Hyperlipidemia is a major risk factor for erectile dysfunction(ED).Oxidative stress and phenotypic modulation of corpus cavernosum smooth muscle cells(CcSMCs)are the key pathological factors of ED.n-acetylcysteine(nAC)can inhibit oxidative stress;however,whether nAc can alleviate pathological variations in the corpus cavernosum and promote erectile function recovery in hyperlipidemic rats remains unclear.A hyperlipidemia model was established using 27 eight-week-old male Sprague-Dawley(SD)rats fed a high-fat and high-cholesterol diet(hyperlipidemic rats,HR).In addition,9 male SD rats were fed a normal diet to serve as controls(nC).HR rats were divided into three groups:HR,HR+normal saline(nS),and HR+nAC(n=9 for each group;nS or nAc intraperitoneal injections were administered daily for 16 weeks).Subsequently,the lipid profiles,erectile function,oxidative stress,phenotypic modulation markers of ccsMCs,and tissue histology were *** experimental results revealed that erectile function was significantly impaired in the HR and HR+nS groups,but enhanced in the HR+nAC *** lipid levels,over-activated oxidative stress,and multi-organ lesions observed in the HR and HR+nS groups were improved in the HR+nAC ***,the HR group showed significant phenotypic modulation of CCSMCs,which was also inhibited by.nAC *** report focuses on the therapeutic effect of nAc in restoring erectile function using a hyperlipidemic rat model by preventing CcSMC phenotypic modulationand attenuating oxidativestress.

Objective To explore the neuroprotective effects of the Shaoyao Gancao decoction(SGD)against excitatory damage in PC12 cells and the role of the Src-nR2-nnOS pathway mediation by SGD in regulatingγ-aminobutyric acid(GABA)-glutamate(Glu)*** n-Methyl-d-aspartic acid(nMDA)was used to establish a PC12 cell excitability injury *** investigate the neuroprotective effect of SGD,a cell counting kit-8(CCK-8)assay was used to determine PC12 cell viability,Annexin V/Propidium Iodide(Annexin V/PI)double staining was used to determine PC12 cell apoptosis,and Ca^(2+)concentration was observed using laser confocal *** receptor agonists and antagonists were used to analyze the neuroprotective interactions betweenγ-aminobutyric acid(GABA)and nMDA ***,molecular biology techniques were used to determine mRnA and protein expression in the Src-nR2-nnOS *** analyzed the correlations between the regulatory sites of GABA and nMDA interactions,excitatory.neurotoxicity,and brain damage at the molecular *** nMDA excitotoxic injury manifested as a significant decrease in cell activity,increased apoptosis and caspase-3 protein expression,and a significant increase in intracellular Ca^(2+)*** of SGD,a GABAA receptor agonist(muscimol),or a GABAB receptor agonist(baclofen)decreased intracellular Ca^(2+)concentrations,attenuated apoptosis,and reversed nMDA-induced upregulation of caspase-3,Src,nMDAR2A,nMDAR2B,and ***,a GABA_(A)receptor antagonist(bicuculline)and a GABA_(B)receptor antagonist(saclofen)failed to significantly increase excitatory *** Taken together,these results not only provide an experimental basis for SGD administration in the clinical treatment of central nervous system injury diseases,but also suggest that the Src-nR2A-nnOS pathway may be a valuable target in excitotoxicity treatment.

微塑料是近年来广受关注的一类新污染物.淡水系统沉积物作为微塑料重要的汇,能反映出当地微塑料的长期污染水平.为探究典型城市河网沉积物中微塑料的时空分布特征,于2018年12月和2019年6月在望虞河西岸河网区分别采集了17个采样点的沉积物样品,使用体视显微镜和显微傅立叶变换红外光谱仪对微塑料进行形态观察以及成分鉴定.结果表明,微塑料的平均丰度(以dw计)为323.37 n·kg^(-1),形状主要为碎片和纤维,颜色以黑色和蓝色为主,粒径主要分布在50~500μm之间,成分以PET、PE、PVE、PS和PP为主.夏季微塑料的丰度低于冬季,河网区上游地区微塑料的丰度高于中游和下游地区,最高丰度出现在污水处理厂的出水口采样点,沉积物中微塑料的丰度与表层水中微塑料的丰度无显著相关性.应用正定矩阵因子分解(PMF)模型解析微塑料的来源,发现微塑料的主要来源有农用塑料薄膜、生活废水、塑料垃圾以及工业生产.研究可为典型城市河网微塑料污染防控提供支撑.

The main protease(M^(pro))of SARS-CoV-2 is an attractive target in anti-COVID-19 therapy for its high conservation and major role in the virus life *** covalent M^(pro)inhibitor nirmatrelvir(in combination with ritonavir,a pharmacokinetic enhancer)and the non-covalent inhibitor ensitrelvir have shown efficacy in clinical trials and have been approved for therapeutic *** antiviral drugs are needed to fight the pandemic,while non-covalent M^(pro)inhibitors could be promising alternatives due to their high selectivity and favorable *** non-covalent M^(pro)inhibitors with desirable properties have been developed based on available crystal structures of M^(pro).In this article,we describe medicinal chemistry strategies applied for the discovery and optimization of non-covalent M^(pro)inhibitors,followed by a general overview and critical analysis of the available *** viewpoints and insights into current strategies for the development of non-covalent M^(pro)inhibitors are also discussed.
