Fungal cell-wall-degrading enzymes have great utility in the agricultural and food *** cell-wall-degrading enzymes are known to have functions that can help defend against pathogenic *** existing methods used to discover these enzymes are not well adapted to fungi culture and morphology,which prevents the proper evaluation of these *** report the first droplet-based microfluidic method capable of long-term incubation and low-voltage conditions to sort filamentous fungi inside nanoliter-sized *** new method was characterized and validated in solid-phase media based on colloidal chitin such that the incubation of single spores in droplets was possible over multiple days(2–4 days)and could be sorted without droplet *** long-term culture,we examined the activity of cell-wall-degrading enzymes produced by fungi during solid-state droplet fermentation using three highly sensitive fluorescein-based *** also used the low-voltage droplet sorter to select clones with highly active cell-wall-degrading enzymes,such as chitinases,β-glucanases,and β-Nacetylgalactosaminidases,from a filamentous fungi droplet library that had been incubated for>4 *** new system is portable,affordable for any laboratory,and user-friendly compared to classical droplet-based microfluidic *** propose that this system will be useful for the growing number of scientists interested in fungal microbiology who are seeking high-throughput methods to incubate and sort a large library of fungal cells.

Uncovering the uncertainty: Risk factors and clinical relevance of P1 lesions on small bowel capsule endoscopy of anemic patients

AIM To identify risk factors for P1 lesions on small bowel capsule endoscopy(SbcE) and to describe the natural history of anemic patients with such type of *** One hundred patients were consecutively selected for a case-control analysis performed between 37 cases with P1 lesions and 63 controls with negative SbcE. Age, gender, comorbidities and regular medication were collected. Rebleeding, further investigational studies and death were also analyzed during the *** No significant differences on gender, median age or charlson index were found between groups. Although no differences were found on the use of proton pump inhibitors, acetylsalicylic acid, anticoagulants or antiplatelet agents, the use of non-steroidal antiinflammatory drugs(NSAID) was associated with a higher risk of P1 lesions(OR = 12.00, 95%cI: 1.38-104.1). From the 87 patients followed at our center, 39 were submitted to additional studies for investigation of iron-deficiency anemia(IDA), and this was significantly more common in those patients with no findings on SbcE(53.7% vs 30.3%, P = 0.033). A total of 29 patients had at least one rebleeding or IDA recurrence episode and 9 patients died of non-anemia related causes but no differences were found between cases and *** P1 lesions are commonly found in patients with IDA submitted to SbcE. The use of NSAID seems to be a risk factor for P1 lesions. The outcomes of patients with P1 lesions do not differ significantly from those with P0 lesions or normal SbcE.

Members of the botryosphaeriaceae are well known fungi associated with dieback,canker and fruit rot on various hosts worldwide,including *** aim of this study was identify a large collection of botryosphaeriaceae species associated with dieback and stem-end rot of mango in the semi-arid region of Northeastern brazil,and compare the species in relation to mycelial growth,pathogenicity and virulence.A total of 115 isolates were sampled and based on morphology and DNA sequence data(ITS and EF1-α)seven taxa were identified,namely,botryosphaeria dothidea,***,Fusicoccum fabicercianum,Neofusicoccum parvum,*** ***,Neoscytalidium dimidiatum and Pseudofusicoccum *** and *** were the most commonly isolated species,which represented 37%and 33%of all isolates *** is reported for the first time in association with mango diseases *** were significant differences among the species obtained in this study in relation to optimum temperature for mycelial growth and mycelial growth *** species were pathogenic on mango *** were significant differences in virulence among the species,with *** and *** being the most virulent species,while *** was the least virulent.

colletotrichum species are the most important and widespread form of decay affecting mango fruit *** this study,colletotrichum species associated with fruit anthracnose isolated from mango in northeastern brazil were subject to molecular and morphological *** partial sequences of the glyceraldehyde-3-phosphate dehydrogenase gene of 143 colletotrichum isolates was amplified,as an initial measure of genetic diversity.A subset of 47 isolates,selected to represent the range of genetic diversity and geographic origin,were further sequenced using the partial actin,β-tubulin,calmodulin,glutamine synthetase genes and rDNA-ITS *** multilocus sequence analysis,together with a critical examination of the phenotypic characters,revealed four previously described species(colletotrichum asianum,colletotrichum fructicola,colletotrichum tropicale and colletotrichum karstii)and one new *** new species is introduced as colletotrichum dianesei and formally described,illustrated and compared with similar *** *** and *** have previously been reported from mango,while the other species represent the first report associated with the mango fruits *** species are reported for the first time associated with the mango fruits in brazil.

