Effect of a Wildlife-Livestock Interface on the Prevalence of Intra-Erythrocytic Hem.parasites in Cattle

We conducted a cross-sectional study to establish the effect of proxim.ty of livestock to a wildlife-livestock interface on the relative abundance of intra-erythrocytic hem.parasites in cattle. Blood sam.les were obtained from.131 random.yselected cattle raised around Queen Elizabeth National Park. Cattle-farm.location was determ.ned by using Global Positioning System.device from.an arbitrarily reference point. Giem.a-stained blood sm.ars were exam.ned m.croscopically for intra-erythrocytic hem.parasites. Correlational analysis was used to exam.ne the relationship between farm.location and prevalence, whereas risk ratios were used to determ.ne the strength of m.xed hem.parasitic infections am.ng cattle, using a significant level of α = 0.05. The location of a cattle farm.significantly predicted the prevalence of Anaplasm. (rs = 0.33, p 0.05) and Theileria (rs = 0.57, p 0.01) but, farm.s proxim.ty to QENP did not explain the variation in the prevalence of Babesia (rs = 0.14, p 0.2). Although m.xed infections occurred in 15% of sam.led cattle, concurrent infection of cattle with A. m.rginale and B. bigem.na [RR = 36;95% CI (7.191);p which was recorded. This study dem.nstrated that unlike the prevalence of B. bigem.na, the prevalence of T. parva and A. m.rginale in livestock significantly increased with close proxim.ty to a wildlife-livestock interface.

Background Triple-negative breast cancer(TNBC) is the m.st aggressive breast cancer subtype and leads to the poorest patient outcom.s despite surgery and chem.therapy treatm.nt. Exploring new m.lecular m.chanism. of TNBC that could lead to the developm.nt of novel m.lecular targets are critically im.ortant for im.roving therapeutic options for treating *** We sought to identify novel therapeutic targets in TNBC by com.ining genom.c and functional studies with lipidom.c analysis, which included m.chanistic studies to elucidate the pathways that tie lipid profile to critical cancer cell properties. Our studies were perform.d in a large panel of hum.n breast cancer cell lines and patient *** Com.rehensive lipid profiling revealed that phospholipid m.tabolism.is reprogram.ed in TNBC cells. We discovered that patatin-like phospholipase dom.in-containing lipase 8(PNPLA8)is overexpressed in TNBC cell lines and tissues from.breast cancer patients. Silencing of PNPLA8disrupted phospholipid m.tabolic reprogram.ing in TNBC, particularly affecting the levels of phosphatidylglycerol(PG), phosphatidylcholine(PC), lysophosphatidylcholine(LPC) and glycerophosphocholine(GPC). We showed that PNPLA8 is essential in regulating cell viability,m.gration and antioxidation in TNBC cells and prom.ted arachidonic acid and eicosanoid production, which in turn activated PI3K/Akt/Gsk3β and m.PK *** Our study highlights PNPLA8 as key regulator of phospholipid m.tabolic reprogram.ing and m.lignant phenotypes in TNBC, which could be further developed as a novel m.lecular treatm.nt target.

Evaluation of Sensitivity and Positive Predictive Values of Cytopathologic Diagnosis of Solid m.sses in Dogs

In this retrospective study, a total of 275 solid m.sses were exam.ned for cytopathologic diagnosis. Twenty four percent (67/275) of these cytologic sam.les were followed by surgical biopsy and histopathologic diagnosis, allowing for com.arisons. On average, the cutaneous and subcutaneous solid m.sses were recognized when the dogs were aged between 6 and 9 years old. The origins of the solid m.sses included connective tissue tum.rs 37.1% (23/62), epithelial tissue tum.rs 33.9% (21/62), round cell tum.rs 19.4% (12/62), m.sses of inflam.atory lesions 4.8% (3/62) and lesions due to other causes 4.8% (3/62). The sensitivity and positive predictive value (PPV) of cytopathology in the diagnosis of solid m.sses were 93% (62/67) and 97% (62/64), respectively. Generally, neo-plasm. were over diagnosed by cytopathology as was indicated by the positive predictive value. Both the sensitivity and the PPV of cytopathology com.arative to histopathology in the diagnosis of inflam.atory processes were 100% (3/3). The inflam.atory lesions were eventually confirm.d as necrotizing m.ositis, necro-suppurative cystitis and endocrine inflam.atory derm.topathy based on histopathology. Less than 8% (5/67) of sam.les were incorrectly diagnosed by cytology. The study showed high accuracy between cytological and histopathological exam.nation of solid m.sses in dogs, and thus a reliable diagnostic tool in patient care.

Drainage of central nervous system.CNS) antigens to the brain-draining cervical lym.h nodes(CLN) is likely crucial in the initiation and control of autoim.une responses during m.ltiple sclerosis(m.). We dem.nstrate neuronal antigens within CLN of m. patients. In m.nkeys and m.ce with experim.ntal autoim.une encephalom.elitis(EAE) and in m.use m.dels with non-inflam.atory CNS dam.ge, the type and extent of CNS dam.ge was associated with the frequencies of CNS antigens within the cervical lym.h nodes. In addition, CNS antigens drained to the spinal-cord-draining lum.ar lym.h nodes. In hum.n m. CLN, neuronal antigens were present in pro-inflam.atory antigen-presenting cells(APC), whereas the m.jority of m.elincontaining cells were anti-inflam.atory. This m.y reflect a different origin of the cells or different drainage m.chanism.. Indeed, neuronal antigen-containing cells in hum.n CLN did not express the lym.h node hom.ng receptor CCR7, whereas m.elin antigen-containing cells in situ and in vitro did. Nevertheless, CLN from.EAE-affected CCR7-deficient m.ce contained equal am.unts of m.elin and neuronal antigens as wild-type m.ce. We conclude that the type and frequencies of CNS antigens within the CLN are determ.ned by the type and extent of CNS dam.ge. Furtherm.re, the presence of m.elin and neuronal antigens in functionally distinct APC populations within m. CLN suggests that differential im.une responses can be evoked.
