菌多样性,而青霉属、鹅掌青霉促进燕麦生长及抗逆性。本研究揭示了青藏高原内生真菌浸种通过促进燕麦生长,降低干旱造成的氧化损伤及改变根系内生真菌群落结构,增强了燕麦对干旱胁迫的响应能力。
