Autoantibody profiles in autoimmune hepatitis and chronic hepatitis C identifies similarities in patients with severe disease

To determine how the auto-antibodies (Abs) profiles overlap in chronic hepatitis C infection (ChC) and autoimmune hepatitis (AIh) and correlate to liver *** of antinuclear Ab, smooth muscle antibody (SMA) and liver/kidney microsomal-1 (LKM-1) Ab and markers of liver damage were determined in the sera of 50 patients with ChC infection, 20 AIh patients and 20 healthy controls using enzyme linked immunosorbent assay and other immune *** found that AIh patients had more severe liver disease as determined by elevation of total IgG, alkaline phosphatase, total serum bilirubin and serum transaminases and significantly higher prevalence of the three non-organ-specific autoantibodies (auto-Abs) than ChC patients. Antinuclear Ab, SMA and LKM-1 Ab were also present in 36% of ChC patients and related to disease severity. ChC cases positive for auto-Abs were directly comparable to AIh in respect of most markers of liver damage and total IgG. These cases had longer disease duration compared with auto-Ab negative cases, but there was no difference in gender, age or viral load. KLM-1+ Ab ChC cases showed best overlap with ***-Ab levels in ChC may be important markers of disease severity and positive cases have a disease similar to AIh. Auto-Abs might have a pathogenic role as indicated by elevated markers of liver damage. Future studies will unravel any novel associations between these two diseases, whether genetic or other.

Numerical simulations of smoke spread during solar roof fires

As the installation of solar roofs increases,so has the concern over *** from a solar roof fire could spread into a building through roof openings and presents a challenge for existing fire protection *** date,there have been insufficient studies on solar roof fire-induced smoke *** this study,we conducted computational fluid dynamics(CDF)simulations using Fire Dynamics Simulator(FDS)to better understand the mechanisms of solar roof fire-induced smoke spread and help with solar roof ***,the photovoltaic(PV)combustion model was created in FDS and validated by experimental data.A parametric study was then simulated to investigate the impacts of roof slopes and vent sizes on the smoke spread of the solar *** was found that the roof slope has a significant effect on the fire smoke *** the roof slope increases,the region of separation,where the smoke and air are mixed,can extend from the leeward side of the building to the roof *** a result,smoke could fill the attic and room more slowly,leading to a lower soot density and lower indoor *** design a solar roof,both fire smoke protection and PV energy performance should be considered,especially for the low latitude regions where the PV optimal title angle regarding energy performance is small and leads to a higher risk of smoke infiltration.

The comparable feature analysis of NAMD and GROMACS molecular dy-namics packages has been *** benchmarks of 72 and 128 Dipalmitoylphos-phatidylcholine(DPPC)/water have been constructed using a cluster(3Ghz-Xeon proces-sors and Myrinet network)and the comparison has been performed using GROMOS87 and ChARMM27 force fields modified for lipids with GROMACS and NAMD software packages,*** GROMACS has been displayed as faster than NAMD,likely due to united-atom character of GROMACS and good implementation *** GROMACS reaches saturation and goes to the worst results,the reason of which is that the program spends more time on communications between processors.

Assessment of trace metals contamination in stream sediments and soils in Abuja leather mining, southwestern Nigeria

This study is aimed at determining the level of environmental degradation as well as the concentration of trace elements in soil and stream sediments in order to evaluate the environmental impact of the mining ***-five(25) soils and ten(10) stream sediment samples were collected from the study area. The physicochemical parameters were determined using appropriate instrumentation with the aid of a digital p h meter(Milwaukee meter) to measure the p h and electrical conductivity, total dissolved solids, moisture content and loss on ignition of the soil and stream sediment samples. The p h of the soil sample ranged from(6.10 to 7.19); Electrical conductivity ranged from(21.3 to 279.4 μS/cm), moisture content varied from(0.60% to 7.20%), and the LOI ranged from(2.03% to 18.62%). The results of the analysis showed that the concentrations of the trace elements in the soils and stream sediment samples were slightly higher than the background values. Plots of the trace elements in stream sediment samples show moderate, consistent decrease downstream except at points where there was mine water discharge into the main river. The pollution levels of heavy metals were examined in stream sediment and soil samples using different assessable indices, such as the enrichment factor, which showed significant-moderate enrichment for Cr, Th, Nb, Zn, Pb, Y and Zr and the geoaccumulation index, which showed practically moderate contamination with Cr, Ni and Sr based on regional background reference values. Geo-accumulation index and contamination index for soils and stream sediment revealed uncontaminated to moderate contamination. Likewise,elements with moderate contamination were Cr, Ni and *** Pearson correlation showed that there were significant positive associations among selected metals in soil and stream sediment samples.

