The aim of this study was to evaluate the effects of silver nitrate in the in vitro development and conservation of passion fruit plants. Experiments were carried out at the Laboratory of Culture and Tissues, in the National Cassava and Fruits Research Center (EMbrAPA), using microcuttings of Passiflora gibertii N. E. br., previously cultivated in vitro. The microcuttings were placed in MS media supplemented with silver nitrate at concentrations of 0, 1, 2, 4 and 8 mg·L-1, and, during 30, 60 and 90 days, the following variables were identified: Shooting length (cm), number of roots, number and coloration of leaves. A completely randomized design with 30 replications in a split-plot scheme was employed. Polynomial regression equations were adjusted in analysis of variance to compare averages of treatments. The obtained results support the assumption of high sensitivity of passion fruit tissues to ethylene, which reflects in the lost of vigor and in the induction of culture senescence by its accumulation. Based on obtained results, and in order to mitigate this problem, it is possible to indicate an addition of 2 mg·L-1?of silver nitrate in culture media, for the micropropagation of passion fruit plant, allowing the controlling of the atmosphere inside the culture test tubes, during the establishment of explants and other steps of the process.

Byrsonima intermedia A Juss. is a species with pharmacological properties from the brazilian Cerrado that shows difficulties related to sexual propagation. Research on cell viability may provide useful information for the selection of cells with embryogenic potential during the callus culture, Within this context, our research is aimed at establishing the cell viability of calli from Byrsonima intermedia leaf segments. The calli went through three subculture phases, of 60 days each, in MS medium with 0.09 M sucrose, 0.6% agar, pH 5.8 and 4.52 laM 2,4-D. The calli were stored in dark conditions and samples were collected every 10 days from each subculture for viability tests with fluorescein 3,6-diacetate （FDA） and 2,3,5-triphenyltetrazolium chloride （TTC）. The staining methods allowed quantifying cell viability in each subculture. The best results from the FDA tests were obtained at 21, 25 and 29 days for the first, second and third subcultures respectively, with 53,86%, 61.88% and 53.73% viable cells. Regarding the TTC test, the largest absorbance values were obtained at 21, 27 and 28 days for the first, second and third subcultures respectively. Fluorescence and spectrophotometry analyses were efficient for determination of cell viability during callus cultivation period.

The abscisic acid (ABA) is related to the formation of certain polyphenols, such as anthocyanin, improving the nutritional and commercial quality of the red grapes. In this study, we verified whether the exogenous application of?S-ABA favors the accumulation of anthocyanin, the total phenolic compounds and the antioxidant activity of the?‘Rubi’?grape. The experiment was performed during the cycle 2014/2015 with the?‘Rubi’?vine (Vitis vinifera?L.) grafted in the rootstock 420-A. The experimental design was composed of randomized blocks, with four treatments and six blocks, in a total of 24 experimental parcels, constituted of one plant each. We used the?S-ABA isomer in different concentrations and times of application, 400 mg·L-1?S-ABA in the beginning of the ripening (br);400 mg·L-1?at br + 200 mg·L-1?at 25 days after first application (DAFA);400 mg·L-1at br + 400 mg·L-1?at 25 DAFA;besides the control (0 mg·L-1). After 40 days of the first application, the samples were harvested and we verified that the?S-ABA induced the accumulation of anthocyanins and total phenolic compounds in the?‘Rubi’?grape. From these results we can recommend two applications of?S-ABA, 400 mg·L-1?at br + 200 mg·L-1?at 25 DAFA.

The goal of this work is to perform the risks analysis of the mission NANOSATC-br1 and NANOSATC-br2 and then compare them linearly. The NANOSATC-br1 and NANOSATC-br2 are the first and the second satellite, respectively. They belonged to the project NANOSATC-br-development of CubeSats, which is performed in the facilities built by the partnership between the National Institute of Space Research and the Technological Center from Federal University of Santa Maria. The project focuses on the development of a scientific instrumentation and, simultaneously, the design development, construction, qualification and launch of a national scientific nanosatellite, in a cube shape with 100 mm of edge and near to 1.33 kg of mass, per unit （U）. The risk analysis was held to identify and minimize the project＇s risks of failure, due to its complexity, assuring the mission success, preventing extra pays and rework. The software, CubeSat Mission Design Software Tool for Risk Estimating, which uses statistical regression methods, was used. So, we were capable to measure the project＇s most critical steps assuring its success. The NANOSATC-br1 was launched in June 19 and it is orbiting the Earth in a nominal regime and the NANOSATC-br2 has been scheduled to be launched in 2016.

Microbial Attributes of Infested Soil Suppressive to Bacterial Wilt by Bokashi Amendments

Bacterial wilt, caused by Ralstonia solanacearum, is a major tomato disease in tropical and sub-tropical regions. It is difficult to be managed, since no single measure confers significant contribution for disease control. Among the cultural practices available for disease management, bokashi provides nutrients to the plants, increasing the microbial biomass, improving the quality of the soil and, in some cases, protecting plants against diseases. In this work, we evaluated the effect of three different bokashis (Embrapa—BE;poultry—BP and cattle—BC) in two soils artificially and naturally infested with R. solanacearum, on the suppression of bacterial wilt in tomato. Disease control is discussed upon measurements on the contents of microbial biomass carbon (MBC), on total organic carbon (TOC), on basal respiration (br), on metabolic coefficient (qCO2) and on microbial coefficient (qMIC). The experiment was implemented in greenhouse, with completely randomized design and factorial arrangement of treatments 2 × 3 (two soils × three bokashis). Disease suppression, assessed through wilt incidence 20 and 30 days after transplanting, was better observed in the naturally infested soil, where BP and BE were more efficient in controling the disease. TOC contents were higher in the artificially infested soil compared to that naturally infested, whereas the qMIC presented higher value for the naturally infested soil, which had greater contribution of MBC. Higher rates of br and qCO2 were observed for the naturally infested soil with BC, probably indicating high plant stress caused by the disease in this treatment. Moreover, a high and positive correlation coefficient was found between the variables qCO2 and the number of infected plants at 30 days after transplanting. In the artificially infested soil, a negative correlation was found between the number of infected plants at 20 days after transplanting and TOC.

The aim of this work was to estimate the adaptability and stability of grain yield per hectare and percentage of crude protein of maize grains combined in an index, and to establish a multicharacter selection through mixed models based on an objective character and 15 auxiliary traits. The trials were conducted in the 2013/2014 agricultural year in four growing environments of the Rio Grande do Sul, br state. The experimental design was randomized blocks arranged in a factorial scheme, being four growing sites × 15 single cross maize hybrids, arranged in three repetitions. The genotypic index, composed by the grain yield and the crude protein percentage in the grains, is the best selection strategy to achieve maize superior genotypes. The multivariate genotypes selection, considering grain yield and crude protein, is efficient. The genotypes FORMULA TL®, AS1656PRO®, P30F53Hx®, LG6304YG®and 30F53 are more adapted and stable for grain yield and percentage of crude protein, in the conditions of this study. The mixed models were efficient to employ the multicharacter selection and to contribute for maize genetic breeding.
