We previously reported a spotted-leaf mutant pelota（originally termed HM_（47）） in rice displaying arrested growth and enhanced resistance to multiple races of Xanthomonas oryzae pv. oryzae. Here, we report the mapbased cloning of the causal gene OsPELOTA（originally termed spl^（HM47））. We identified a single base substitution from T to A at position 556 in the coding sequence of OsPELOTA, effectively mutating phenylalanine to isoleucine at position 186 in the translated protein sequence. Both functional complementation and over-expression could rescue the spotted-leaf phenotype. OsPELOTA, a paralogue to eukaryotic release factor 1（eRF_1）, shows high sequence similarity to Drosophila Pelota and also localizes to the endoplasmic reticulum and plasma *** is constitutively expressed in roots, leaves,sheaths, stems, and panicles. Elevated levels of salicylic acid and decreased level of jasmonate were detected in the pelota mutant. RNA-seq analysis confirmed that genes responding to salicylic acid were upregulated in the mutant. Our results indicate that the rice PELOTA protein is involved in bacterial leaf blight resistance by activating the salicylic acid metabolic pathway.

here has been a strong need for simulation environments that are capable of modeling deep interdependencies between complex systems encountered during natural hazards,such as the interactions and coupled effects between civil infrastructure systems response,human behavior,and social policies,for improved community *** such complex components with an integrated simulation requires continuous data exchange between different simulators simulating separate models during the entire simulation *** can be implemented by means of distributed simulation platforms or data passing *** order to provide a systematic reference for simulation tool choice and facilitating the development of compatible distributed simulators for deep interdependent study in the context of natural hazards,this article focuses on generic tools suitable for integration of simulators from different fields but not the platforms that are mainly used in some specific *** this aim,the article provides a comprehensive review of the most commonly used generic distributed simulation platforms(Distributed Interactive Simulation(DIS),High Level Architecture(HLA),Test and Training Enabling Architecture(TENA),and Distributed Data Services(DDS))and data passing tools(Robot Operation System(ROS)and Lightweight Communication and Marshalling(LCM))and compares their advantages and *** specific limitations in existing platforms are identified from the perspective of natural hazard *** mitigating the identified limitations,two platform design recommendations are provided,namely message exchange wrappers and hybrid communication,to help improve data passing capabilities in existing solutions and provide some guidance for the design of a new domain-specific distributed simulation framework.

Tandem repetitions in transcriptomes of some Solanaceae species

Characterization of occurrence, density and motif sequence of tandem repeats in the transcribed regions is helpful in understanding the functional significance of these repeats in the modern genomes. We analyzed tandem repeats present in expressed sequences of thirteen species belonging to genera Capsicum, Nicotiana, Petunia and Solanum of family Solanaceae and the genus Coffea of Rubiaceae to investigate the propagation and evolutionary sustenance of these repeats. Tandem repeat containing sequences constituted 1.58% to 7.46% of sequences analyzed. Tandem repetitions of size 2, 15, 18 and 21 bp motifs were more frequent. Repeats with unit sizes 21 and 22 bp were also abundant in genomic sequences of potato and tomato. While mutations occurring in these repeats may alter the repeat number, genomes adjust to these changes by keeping the translated products unaffected. Surprisingly, in majority of the species under study, tandem repeat motif length did not exceed 228 bp. Conserved tandem repeat motifs of sizes 180, 192 and 204 bp were also abundant in the genomic sequences. Our observations lead us to propose that these tandem repeats are actually remnants of ancestral megasatellite repeats, which have split into multiple repeats due to frequent insertions over the course of evolution.

Anxiety is an unpleasant state,which can critically decrease the quality of life is often accompanied by nervous behaviour and *** is a lignan isolated from various Phyllanthus *** literature survey on niranthin highlights wide ranges of the therapeutic *** a present study,based on our previous investigations,we evaluated pure,isolated and characterized niranthin as an anxiolytic *** niranthin[6-[(2R,3R)-3-[(3,4-dimethoxyphenyl)methyl]-4-methoxy-2-(methoxymethyl)butyl]-4-methoxy-1,3-benzodioxole]was purchased from commercial source and further subjected for assessment of its anxiolytic potentials using popular animal models including Elevated plus-maze model/test(EPM)and Light&Dark Exploration test(L&D).GABA-A receptor mediation was evaluated by pretreating the mice with the GABA-A receptor antagonist Flumazenil before the EPM *** docking simulation studies(pdb id:4COF)carried out by Vlife QSAR software showed that niranthin(docking score:−62.1714 kcal/mol)have shown comparatively best docking score compared to the standard drug Diazepam(docking score:−63.1568 kcal/mol).To conclude,Niranthin has probable potential in the management of anxiety *** in-silico and in-vivo analysis(indirectly)indicated the plausible role of GABA mediation for anxiolytic ***,these studies are preliminary,future in depth experimental explorations will be required to use Niranthin as anti-anxiety drug in near future.

Aim:The present in silico study aimed to evaluate the ATP-binding cassette(ABC)transporter inhibition potential of Bulbine frutescens(***)***:Several previous studies and databases were used to retrieve the ligands and target protein *** molecular docking study was performed using the Auto Dock Tools,and the GROMACS package was applied to accomplish molecular dynamics ***:Utilizing the molecular docking and simulation approach,~25 phytochemicals were screened against the ABC transporter *** score analysis revealed that *** phytochemical 4’-Demethylknipholone 2’-β-D-glucopyranoside exhibited strong binding on the ABC transporter protein with a minimum binding score-9.8 kcal/mol in comparison to the standard ABC transporter inhibitor diltiazem(-6.86 kcal/mol).Furthermore,molecular dynamics simulation for 4’-Demethylknipholone 2’-β-D-glucopyranoside showed an acceptable root mean square deviation,radius of gyration,root mean square fluctuation,and hydrogen bond,in addition to other lead ***:The in-silico study demonstrated that *** phytochemical 4’-Demethylknipholone 2’-β-D-glucopyranoside possesses anti-drug resistance properties and requires further testing in preclinical settings.

Androgen deprivation therapy targeting the androgens/androgen receptor(AR)signaling continues to be the mainstay treatment of advanced-stage prostate *** use of second-generation antiandrogens,such as abiraterone acetate and enzalutamide,has improved the survival of prostate cancer patients;however,a majority of these patients progress to castration-resistant prostate cancer(CRPC).The mechanisms of resistance to antiandrogen treatments are complex,including specific mutations,alternative splicing,and amplification of oncogenic proteins resulting in dysregulation of various signaling *** this review,we focus on the major mechanisms of acquired resistance to second generation antiandrogens,including AR-dependent and AR-independent resistance mechanisms as well as other resistance mechanisms leading to CRPC *** knowledge of resistance mechanisms to AR targeted treatments will lead to additional research on designing more effective therapies for advanced-stage prostate cancer.
