Epithelial-mesenchymal plasticity(EMP)of cancer cells contributes to cancer cell heterogeneity,and it is well established that EMP is a critical determinant of acquired resistance to cancer treatment modalities including radiation therapy,chemotherapy,and targeted ***,we aimed to explore how EMP contributes to cancer cell camouflage,allowing an ever-changing population of cancer cells to pass under the radar of our immune system and consequently compromise the effect of immune checkpoint blockade *** ultimate clinical benefit of any combination regimen is evidenced by the sum of the drug-induced alterations observed in the variety of cellular populations composing the tumor immune *** finely-tuned molecular crosstalk between cancer and immune cells remains to be fully elucidated,particularly for the spectrum of malignant cells along the epithelial to mesenchymal ***-dimensional single cell analyses of specimens collected in ongoing clinical studies is becoming a key contributor to our understanding of these *** review will explore to what extent targeting EMP in combination with immune checkpoint inhibition represents a promising therapeutic avenue within the overarching strategy to reactivate a halting cancer-immunity cycle and establish a robust host immune response against cancer *** strategies currently in clinical development will be discussed.

Design Safe addresses the challenges of supporting integrative data-driven research in natural hazards *** is an end-to-end data management,communications,and analysis platform where users collect,generate,analyze,curate,and publish large data sets from a variety of sources,including experiments,simulations,field research,and post-disaster *** achieves key objectives through:(1)integration with high performance and cloud-computing resources to support the computational needs of the regional risk assessment community;(2)the possibility to curate and publish diverse data structures emphasizing relationships and understandability;and(3)facilitation of real time communications during natural hazards events and disasters for data and information *** resultant services and tools shorten data cycles for resiliency evaluation,risk modeling validation,and forensic *** article illustrates salient features of the *** summarizes its design principles,architecture,and *** focus is on case studies to show the impact of Design Safe on the disaster risk *** Next Generation Liquefaction project collects and standardizes case histories of earthquake-induced soil liquefaction into a relational database—Design Safe—to permit users to interact with the *** can correlate in Design Safe building dynamic characteristics based on data from building sensors,with observed damage based on ground motion *** groups upload,curate,and publish wind,seismic,and coastal damage data they gather during field reconnaissance missions,so these datasets are available shortly after a *** a part of the education and community outreach efforts of Design Safe,training materials and collaboration space are also offered to the disaster risk management community.

Understanding the immune responses elicited by SARS-CoV-2 infection is critical in terms of protection against reinfection and,thus,for public health policy and vaccine development for *** this study,using either live SARS-CoV-2 particles or retroviruses pseudotyped with the SARS-CoV-2 S viral surface protein(Spike),we studied the neutralizing antibody(nAb)response in serum samples from a cohort of 140 SARS-CoV-2 qPCR-confirmed infections,including patients with mild symptoms and also more severe forms,including those that required intensive *** show that nAb titers correlated strongly with disease severity and with anti-spike IgG ***,patients from intensive care units exhibited high nAb titers;conversely,patients with milder disease symptoms had heterogeneous nAb titers,and asymptomatic or exclusive outpatient-care patients had no or low *** found that nAb activity in SARS-CoV-2-infected patients displayed a relatively rapid decline after recovery compared to individuals infected with other ***,we found an absence of cross-neutralization between endemic coronaviruses and SARS-CoV-2,indicating that previous infection by human coronaviruses may not generate protective nAbs against ***,we found that the D614G mutation in the spike protein,which has recently been identified as the current major variant in Europe,does not allow neutralization ***,our results contribute to our understanding of the immune correlates of SARS-CoV-2-induced disease,and rapid evaluation of the role of the humoral response in the pathogenesis of SARS-CoV-2 is warranted.