Promoting effect of licorice extract on induction of β-glucuronidase in Penicillium purpurogenum Li-3

PeniciUium purpurogenum Li-3, a fungus producing β-glucuronidase （PGUS）, can con- vert glycyrrhizin （GL） to glycyrrhetinic acid monoglucuronide （GAMG） when grown in medium with GL as the sole carbon source. In order to improve the conversion rate of GL and the yield of GAMG, licorice extract （LE） was added as an inducer to enhance the production of GAMG by the PGUS. In this work, the influence of LE on the conversion rate of GL to GAMG was studied. When the Penicil- lium purpurogenum Li-3 was grown in the medium containing LE and GL （ concentration ratio of LE to GL was 2： 3）, the conversion rate of GL was 84. 12% with 38. 18% increase and the yield of GAMG was 80. 47% with 37. 18% increase, comparing with to the medium only containing GL at 48 h. The enzyme activity of ^-glucuronidase was also enhanced from 22. 4 U/mL to 82.3 U/mL, which in- creased up to about 3. 67 fold. The results showed that LE could significantly improve the induced expression level of PGUS.

A series of novel unsymmetrical N-（4-oxo-2-phenyl-3（4h）-quinazolinoyl）-N-（aryl） acetamidines was synthesized by reacting ethyl（1h）-N-（4-oxo-2-phenylquinazolin-3（4h）-y1）ethanimidoate（2） and suitable reactive aromatic ***＇ determination of the synthesized compounds was carried out using spectroscopic techniques including IR,~1h NMR,and mass spectrometry. Structural effects on reactivity were also studied.

Purification and characterization of Chpro1, a thermotolerant, alkali-stable and oxidation-resisting protease of Chumathang hotspring

Metagenomic approaches are recently used for searching novel open reading frames （ORFs） coding enzymes employed in pharmaceutical, rood industries, etc. In this study, a metagenornic library was constructed from Chumathang hotspring sediment DNA. The library consisted of approximately 9,000 clones and was screened for protease activity. A clone exhibiting protease activity was identified and named Chprol. Sequencing of Chprol revealed that the ORF encoded a functional protein of 363 amino acids belonging to peptidase S8-S53 superfamily. Chprol shared 41% sequence similarity with a reported protease （subtilase family） and 35 % structural similarity with the crystal structure of Pro-Tk sps. of Thermococcus kodarkaenasis. In silico modeling the 3D structure of Chprol showed that it has two beta sheets, 10 alpha helices and 11 strands. Catalytic triad prediction implied Chprol to be a serine protease. The optimum temperature and ph of the purified protease were found to be 80 ℃ and 11.0, respectively. The enzyme was active at 5 % concentration of hydrogen peroxide and retained 60 % of activity at 10 % concentration. The thermotolerant, alkalophilic and oxidant stable properties of the protease make it a potential can- didate for biotechnological applications.

Piperine,an active ingredient of black pepper attenuates acetaminophen-induced hepatotoxicity in mice

Objective:To explore the hepatoprotective and antioxidant effects of piperine against acetaminophen-induced hepatotoxicity in ***:In mice,hepatotoxicity was induced by a single dose of acetaminophen(900 mg/kg b.w.i.p.).Piperine(25 mg/kg b.w.i.p.) and standard drug silymarin(25 mg/kg b.w.i.p.) were given to mice,30 min after the single injection of *** 4 h,the mice were *** of liver marker enzymes [(aspartate transaminase(AST),alanine transaminase(ALT),and alkaline phosphatase(ALP)]and inflammatory mediator tumour necrosis factor-alpha(TNF-α) were estimated in serum,while lipid peroxidation and antioxidant status(superoxide dismutase,catalase,glutathione peroxidase, glutathione reductase,glutathione-s-transferase and glutathione) were determined in liver homogenate of control and experimental ***:Acetaminophen induction(900 mg/kg b.w.i.p.) significantly increased the levels of liver marker enzymes,TNF-α,and lipid peroxidation,and caused the depletion of antioxidant *** and silymarin treatment to acetaminophen challenged mice resulted in decreased liver marker enzymes activity,TNF-αand lipid peroxidation levels with increase in antioxidant ***:The results clearly demonstrate that piperine shows promising hepatoprotective effect as comparable to standard drug silymarin.

Water promoted, microwave-assisted oxidative novel deamination of N-aminoquinazolinones

A novel deamination of 2-alkyl/aryl 3-amino-4（3h）-quinazolinones series using aqueous KMnO4 under thermal condition and microwave irradiation is described. Compared to thermal condition, significantly higher yields in much shorter times were observed for reactions under microwave irradiation. A plausible mechanism has been proposed for the oxidative water-promoted deamination.
