为缩短竹材生物脱胶时间,得到能够满足后道加工的工艺纤维(竹束),并提高成品质量,探索了温水沤竹过程中细菌菌群结构的演替规律及作用机制。采用高通量测序与传统可培养技术相结合的方法,确定了沤竹过程中的优势菌;并结合水沤过程中竹块的形态变化以及竹块与竹束的化学成分差异,分析了菌群在水沤脱胶过程中的作用。结果表明:竹材的水沤脱胶过程以木质素的降解为主,以竹粉为唯一碳源筛选得到的5株菌株,在属水平上仅占总菌群的0.70%~3.21%,优势可培养菌种为黏质沙雷氏菌(Serratia marcescens)和枯草芽孢杆菌枯草亚种(Bacillus subtilis subsp.subtilis)。筛选获得的可培养菌种中缺少起发酵作用使竹块膨胀的菌种,因此,预处理-优势菌种复配的联合工艺可以作为一种提高竹脱胶效率的手段。

Aim： To investigate mechanisms of tryptase-induced reduction of sperm motility and explore whether epidermal growth factor receptor （EGF-R） and protease activated receptor 2 （PAR-2）- associated pathways are involved. Methods： Fresh semen was collected from healthy donors （n = 15）. Semen parameters and quality were assessed in accordance with the World Health Organization （WHO） criteria. Swim-up sperm were fixed and subjected to immunocytochemistry and immunoelectronmicroscopy with specific antibodies directed against PAR-2 and EGF-R. Protein extractions from swim-up spermatozoa were analyzed by Western blotting with antibodies for both receptors. Motility of spermatozoa was evaluated by computer-assisted semen analysis. Results： Immunocytochemistry found PAR-2 and EGF-R in approximately 30% of examined human ejaculated spermatozoa. Both receptors were localized in the plasma membrane. Like tryptase, the PAR-2 synthetic agonist SLIGKV reduced sperm motility, and this effect was inhibited by application of two specific EGF-R pathway blockers （AG1478 and PD168393）. Conclusion： The observed reduction of sperm motility by tryptase through the PAR-2 receptor involves EGF-R pathways.

There is a very urgent need and clinical demand of robust biomarkers in cancer screening,especially in early hepatocellular carcinoma(HCC)surveillance as outlined by various professional international associations as the European Association for the study of the Liver(EASL),or the Asian Pacific Association for the Study of the Liver(APASL)and national professional organisations as the American Association for the Study of Liver Diseases(AASLD),besides others.Newest data on primarily HCC is still depicting this cancer entity as the sixth most frequently diagnosed cancer,with over 900,000 new cases and over 830,000 deaths worldwide(1).This type of cancer,therefore,remains at the forefront of malignant diseases.Thus,every element of patient management,including early diagnosis and prognosis of treatment response and survival,is a challenge.

Bladder cancer(BC)is one of the most frequently diagnosed types of urinary cancer.Despite advances in treatment methods,no specific biomarkers are currently in use.Targeted and untargeted profiling of metabolites and elements of human blood serum from 100 BC patients and the same number of normal controls(NCs),with external validation,was attempted using three analytical methods,i.e.,nuclear magnetic resonance,gold and silver-109 nanoparticle-based laser desorption/ionization mass spectrometry(LDI-MS),and inductively coupled plasma optical emission spectrometry(ICP-OES).All results were subjected to multivariate statistical analysis.Four potential serum biomarkers of BC,namely,isobutyrate,pyroglutamate,choline,and acetate,were quantified with proton nuclear magnetic resonance,which had excellent predictive ability as judged by the area under the curve(AUC)value of 0.999.Two elements,Li and Fe,were also found to distinguish between cancer and control samples,as judged from ICP-OES data and AUC of 0.807(in validation set).Twenty-five putatively identified compounds,mostly related to glycans and lipids,differentiated BC from NCs,as detected using LDI-MS.Five serum metabolites were found to discriminate between tumor grades and nine metabolites between tumor stages.The results from three different analytical platforms demonstrate that the identified distinct serum metabolites and metal elements have potential to be used for noninvasive detection,staging,and grading of BC.