Mango(Mangifera indica)is a major tropical fruit species cultivated in *** objective of this study was to identify species of Lasiodiplodia associated with dieback and stem-end rot of mango in the semi-arid region of Northeastern brazil,and compare the species in relation to mycelial growth,pathogenicity and virulence.A total of 120 isolates of Lasiodiplodia were used and identifications were made using a combination of morphology and phylogenetic analysis based on partial translation elongation factor 1-αsequence(EF1-α)and internal transcribed spacers(ITS).The following species were identified:Lasiodiplodia crassispora,***,***,***,***,*** and Lasiodiplodia sp..Lasiodiplodia theobromae was the most frequently isolated species,which represented 41%of all the *** this species had been previously reported on mango in brazil,while the other species represent the first report associated with mango tree diseases in this *** crassispora is reported for the first time associated with mango diseases *** were significant differences in mycelial growth rates among the Lasiodiplodia species and also in the optimum temperature for *** species of Lasiodiplodia were pathogenic on mango *** were significant differences in virulence among the species,wherein *** and Lasiodiplodia *** the most virulent,while the least virulent were ***,***,*** and ***.

Endothelin and neonatal capsaicin regulate gastric resistance to injury in bDL rats

AIM: To investigate the relationship between primary afferent neurons, endothelin (ET) and the role of its receptors on ethanol-induced gastric damage in cirrhotic rats. METHODS: cirrhosis and portal hypertension were induced in rats by bile duct ligation (bDL) while controls had a sham operation. The association between ET and afferent neurons on the gastric mucosa was evaluated by capsaicin treatment in newborn rats, the use of ET agonists or antagonists, gastric ET-1 and -3 mRNA and synthetic capacity. Ethanol-induced damage was assessed using ex vivo gastric chamber *** blood flow was measured by laser-Doppler flow-metry. RESULTS: ET-3 and an ETb receptor antagonist sig- nificantly reduced the extent of ethanol-induced gastric damage in bDL rats. Gastric ET-1 and -3 levels were 30% higher in bDL rats compared to control rats. cap-saicin treatment restored the gastric resistance and blood flow responses to topical application of ethanol in bDL rats and ET-1 and -3 production to levels observed in controls. cONcLUSION: Our results suggest that the reduced resistance of the gastric mucosa of cirrhotic rats to ethanol-induced injury is a phenomenon modulated by ET through the ET b receptor and by sensory afferent neurons.

Oral purgative and simethicone before small bowel capsule endoscopy

AIM:To evaluate small bowel cleansing quality,diagnostic yield and transit time,comparing three cleansing protocols prior to capsule ***:Sixty patients were prospectively enrolled and randomized to one of the following cleansing protocols:patients in Group A underwent a 24 h liquid diet and overnight fasting;patients in Group b followed protocol A and subsequently were administered 2 L of polyethylene glycol(PEG) the evening before the procedure;patients in Group c followed protocol b and were additionally administered 100 mg of simethicone 30 min prior to capsule *** bowel cleansing was independently assessed by two experienced endoscopists and classified as poor,fair,good or excellent according to the proportion of small bowel mucosa under perfect conditions for *** there was no agreement between the two endoscopists,the images were reviewed and discussed until a consensus was *** preparation was considered acceptable if > 50% or adequate if > 75% of the mucosa was in perfect cleansing *** amount of bubbles was assessed independently and it was considered significant if it prevented a correct interpretation of the *** endoscopic findings,gastric emptying time(GET) and small bowel transit time(SbTT) were recorded for each ***:There was a trend favoring Group b in achieving an acceptable(including fair,good or excellent) level of cleansing(Group A:65%;Group b:83.3%;Group c:68.4%) [P = not significant(NS)] and favoring Group c in attaining an excellent level of cleansing(Group A:10%;Group b:16.7%;Group c:21.1%)(P = NS).The number of patients with an adequate cleansing of the small bowel,corresponding to an excellent or good classification,was 5(25%) in Group A,5(27.8%) in Group b and 4(21.1%) in Group c(P = 0.892).conversely,7 patients(35%) in Group A,3 patients(16.7%) in Group b and 6 patients(31.6%) in Group c were considered to have poor small bowel cleansing(P = 0.417),with sig

Aim:To investigate the correlation of p53 and prohibitin (PHb) expression in the spectrum of hepatitis, cirrhosis, and human hepatocellular carcinoma (Hcc).Methods:Hepatic biopsies from patients with Hcc (n = 60), cirrhosis (cIR,n= 30), hepatitis c virus (HcV,n = 30), and normal livers (NL,n = 20) were examined for immunohistochemical expression of RelA/p65, tumor necrosis factor receptor-1 (TNFR1), TNF-related apoptosis-inducing ligand (TRAIL), p53, and PHb. The samples were also analysed by nuclear factor kappa b (NFκb) Southwestern histochemistry and a transferase-mediated dUTP-biotin nick-end labelling ***: Expression of NFκb and RelA/p65 was detected increasingly from NL to cIR, but had a diminished labelling in the Hcc cases (P creased in Hcc, but was progressively reduced from cIR to NL (P b nuclear expressions were ampliifed in cases of Hcc, but diminished in NL, HcV, and cIR (Pconclusion: These results suggest that in addition to well-understood sequences of proinlfammatory events such as TNF-induced NFκb activation and NFκb/TRAIL pathway-mediated apoptosis, development of Hcc is also inlfuenced by regulation ofp53 andPHb tumour suppressor function. Additional studies are necessary to explain the contradictory mechanisms of the tumour microenvironment observed in the sequence of HcV, cIR, and Hcc.
