Efficiency in hydrocarbon degradation and biosurfactant production by Joostella sp.A8 when grown in pure culture and consortia

Joostella strains are emerging candidates for biosurfactant production. Here such ability was analyzed for Joostella strain A8 in comparison with Alcanivorax strain A53 and Pseudomonas strain A6, all previously isolated from hydrocarbon enrichment cultures made of polychaete homogenates. In pure cultures Joostella sp. A8 showed the highest stable emulsion percentage（78.33%）, hydrophobicity rate（62.67%）, and an optimal surface tension reduction during growth in mineral medium supplemented with diesel oil（reduction of about 12 mN/m）, thus proving to be highly competitive with Alcanivorax and Pseudomonas strains. During growth in pure culture different level of biodegradation were detected for Alcanivorax strain A53（52.7%）,Pseudomonas strain A6（38.2%） and Joostella strain A8（26.8%）. When growing in consortia,isolates achieved similar abundance values, with the best efficiency that was observed for the Joostella-Pseudomonas co-culture. Gas-chromatographic analysis revealed an increase in the biodegradation efficiency in co-cultures（about 90%）, suggesting that the contemporary action of different bacterial species could improve the process. Results were useful to compare the efficiencies of well-known biosurfactant producers（i.e. Pseudomonas and Alcanivorax representatives） with a still unknown biosurfactant producer, i.e. Joostella, and to confirm them as optimal biosurfactant-producing candidates.

Prevalence and ecological factors affecting the distribution of plant-parasitic nematodes in Prunus groves in Spain

A wide survey was conducted to study plant-parasitic nematodes(PPNs)associated with Prunus groves in *** research aimed to determine the prevalence and distribution of PPNs in Prunus groves,as well as the influence of explanatory variables describing soil,climate and agricultural management in structuring the variation of PPNs community composition.A total of 218 sampling sites were surveyed and 84 PPN species belonging to 32 genera were identified based of an integrative taxonomic *** species considered as potential limiting factors in Prunus production,such as Meloidogyne arenaria,***,***,Pratylenchus penetrans and ***,were identified in this *** soil physico-chemical(C,Mg,N,Na,OM,P,pH and clay,loamy sand and sandy loam texture classes),four climate(Bio04,Bio05,Bio13 and Bio14)and four agricultural management variables(grove-use history less than 10 years,irrigation,apricot seedling rootstock,and Montclar rootstock)were identified as the most influential variables driving spatial patterns of PPNs *** particular,younger plantations showed higher values for species richness and diversity indices than groves cultivated for more than 20 years with Prunus *** study increases the knowledge of the distribution and prevalence of PPNs associated with Prunus rhizosphere,as well as on the influence of explanatory variables driving the spatial structure PPNs communities,which has important implications for the successful design of sustainable management strategies in the future in this agricultural system.

Four Cases of X-Linked Hypophosphatemic Rickets, Clinical Description and Genetic Testing

One of the major causes of congenital hypophosphatemic rickets is the X-linked hypophosphatemic rickets (XHR), due to a defect on PHEX gene. The XHR increases the renal elimination of phosphate, that condition leads a defective mineralization of bones and also affects the growth in children. Clinical diagnosis should be suspected in children with signs of rickets and hypophosphatemia with normal calcium levels. We describe clinical characteristics and genetic results of four patients diagnosed and treated in our Nephrology Section. All patients have a “de novo” XHR as none familiars are affected. Early diagnosis should be suspected before the bone deformities have been submitted and the growth would have been impaired.

Sensory analysis of Creole turkey meat with flash profile method

Different kind of feed can result in development of new meat odors, aroma and texture. The aim of the study was to use flash profile method to determine differences;if any, in sensory traits of Creole turkey meat with different alimentation programs. Treatments were kitchen leftovers + fresh forage, commercial feed, kitchen leftovers, commercial feed + fresh forage, and broken maize + fresh forage. Cooked thighs, drumstick and breast were used. For the meat evaluation two different groups of people were recruited. In order to perform the sensory profile of breast six persons were recruited, in a second group nine persons;in both cases they did not know what kind of meat they were evaluating. Each sensory profile had three replicas, previously three training sessions and establishment of sensory attributes criteria were held. Attribute discrimination was evaluated one-way ANOVA. To obtain consensus and treatment mean position, attributes of the subjects a Generalized Procrustes Analysis was used and comparison of treatments through an ascendant hierarchy classification. Thirty five different sensory descriptive were generated. There were differences in meat sensory profile, it can be said that different treatments influenced in different ways the muscle development of Creole turkey, creating new sensory attributes.

Productive evaluation of slow-growing Mexican turkeys with different diets in confinement

Were assessed the effect of five traditional diets backyard turkeys in confinement. The experiment lasted 36 weeks, the turkeys were randomized to one of five diets: kitchen waste + fresh forage (T1), kitchen waste (T2), commercial feed + fresh forage (T3), cracked corn + fresh forage (T4) and commercial feed (T5). Food and water were provided ad libitum. Each treatment with birds of both sexes had two repetitions with four birds and each bird was considered as an experimental unit. Were evaluated daily weight gain, total weight gain, final live weight, total length peak-tail, monthly gain peak-tail, total length, total length of wings, monthly gain wings length, and feed cost. The data were analyzed in a completely randomized arrangement with adjusted means and Tukey’s mean comparison, then an evaluation of treatment groups by analyzing clusters for semi quantitative data. The biggest daily gain, total weight gain and final live weight was obtained with treatment T3, while the highest increase in peak-length tail and wings was obtained with treatment T1 (P < 0.05). T2 treatment turned out to be most economical, but the treatment that best scores obtained was T1. We conclude that the use of kitchen waste + fresh forage is a feasible strategy to feed slow-growing turkeys.

