Macrophages-Based Immunotherapy for the Brain Diseases

The fundamental difficulties of immunotherapy for the brain diseases are delivery of therapeutic molecules to cross the blood-brain barrier(BBB),to transfect non-dividing neurons and macrophages,and to edit gene in and/or out in the diseased tissues. Regulation of immune responses is particular important in immunotherapy in brain cancer,stroke,traumatic brain injury,and brain infections. Immunotherapy appears to have significant therapeutic potential for the treatment of neurological conditions by regulation of immune responses to prevent irreversible destructive mechanisms. The systemic and brain immune responses to treatment closely contributed to long-term therapeutic efficacy. We have been developed three platforms by targeting macrophages. First,manipulating macrophages to "turn off" neuropathologyical signals and to "turn on" therapeutic immune responses. This immunoswitch allows to develop the effective immunotherapies for the brain diseases. Second,we used a targeting peptide,rabies virus glycoprotein(RVG),to bind core-shell nanoparticles(RVG-NPs) for dual targeting the brain and macrophages. RVG-NPs enhanced trans-BBB and favored to macrophages and microglia. Using NPs to deliver plasmid DNA(pDNA) into macrophages,we obtained the higher transfection efficiency. The pDNA-NPs showed to effectively edit foreign gene in and delete target gene out. Moreover,macrophage is cellular immune regulator that target to the brain diseased site treatment of brain diseases. Thus,macrophages based immunotherapy is a promising strategy in neurological ailments.

Regulation of immune responses is particular important in recovery after stroke,traumatic brain injury,and brain infections by inhibiting inflammation and reinstating rescue *** delivery based immunotherapy appears to have significant therapeutic potential for the treatment of neurological conditions by inhibition of neuroinflammation to prevent irreversible destructive ***,current gene delivery systems lack desired convenience,efficiency,and *** offers an attractive gene delivery system that overcome the immunogenic and long-term oncologic effects of the viral *** branched PEI lipids with PLGA,we developed a highly efficient core-shell nanoparticle(NP)gene delivery *** IL-4 plasmid DNA(pIL-4)loaded NPs(pIL-4-NPs)were effectively delivered pIL-4 to the human blood monocytes derived macrophages(MDM)and mouse bone marrow-derived macrophages(BMM).Furthermore,pIL-4-NPs quickly escaped lysosomes and entered the nuclei of these *** in vitro transfection assays,the pIL-4-NPs had higher transfection efficiency,in contrast,PEI25KPEI liposomes and PLGA NPs were unable to transfect MDM and *** LPSinduced inflammatory in vitro and in vivo models,transfection of pIL-4-NPs decreased LPSinduced pro-inflammatory cytokines,such as IL-1,IL6,and TNF-.Our findings indicate that pIL-4-NPs delivery system is capable of effectively deliver IL-4 plasmid DNA into human and murine macrophages,which consistently express IL-4 in vitro and in *** pIL-4-NPs dramatically diminished pro-inflammatory cytokines expression,exhibited antineuroinflammatory and neuroprotective efficacy when macrophages were activated by ***,branched PEI lipids and PLGA formed core-shell NPs are a promising gene delivery platform for immunotherapy in neurological ailments.

T cell activation is a main component of adaptive immunity,which involves two types of signals transmitted by key immune receptors upon engagement with their respective ligands present on the antigen-presenting cells(APCs)[1].The first signal is induced via the T cell receptor(TCR)upon binding to antigenic peptide–major histocompatibility complex(pMHC);the second signal is induced via the costimulatory receptors,the prototype of which is CD28,which can bind to either B7-1(also known as CD80)or B7-2(also known as CD86)[2,3].

Stimulator of interferon genes(STING)-mediated innate immune activation plays a key role in tumor-and self-DNA-elicited antitumor immunity and ***,STING can also suppress tumor immunity and *** signaling In host nonhematopoietic cells was reported to either protect against or promote graft-versus-host disease(GVHD),a major complication of allogeneic hematopoietic cell transplantation(allo-HCT).Host hematopoietic antigen-presenting cells(APCs)play key roles in donor T-cell priming during GVHD ***,how STING regulates host hematopoietic APCs after allo-HCT remains *** utilized murine models of allo-HCT to assess the role of STING in hematopoietic ***-deficient recipients developed more severe GVHD after major histocompatibility complex-mismatched *** bone marrow chimeras,we found that STING deficiency in host hematopoietic cells was primarily responsible for exacerbating the ***,STING on host CD11c+cells played a dominant role in suppressing allogeneic T-cell ***,STING deficiency resulted in increased survival,activation,and function of APCs,including macrophages and dendritic ***,constitutive activation of STING attenuated the survival,activation,and function of APCs isolated from STING V154M knock-in ***-deficient APCs augmented donor T-cell expansion,chemokine receptor expression,and migration into intestinal tissues,resulting in accelerated/exacerbated *** pharmacologic approaches,we demonstrated that systemic administration of a STING agonist(bis-(3'-5')-cyclic dimeric guanosine monophosphate)to recipient mice before transplantation significantly reduced GVHD *** conclusion,we revealed a novel role of STING in APC activity that dictates T-cell allogeneic responses and validated STING as a potential therapeutic target for controlling GVHD after allo-HCT.
