Cinnamyl alcohol dehydrogenase （CAD） is a key enzyme involved in the last step of monolignol biosynthesis. The effect of CAD down-regulation on lignin production was investigated through a transgenic approach in maize. Trans- genic CAD-RNAi plants show a different degree of enzymatic reduction depending on the analyzed tissue and show alter- ations in cell wall composition. Cell walls of CAD-RNAi stems contain a lignin polymer with a slight reduction in the s-to-G ratio without affecting the total lignin content. In addition, these cell walls accumulate higher levels of cellulose and ara- binoxylans. In contrast, cell walls of CAD-RNAi midribs present a reduction in the total lignin content and of cell wall polysaccharides. In vitro degradability assays showed that, although to a different extent, the changes induced by the repression of CAD activity produced midribs and stems more degradable than wild-type plants. CAD-RNAi plants grown in the field presented a wild-type phenotype and produced higher amounts of dry biomass. Cellulosic bioethanol assays revealed that CAD-RNAi biomass produced higher levels of ethanol compared to wild-type, making CAD a good target to improve both the nutritional and energetic values of maize lignocellulosic biomass.

Resilience in a Mexican Pacific Mangrove after Hurricanes: Implications for Conservation Restoration

High resilience is a mangrove trait that is compatible with life in a dynamic environment, however, catastrophic disturbance can lead to the entire forest structure being re-defined. In the Pochutla district of Oaxaca, two hurricanes made landfall in 1997 and one more in 2012. Following the 1997 hurricanes, extensive mangrove restoration was carried out in the study area. A cohort of Rhizophora mangle saplings planted in 2007 showed 10% mortality during the first year after transplantation, an average growth of 39 cm, and a positive association between growth rate and the level of water in the lagoon. Following Hurricane Carlotta of 2012, measurements of structural impact and tree mortality were taken in restored R. mangle stands and in naturally regenerated patches of Laguncularia racemosa. The role of tree girth in R. mangle susceptibility to wind damage was also investigated. The stands of R. mangle suffered 80% reduction in stem density and 86% loss of basal area, whereas the corresponding values for L. racemosa were 26% and 15%, respectively. Within stands of R. mangle, mortality conserved a positive relationship with structural impact categories and the frequency of snapped stems was segregated across girth classes. The results suggest that L. racemosa has greater resilience to hurricane damage, which has some consistency with previous research and implications for conservation restoration protocols. In order to promote a system with higher resilience, we recommend an on-going restoration effort with mixed mangrove species.

The present study aimed to develop a protocol for somatic embryogenesis and encapsulation of coffee embryos(Coffea arabica L.),for the conservation of genotypes with characteristics of commercial *** embryos were induced from leaf explants in Murashige and skoog medium(Ms)supplemented with 1 mg·L^(−1) of 2,4-dichlorophenoxiacetic acid(2,4-D)combined with 2 mg·L^(−1) of benzyladenine(BA).somatic embryos(sE)at the globular stage were encapsulated in a sodium alginate matrix;two treatments were tested:Ms+5 mg·L^(−1) BA+1 mg·L^(−1) NAA+3%(w/v)alginate,and Ms+7 mg·L^(−1) BA+5.7 mg·L^(−1) indoleacetic acid(IAA)+3%(w/v)*** was complexed with 100 mM calcium chloride(CaCl_(2)).Viability of the encapsulated sE was determined by staining with 0.01%fluorescein diacetate(FDA)after 0,15,30,and 45 days of storage at 4℃.Embryo viability was 100%in both treatments.

Comparative study of the efficacy and safety of bromfenac, nepafenac and diclofenac sodium for the prevention of cystoid macular edema after phacoemulsification

AIM： To compare the efficacy, tolerability and safety of bromfenac 0.09%, nepafenac 0.1% or diclofenac 0.1% for the prophylaxis of the cystoid macular edema（CME） after phacoemulsification. METHODs： Group sequential observational comparative study. After phacoemulsification, patients received two months for topical treatment of either diclofenac sodium, bromfenac or nepafenac. All patients received concomitant topical tobramycin 0.3% and topical prednisolone 1%. We measured CME using optical coherence tomography（OCT） central foveal thickness, macular thickness and total macular volume. REsULTs： We enrolled 243 patients from January to June 2015, and 35% received diclofenac, 32.9% bromfenac and 32.1% nepafenac. When we compared pre-operative to three weeks to two months, bromfenac was more effective in reducing foveal volume（21.3 and 35.4 mm3, respectively）, compared with the diclofenac（1.3 and 11.5 mm3, respectively）, and the nepafenac group, became more edematous 6.4 and 5.3, respectively. Totally 133 patients completed the post-surgical satisfaction questionnaire. Patients complained of eye stickiness in 13.8% whom we gave nepafenac, versus 10.3% whom we gave diclofenac sodium, and in 0 whom we gave bromfenac. CONCLUsION： Bromfenac is the best tolerated and is more effective than diclofenac and nepafenac in reducing CME after phacoemulsification.

Laparoscopic surgery is applied today worldwide to most digestive procedures. In some of them, such as cholecystectomy, Nissen's fundoplication or obesity surgery, laparoscopy has become the standard in practice. In others, such as colon or gastric resection, the laparoscopic approach is frequently used and its usefulness is unquestionable. More complex procedures, such as esophageal, liver or pancreatic resections are, however, more infrequently performed, due to the high grade of skill necessary. As a result, there is less clinical evidence to support its implementation. In the recent years, robot-assisted laparoscopic surgery has been increasingly applied, again with little evidence for comparison with the conventional laparoscopic approach. This review will focus on the complex digestive procedures as well as those whose use in standard practice could be more controversial. Also novel robot-assisted procedures will be updated.