Recoverable strain induced by martensitic transformation during holding of NiTi-based shape memory alloys under constant stress and temperature

The aim of the present paper was to study a new phenomenon – the strain variation in the NiTi-based shape memory alloys during isothermal holding under a stress. The recoverable strain was studied in NiTiand TiHfNiCualloys on holding under a stress after cooling under a stress (Regime 1)， after active loading (Regime 2)， or during holding without stress in the sample that demonstrates the two-way shape memory effect (Regime 3). The strain variation was found during holding under a stress in all regimes. This strain was recovered on subsequent heating or unloading thus， the strain variation on holding was caused by the isothermal martensitic transformation. This isothermal strain depended on the chemical composition of the alloy， regime for the isothermal holding， the stress and holding temperature. The maximum isothermal strain was 3.4% in TiHfNiCualloy (Regime 1) and 6.1% in NiTialloy (Regime 2). Holding in Regime 3 was accompanied by a small strain (less than 0.3%). It was discussed the influence of the stored elastic energy on isothermal martensitic transformation on holding and it was shown that a large isothermal strain was found if the transformation was accompanied by a small stored elastic energy.

Arabinogalactan proteins(AGPs)are proteoglycans challenging researchers for decades.However,despite the extremely interesting polydispersity of their structure and essential application potential,studies of AGPs in fruit are limited,and only a few groups deal with this scientific subject.Here,we summarise the results of pioneering studies on AGPs in fruit tissue with their structure,specific localization pattern,stress factors influencing their presence,and a focus on recent advances.We discuss the properties of AGPs,i.e.,binding calcium ions,ability to aggregate,adhesive nature,and crosslinking with other cell wall components that may also be implicated in fruit metabolism.The aim of this review is an attempt to associate well-known features and properties of AGPs with their putative roles in fruit ripening.The putative physiological significance of AGPs might provide additional targets of regulation for fruit developmental programme.A comprehensive understanding of the AGP expression,structure,and untypical features may give new information for agronomic,horticulture,and renewable biomaterial applications.

研究NiTi形状记忆合金在应力下等温保温过程中的应变变化。为了研究可恢复应变,对Ni_(51)Ti_(49)和Ti_(40.7)Hf_(9.5)Ni_(44.8)Cu_(5)合金进行3种制度的保温处理,分别是:应力下冷却后在应力下保温(制度1);无应力和载荷下冷却后在应力下保温(制度2);具有双向形状记忆效应的样品在应力下保温(制度3)。结果表明,所有制度中,在应力下保温后样品均会发生应变变化。该应变在后续热处理或卸载后能恢复,因此表明应变变化是由等温马氏体相变引起的。这种等温应变取决于合金的化学成分、等温保温制度、应力和保温温度。Ti_(40.7)Hf_(9.5)Ni_(44.8)Cu_(5)合金(制度1)和Ni_(51)Ti_(49)合金(制度2)的最大等温应变分别为3.4%和6.1%。制度3中的保温伴随着较小的应变(小于0.3%)。讨论储存弹性能对等温马氏体相变的影响,结果表明,当相变伴随着较小的储存弹性能时,等温应变较大。

Algae(and their extracts)are increasingly important for pharmaceutical applications due to the diversity of useful compounds they contain.The genus Fucus contains one of the most studied species,Fucus vesiculosus.The species F.ceranoides differs from the others of the genus by presenting longitudinal air-vesicles and a capacity to survive at low salinities.It is an alga that inhabits the Mondego River estuary(Portugal),at the southern limit of its distribution,and can serve as a role model to understand the effect of a salt gradient on the production of bioactive compounds.We assessed the phenolic content and antioxidant activity of different F.ceranoides extracts(e.***.methanolic,aqueous and polysaccharide)prepared from samples harvested from two different zones to evaluate if the adaptation of F.ceranoides to different salinity levels influenced its chemical composition.The antioxidant activity of the extracts was determined using 1,2-diphenyl-picrylhydrazyl(DPPH)and 2.2-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)(ABTS)radicals.These assays demonstrated that the methanolic extract of lyophilized F.ceranoides that grew at low salinities was the most bioactive,i.e.DPPH(IC 50=50.39μg/mL)and ABTS(TEAC=2.42).The total phenolic content(Folin-Ciocalteu method)and the methanolic extract of the lyophilized F.ceranoides collected from a low salinity habitat exhibited the highest phenolic content(PGE=49.48μg/mg of lyophilized extract)amongst those sampled.Thin layer chromatography(TLC)and Fourier transform infrared spectroscopy(FTIR)were used for the identification of compounds in the extracts.This characterization allowed confirmation that the various extracts contained almost the same compounds but with notable quantitative differences.Based on these results,we conclude that there were differences in the quantity of the compounds due to the effect of salinity.The drying methods used were also found to have influenced the quality of the extracted compounds.
