Probiotics and postbiotics in colorectal cancer:Prevention and complementary therapy

Colorectal cancer(CRC)is a leading cause of human mortality *** conventional anticancer therapy not always being effective,there is growing interest in innovative“drug-free”cancer treatments or interventions that improve the efficacy of established *** is associated with microbiome alterations,a process known as dysbiosis that involves depletion and/or enrichment of particular gut bacterial species and their metabolic *** patient treatment with traditional probiotics(with or without prebiotics),nextgeneration probiotics(NGP),or postbiotics represents a potentially effective and accessible complementary anticancer strategy by restoring gut microbiota composition and/or by signaling to the *** this capacity,restoration of the gut microbiota in cancer patients can stabilize and enhance intestinal barrier function,as well as promote anticarcinogenic,anti-inflammatory,antimutagenic or other biologically important biochemical pathways that show high specificity towards tumor *** benefits of traditional probiotics,NGP,and postbiotics include modulating gut microbiota composition and function,as well as the host inflammatory *** application in CRC prevention is highlighted in this review,where we consider supportive in vitro,animal,and clinical *** on emerging research,NGP and postbiotics hold promise in establishing innovative treatments for CRC by conferring physiological functions via the production of dominant natural products and metabolites that provide new host-microbiota signals to combat *** favorable results have been reported,further investigations focusing on strain and dose specificity are required to ensure the efficacy and safety of traditional probiotics,NGP,and postbiotics in CRC prevention and treatment.

Survival predictors in patients treated with a molecular adsorbent recirculating system

AIM:To identify prognostic factors for survival in patients with liver failure treated with a molecular adsorbent recirculating system(MARS). METHODS:MARS is a liver-assisting device that has been used in the treatment of liver failure to enable native liver recovery,and as a bridge to liver transplantation(LTX).We analyzed the 1-year outcomes of 188 patients treated with MARS,from 2001 to 2007, in an intensive care unit specializing in liver disease. Demographic,clinical and laboratory parameters were recorded before and after each ***-year survival and the number of LTXs were *** regression analysis was performed to determine factors predicting survival. RESULTS:The study included 113 patients with acute liver failure(ALF),62 with acute-on-chronic liver failure(AOCLF),11 with graft failure(GF),and six with miscellaneous liver *** was performed for 29% of patients with ALF,18% with AOCLF and 55%with *** overall 1-year survival rate was 74% for ALF,27% for AOCLF,and 73% for *** poorest survival rate,6%,was noted in non-transplanted patients with alcohol-related AOCLF and cirrhosis,whereas,patients with enlarged and steatotic liver had 55% *** etiology of liver failure was the most important predictor of survival(Pv levels(P =0.022)in ALF of unknown etiology. CONCLUSION:The etiology of liver disease was the most important prognostic *** treatment appears to be ineffective in AOCLF with end-stage cirrhosis without an LTX option.

Nanoparticles(NPs)can be transported via the nose-to-brain(N_(2)B)***,quantitative data on their spatiotemporal dynamics and regulation of the N_(2)B transport are largely *** surveyed metal oxide/hydroxide NPs as magnetic resonance imaging(MRI)contrasts for quantitative N_(2)B *** containing divalent transition metals were the only ones capable of N_(2)B *** T1-weighted(T1W)MRI,we showed that Mn_(3)O_(4)-NPs were readily engulfed by olfactory receptor neurons(ORNs)without disrupting olfactory sensing,we mapped their N_(2)B *** neurons,the Mn_(3)O_(4)-NPs were localized to the cytosol,mitochondria,vesicles,moved at mixed fast and slow axonal transport velocities intraand extra-vesicularly through *** NPs’axonal transport is dependent on neuronal activity and microtubule *** Mn_(3)O_(4)-NPs were trans-synaptically transmitted through at least four synapses across the olfactory ***-synaptic transmission of the NPs was dependent on N-type Ca^(2+)channels and NMDA receptors but blocked by GABAB receptor activation.A five-parameter Weibull signal increase/decrease model fitted to the T1W MRI data allowed for estimating kinetic parameters of Mn_(3)O_(4)-NP accumulation/*** and relative accumulation rates,but not elimination,correlated negatively with the number of synapses from ORNs,indicating a coupling of the NPs’N_(2)B transport with spontaneous neuronal ***,olfactory stimuli(2,5-dimethylpyrazine and acetophenone)significantly modulated and rerouted the Mn_(3)O_(4)-NP N_(2)B transport odor ***,the NPs’trans-synaptic transmission was impaired by aging and the onset of Parkinson’s *** data suggest new approaches to diagnostics,functional neuroimaging,controlling N_(2)B drug delivery.