Degradation of Indigo Carmine Using Advanced Oxidation Processes: synergy Effects and Toxicological study

The physicochemical degradation of Indigo Carmine (IC) dye in aqueous solution was performed using single and combined Advanced Oxidation Processes (AOP’s). Photocatalysis (TiOsub>2sub>-UV), Ozonation (Osub>3sub>) and sonolysis (sN) were tested either in a standalone methodology or by combination of two simultaneous AOP’s. The dye conversion was followed by both measurements: 1) color removal determined by UV-VIs spectrometry and 2) organic and inorganic load determined by the chemical oxygen demand (COD). A complete and quick color disappearance of model water waste has been obtained by using combination of non-irradiated AOP’s, namely, O3sub>/sN, which contrasts to the combination of irradiated photocatalysis with O3sub> or sonolysis. Color removal with simultaneous TiO2sub>-UV/sN reached 77% while TiO2sub>-UV/O3sub> reached 96% at similar reaction time. On the other hand, the standalone O3sub> yielded the highest color removal (94.4%) in 32 minutes whereas sN reached only 39.2% in 4 hours. The standalone light irradiated TiO2sub>-UV reached 93.3% color re-moval in one hour of reaction time. These results indicated that non-irradiated (sN and O3sub>) enhance synergistic effects that provoke structural changes in dye molecule without reaching total degradation. This is evidenced from FTIR of residuals from reaction mixture in which it has been observed the presence of organic molecules such as aromatics, sulfonic and amines refractory compounds that are mechanisti-cally possible to be found during IC degradation. Also, toxicity tests (MicroToxsup>?sup> Technique) were performed using commercially available bacteria culture before and after IC degradation for each AOP and their combination. Reduction of aqueous dye concentration decreased the level of toxicity of the treated water which is the main target of the AOP’s but the presence of the remaining recalcitrant compounds have also toxic effect.

Determining the specific status of the Iberian sturgeons by means genetic analyses of old specimens

To clarify the species status of sturgeon from rivers of the Iberian Peninsula, eight molecular markers (4 nuclear and 4 mitochondrial) have been analysed in different specimens from historical museum samples and prehistoric samples from archaeological sites. These analyses indicate that one of these specimens (UGP captured in the Guadalquivir River in the 19th century) is A. sturio, based on all the eight molecular markers, four of them used from the first time in this study. In previous analyses based on 5 genetic markers, our group assigned two specimens captured in this river in the 1970-80s (EBD8173 and EBD8401) to the species A. naccarii, suggesting the presence of this species in the Iberian Peninsula. In this work, this conclusion is drawn after successfully obtaining a mitochondrial marker in a very old scute from a prehistoric site (Acinipo, about 1500 BC, from the Guadalquivir River basin). On the other hand, in the specimen EBD8174 captured in the Guadalquivir in 1975, we have obtained two new mitochondrial markers confirming that it can be considered A. sturio for all the mitochondrial markers, but nuclear ones identify it as A. naccarii. Finally, two very old samples (Nerja E-VI and Nerja N/62-63) were not successfully characterized by any molecular markers. some aspects and consequences of our results are discussed, such as the origin of the “mosaic” specimen EBD8174 and, above all, the native status of A. naccarii in historic and prehistoric times in the southern Iberian Peninsula.

Thoracic Unilateral spinal Cord syndrome Following Neuraxial Anaesthesia for Periprosthetic Knee Fracture: A Case Report

Neuraxial anaesthesia is widely used in surgical procedures;overall, epidural and intrathecal techniques. Nevertheless, several outcomes should be considered. The incidence of neurologic complications after neuraxial anaesthesia is not perfectly clear (0% - 0.08%), although there are several described cases of spinal cord ischemia. We present a case of thoracic unilateral spinal cord syndrome following lumbar spinal anaesthesia for periprosthetic knee fracture. Our patient suffered monoparesis in her left lower limb as well as decreasing of muscle strength and loss of tendon reflexes. The MNR showed left hyperintense intra-cord images from T7 to T12 attributed to spinal cord oedema and a lineal hypointensity related to minimal haematic component. What made this case surprising was the fact that spinal anaesthesia was performed between L3 and L4 and the patient did not suffer paraesthesia associated with local anaesthetic injection. she was treated with glucocorticoids, gabapentin and amitriptyline. she also was checked by physical rehabilitators, neurologists and Pain Unit physicians. We have found another case reported in the literature about thoracic cord injury after lumbar spinal puncture. In this paper, we report possible aetiologies according to a review and neurological evolution of the patient seven months later.

Agave americana *** were exposed to different concentrations of ethyl methanesulphonate(EMs)0,15,30,45 and 60 mM and to different times of exposure(2 and 4 h).The viability and capacity of shoot formation were shown to be affected when the callus were exposed to high concentrations(30–60 mM).Only the callus exposed to 15 mM EMs presented shoot formation;the exposure time of two hours produced the largest quantity of shoots regenerated per callus(21 shoots/callus).In order to generate somaclonal variants resistant to Fusarium oxysporum,a selection pressure was applied through of a culture filtrate(CF)of 100 ppm of the *** was made in callus obtained in the treatment with 15 mM EMs during 2 h of *** CF caused oxidation and necrosis in 71.25%of the callus;however,they were capable of generating shoots(3.5 shoots/callus).Molecular markers type RAPD,IssR and DAMD were used to evaluate the genetic variation arising from the mutations caused by EMs on control plants and 16-month-old somaclonal *** polymorphic information content(PIC)for each one of the initiating groups was:0.28(DAMD),0.09(IssR)and 0.14(RAPD).DAMD revealed a greater percentage of polymorphism than RAPD and *** bands were detected in the somaclonal *** indicated that the EMs caused genetic variation in the regenerated plants conferring resistance to them against Fusarium oxysporum.