The use of mesenchymal stem-cells(MSC)in cell therapy has received considerable attention because of their *** properties include high expansion and differentiation in vitro,low immunogenicity,and modulation of biological processes,such as inflammation,angiogenesis and ***,the regenerative effect of MSC is partly due to their paracrine *** has prompted numerous studies,to investigate the therapeutic potential of their secretome in general,and specifically their extracellular vesicles(EV).The latter contain proteins,lipids,nucleic acids,and other metabolites,which can cause physiological changes when released into recipient ***,contents of EV can be modulated by preconditioning MSC under different culture *** them,exposure to hypoxia stands out;these cells respond by activating hypoxia-inducible factor(HIF)at low O_(2) *** has direct and indirect pleiotropic effects,modulating expression of hundreds of genes involved in processes such as inflammation,migration,proliferation,differentiation,angiogenesis,metabolism,and cell *** of these genes is reflected in the contents of secreted ***,numerous studies show that MSC-derived EV conditioned under hypoxia have a higher regenerative capacity than those obtained under *** this review,we show the implications of hypoxia responses in relation to tissue *** addition,hypoxia preconditioning of MSC is being evaluated as a very attractive strategy for isolation of EV,with a high potential for clinical use in regenerative medicine that can be applied to different pathologies.

Cirrhosis complicating Shwachman-Diamond syndrome: A case report

BACKGROUND The features of Shwachman-Diamond syndrome (SDS) include exocrine pancreatic insufficiency, skeletal abnormalities and bone marrow dysfunction;an often overlooked feature is hepatic involvement. CASE SUMMARY We report a child who initially presented with failure to thrive and mildly elevated transaminase levels and was determined to have pancreatic insufficiency due to SDS. During follow-up he had persistently elevated transaminase levels and developed hepatosplenomegaly. An investigation was performed to determine the etiology of ongoing liver injury, including a liver biopsy which revealed hepatic cirrhosis. CONCLUSION Cirrhosis has rarely been reported with SDS. While many of the hepatic disorders associated with SDS improve with age, there are rare exceptions with serious implications for long-term outcome.

Rectal tone and compliance affected in patients with fecal incontinence after fistulotomy

AIM: To investigate the anal sphincter and rectal factors that may be involved in fecal incontinence that develops following fistulotomy(FIAF).METHODS: Eleven patients with FIAF were compared with 11 patients with idiopathic fecal incontinence and with 11 asymptomatic healthy subjects(HS). All of the study participants underwent anorectal manometry and a barostat study(rectal sensitivity, tone, compliance and capacity). The mean time since surgery was 28 ± 26 mo. The postoperative continence score was 14 ± 2.5(95%CI: 12.4-15.5, St Mark's fecal incontinence grading system).RESULTS: Compared with the HS, the FIAF patients showed increased rectal tone(42.63 ± 27.69 vs 103.5 ± 51.13, P = 0.002) and less rectal compliance(4.95 ± 3.43 vs 11.77 ± 6.9, P = 0.009). No significant differences were found between the FIAF patients and the HS with respect to the rectal capacity; thresholds for the non-noxious stimuli of first sensation, gas sensation and urge-to-defecate sensation or the noxious stimulus of pain; anal resting pressure or squeeze pressure; or the frequency or percentage of relaxation of the rectoanal inhibitory reflex. No significant differences were found between the FIAF patients and the patients with idiopathic fecal ***: In patients with FIAF, normal motor anal sphincter function and rectal sensitivity are preserved, but rectal tone and compliance are impaired. The results suggest that FIAF is not due to alterations in rectal sensitivity and that the rectum is more involved than the anal sphincters in the genesis of FIAF.

Clostridium difficile (C. difficile) is a spore-forming, toxin-producing, gram-positive anaerobic bacterium that is the principal etiologic agent of antibiotic-associated diarrhea. Infection with C. difficile (CDI) is characterized by diarrhea in clinical syndromes that vary from selflimited to mild or severe. Since its initial recognition as the causative agent of pseudomembranous colitis, C. difficile has spread around the world. CDI is one of the most common healthcare-associated infections and a significant cause of morbidity and mortality among older adult hospitalized patients. Due to extensive antibiotic usage, the number of CDIs has increased. Diagnosis of CDI is often difficult and has a substantial impact on the management of patients with the disease, mainly with regards to antibiotic management. The diagnosis of CDI is primarily based on the clinical signs and symptoms and is only confirmed by laboratory testing. Despite the high burden of CDI and the increasing interest in the disease, episodes of CDI are often misdiagnosed. The reasons for misdiagnosis are the lack of clinical suspicion or the use of inappropriate tests. The proper diagnosis of CDI reduces transmission, prevents inadequate or unnecessary treatments, and assures best antibiotic treatment. We review the options for the laboratory diagnosis of CDI within the settings of the most accepted guidelines for CDI diagnosis, treatment, and prevention of CDI.