Increasing evidence points to a role for killer immunoglobulin-like receptors(KIRs)in the development of autoimmune *** particular,a positive association of KIR3DS1(activating receptor)and a negative association of KIR3DL1(inhibitory receptor)alleles with ankylosing spondylitis(AS)have been reported by several ***,none of the studies analyzed these associations in the context of functionality of polymorphic *** better understand how the KIR3DL1/3DS1 genes determine susceptibility to AS,we analyzed the frequencies of alleles and genotypes encoding functional(KIR3DL1*F)and non-functional(KIR3DL1*004)*** genotyped 83 AS patients and 107 human leukocyte antigen(HLA)-B27-positive healthy controls from the Russian Caucasian population using a two-stage sequence-specific primer PCR,which distinguishes KIR3DS1,KIR3DL1*F and KIR3DL1*004 *** the patients carrying two functional KIR3DL1 alleles,those alleles were additionally genotyped to identify KIR3DL1*005 and KIR3DL1*007 alleles,which are functional but are expressed at low ***3DL1 was negatively associated with AS at the expense of KIR3DL1*F but not of KIR3DL1**** finding indicates that the inhibitory KIR3DL1 receptor protects against the development of AS and is not simply a passive counterpart of the segregating KIR3DS1 allele encoding the activating ***,analysis of genotype frequencies indicates that the presence of KIR3DS1 is a more important factor for AS susceptibility than the absence of KIR3DL1**** activation of either natural killer(NK)or T cells via the KIR3DS1 receptor can be one of the critical events in AS development,while the presence of the functional KIR3DL1 receptor has a protective ***,even individuals with a genotype that carried two inhibitory KIR3DL1 alleles expressed at high levels could develop AS.

Morphofunctional status and the role of mononuclear phagocyte system lung compartment in the pathogenesis of influenza A (H5N1) in mammals

Influenza and other respiratory viral infections account for 80%-90% of infectious pathologies. Influenza A (H5N1) virus has an apparent pneumotropism, and therefore the lung compartment of mononuclear phagocyte system plays an important role in antiviral immunity. Lung macrophages are active phagocytes expressing variety of antiviral factors. The investigation of morphofunctional status of lung macrophages and evaluation of their role in mammal antiviral response in a mouse model were performed within the study. Methods: Light microscopy using standard hematoxylin-eosin, and van-Gizon’s picrofuchsin staining. Immunohistochemistry using influenza A antigen marker specific primary antibodies, myeloperoxidase, cathepsin D, lysozyme, NO synthase, pro-inflammatory cytokines, cells of CD68 macrophage lineage, PCNA proliferative activity. Morphometric and statistical analysis. Results: Influenza A virus antigen was detected in lung macrophages starting from day 1 to day 14 of infection which corresponds with the beginning of convalescence and may be suggestive of prolonged persistence of virus. On the one hand, the cytopathic effects of the virus lead to lung macrophages death mainly via apoptosis through activation of caspase cascade, including caspase-3 and caspase-9. On the other hand, the observed activation of PCNA proliferation marker, perhaps, allows to support the pool of lung macrophages not only by their recruitment from bone marrow but also by their proliferation in situ. The increase of mononuclear phagocyte system cells expressing antiviral factors depended on the stage of infection. In the early stage, there was an increase of number of cells expressing lysozyme, myeloperoxidase, cathepsin D, endothelial NO synthase (eNOS) followed by the increase of number of macrophages expressing inducible NO synthase (iNOS), pro-inflammatory cytokines and interleukins.

The Baraba and Kulunda steppes are located in southwestern Siberia in an area with an arid continental climate. This paper presents results of the first study of the hypersaline Lake Krasnovishnevoye(Baraba steppe, TDS(total dissolved solids)=297 g/L, pH 7.88). The major chemical, mineralogical and biological features of the lake were studied and compared to those of Lake Malinovoe, a typical saline neutral lake of Kulunda steppe(TDS=396 g/L, pH 7.63). The phytoplankton composition and the culturable diversity of anoxygenic phototrophic bacteria from Lake Krasnovishnevoye correspond to the ones in the Kulunda lakes. Nevertheless, the peculiarities of water composition and regime of Lake Krasnovishnevoye reduce the biodiversity to prokaryotes and unicellular algae.

Social Deprivation of Newborn Rats Modifies Exploratory and Defensive Behavior at Mature Age

The effect of complete social deprivation in early postnatal ontogenesis on adult behavior in Wag/Rij rats with a genetic predisposition to the development of absence epilepsy was studied. From the 2nd to the 8th day of life, a 3-hour daily isolation of the pups from the mother and siblings was performed, the control animals were handled. In adulthood, offspring behavior was tested in an elevated plus-maze, an open field, and for an ability to learn the conditioned reflex of active avoidance. In isolated earlier rats, an increase in emotional reactivity, exploratory behavior in a potentially dangerous space, the speed of learning a conditioned reflex with negative painful reinforcement, and a disturbance of the formation of a defensive reaction were revealed. It is assumed that high emotional reactivity creates the prerequisites for the development of absence epilepsy.
