Autologous tissue patch rich in stem cells created in the subcutaneous tissue

AIM:To investigate whether we could create natural autologous tissue patches in the subcutaneous space for organ repair. METHODS: We implanted the following three types of inert foreign bodies in the subcutaneous tissue of rats to produce autologous tissue patches of different geometries:(1) a large-sized polyvinyl tube(l = 25 mm,internal diameter = 7 mm) sealed at both ends by heat application for obtaining a large flat piece of tissue patch for organ repair;(2) a fine polyvinyl tubing(l = 25 mm,internal diameter = 3 mm) for creating cylindrically shaped grafts for vascular or nerve repair; and(3) a slurry of polydextran particle gel for inducing a bladder-like tissue. Implantation of inert materials was carried out by making a small incision on one or either side of the thoracic-lumbar region of rats. Subcutaneous pockets were created by blunt dissection around the incision into which the inert bodies were inserted(1 or 2 per rat). The incisions were closed with silk sutures,and the animals were allowed to recover. In case of the polydextran gel slurry 5 m l of the slurry was injected in the subcutaneous space using an 18 gauge needle. After implanting the foreign bodies a newly regenerated encapsulating tissue developed around the foreign bodies. The tissues were harvested after 4-42 d of implantation and studied by gross examination,histology,and histochemistry for organization,vascularity,and presence of mesenchymal stem cells(MSCs)(CD271+CD34+ cells). RESUlTS: Implanting a large cylindrically shaped polyvinyl tube resulted in a large flat sheet of tissue that could be tailored to a specific size and shape for use as a tissue patch for repairing large organs. Implanting a smaller sized polyvinyl tube yielded a cylindrical tissue that could be useful for repairing nerves and blood vessels. 5-Fluorouracil(5-FU)is an anticancer drug extensively used for different *** metabolic activation leads to several nucleoside and nucleotide metabolites essential to exert its cytotoxic activity on multiple cellular targets such as enzymes,DNA and *** this paper,we describe the development of a method based on liquid chromatography coupled with high resolution mass spectrometry suitable for the simultaneous determination of the ten anabolic metabolites(nucleoside,nucleotide and sugar nucleotide)of *** chromatographic separationwas optimized on a porous graphitic carbon column allowing the analysis of the metabolites of 5-FU as well as endogenous *** detection was performed on an Orbitrap■tandem mass *** of the method was verified in intracellular content and in RNA *** limit of detection was equal to 12 pg injected on column for nucleoside metabolites of 5-FU and 150 pg injected on column for mono-and tri-phosphate nucleotide *** effect was evaluated in cellular contents,DNA and RNA extracts for nucleoside and nucleotides *** method was successfully applied to i)measure the proportion of each anabolic metabolite of 5-FU in cellular contents,ii)follow the consequence of inhibition of enzymes on the endogenous nucleotide pools,iii)study the incorporation of metabolites of 5-FU into RNA and DNA,and iv)to determine the incorporation rate of 5-FUrd into 18 S and 28 S sub-units of rRNA.

Usually, the maize cob is formed by grains of medium size. However, the extremes have larger or smaller size grains. The objective of this study was to investigate the influence of grain size from the same hybrid on the physicochemical properties of isolated starch, crude maize flours and nixtamalized maize flours. Two hybrids, one from CIMMyT-Mexico called IMIC-254 and one commercial sample from Monsanto (Puma) were studied. The isolated starch granules from small, medium, and large grains exhibit the same size and distribution. The grain size has influence in the determination of cooking and steeping times;small grains reach these parameters faster than medium and large ones. The hardness of the grain size for both hybrids does not showed statistical differences between them. The starch from small, medium and large grains is mainly composed of amylopectin;this result is confirmed by X-ray diffraction and Megazine analysis. The apparent viscosity of the isolated starches of small grains showed statistically significant higher peak values. According to these results, it is possible to use small, medium, and large grains to obtain products with the same physicochemical properties, by adjusting the cooking and steeping times and Ca2+ content.

A 3D forest monitoring system,called FORSAT(a satellite very high resolution image processing platform for forest assessment),was developed for the extraction of 3D geometric forest information from very high resolution(VHR)satellite imagery and the automatic 3D change *** is composed of two complementary tasks:(1)the geometric and radiometric processing of satellite optical imagery and digital surface model(DSM)reconstruction by using a precise and robust image matching approach specially designed for VHR satellite imagery,(2)3D surface comparison for change *** allows the users to import DSMs,align them using an advanced 3D surface matching approach and calculate the 3D differences and volume changes(together with precision values)between *** is a single source and flexible forest information solution,allowing expert and non-expert remote sensing users to monitor forests in three and four(time)*** geometric resolution and thematic content of VHR optical imagery are sufficient for many forest information needs such as deforestation,clear-cut and fire severity *** capacity and benefits of FORSAT,as a forest information system contributing to the sustainable forest management,have been tested and validated in case studies located in Austria,Switzerland and Spain.
