The administration of extracellular vesicles(EV)from mesenchymal stromal cells(MSC)is a promising cell-free nanotherapy for tissue repair after myocardial infarction(MI).However,the optimal EV delivery strategy remains ***,we designed a novel MSC-EV delivery,using 3D scaffolds engineered from decellularised cardiac tissue as a cell-free product for cardiac *** from porcine cardiac adipose tissue-derived MSC(cATMSC)were purified by size exclusion chromatography(SEC),functionally analysed and loaded to ***-EV markedly reduced polyclonal proliferation and pro-inflammatory cytokines production(IFnγ,TnFα,IL12p40)of allogeneic ***,cATMSC-EV recruited outgrowth endothelial cells(OEC)and allogeneic MSC,and promoted *** labelled cATMSC-EV were mixed with peptide hydrogel,and were successfully retained in decellularised ***,cATMSC-EV-embedded pericardial scaffolds were administered in vivo over the ischemic myocardium in a pig model of *** days from implantation,the engineered scaffold efficiently integrated into the post-infarcted ***-EV were detected within the construct and MI core,and promoted an increase in vascular density and reduction in macrophage and T cell infiltration within the damaged *** confined administration of multifunctional MSC-EV within an engineered pericardial scaffold ensures local EV dosage and release,and generates a vascularised bioactive niche for cell recruitment,engraftment and modulation of short-term post-ischemic inflammation.

Camivores play a central role in ecosystem processes by exerting top-down control, while fire exerts bottom-up con- trol in ecosystems throughout the world, yet, little is known about how fire affects short-term carnivore distributions across the landscape. Through the use of a long-term data set we investigated the distribution of lions, during the daytime, in relation to burned areas in Serengeti national Park, Tanzania. We found that lions avoid burned areas despite the fact that herbivores, their prey, are attracted to burned areas. Prey attraction, however, likely results from the reduction in cover caused by burning, that may thereby decrease lion hunting success. Lions also do not preferentially utilize the edges of burned areas over unburned areas de- spite the possibility that edges would combine the benefit of cover with proximity to abundant prey. Despite the fact that lions avoid burned areas, lion territory size and reproductive success were not affected by the proportion of the territory burned each year. Therefore, burning does not seem to reduce lion fitness perhaps because of the heterogeneity of burned areas across the landscape or because it is possible that when hunting at night lions visit burned areas despite their daytime avoidance of these ar- eas .

Surface recombination represents a handicap for high-efficiency solar cells. This is especially important for nanowire array solar cells, where the surface-to-volume ratio is greatly enhanced. Here, the effect of different passivation materials on the effective recombination and on the device performance is experimentally analyzed. Our solar cells are large area top-down axial n-p junction silicon nanowires fabricated by means of near-Field Phase-Shift Lithography （nF-PSL）. We report an efficiency of 9.9% for the best cell, passivated with a SiO2/Sinx stack. The impact of the presence of a surface fixed charge density at the silicon/oxide interface is studied.

normothermic ex vivo lung perfusion(nEVLP)has emerged as a modernized organ preservation technique that allows for detailed assessment of donor lung function prior to *** main goal of this study was to identify potential biomarkers of lung function and/or injury during a prolonged(19 h)nEVLP procedure using in vivo solid-phase microextraction(SPME)technology followed by liquid chromatography-high resolution mass spectrometry(LC-HRMS).The use of minimally invasive in vivo SPME fibers for repeated sampling of biological tissue permits the monitoring and evaluation of biochemical changes and alterations in the metabolomic profile of the *** in vivo SPME fibers were directly introduced into the lung and were also used to extract metabolites(on-site SPME)from fresh perfusate samples collected alongside lung samplings.A subsequent goal of the study was to assess the feasibility of SPME as an in vivo method in metabolomics studies,in comparison to the traditional inlab metabolomics *** upregulated biochemical pathways involved in pro-and antiinflammatory responses,as well as lipid metabolism,were observed during extended lung perfusion,especially between the 11th and 12th hours of the procedure,in both lung and perfusate ***,several unstable and/or short-lived metabolites,such as neuroprostanes,have been extracted from lung tissue in vivo using SPME ***-site monitoring of the metabolomic profiles of both lung tissues through in vivo SPME and perfusate samples on site throughout the prolonged nEVLP procedure can be effectively performed using in vivo SPME technology.

Many studies have demonstrated the linkage between the IBD3 region (6p21.1-23), an area which encompasses the famous human leukocyte antigen (HLA) complex, and Crohn’s disease (CD) or ulcerative colitis (UC). IBD3 is the only region that meets genome-wide significance, and provides stronger evidence of the linkage than 16p13.1-16q12.2 (IBD1), the locus that contains the susceptibility gene CARD15. However, despite these findings, IBD3 susceptibility genes remain elusive and unclear due to the strong linkage disequilibrium, extensive polymorphism, and high gene density that characterize this area and also due to varying allele frequencies in populations around the world. This area presents an extremely high abundance of genes, including the classical and non-classical major histocompatibility complex (MHC) class I and II genes, and other genes, namely MHC class III genes tumor necrosis factor (TnF)-α and -β, and Hsp, whose proteins play key functions in immunological processes. To date, it is not clear which genes within the MHC family contribute to the IBD pathogenesis, although certain HLA alleles have been associated with IBD. Recent insights into the biological function of other genes encoded within the IBD3 region, such as the MHC class I chain-related (MIC) genes, have led investigators to a more comprehensive exploration of this region. MHC class I chain-related molecule A (MICA) is highly polymorphic and interacts with nKG2D, its receptor on the surface of nK, Tγδ and T CD8+ cells. Increased expression of MICA in intestinal epithelial cells and increased expression of nKG2D in CD4+ T cells (lamina propria) in patients with CD have also been reported. A melt maintained for hours in a press pour unit allowed the following changes over time from spheroidal graphite to compacted graphite iron by casting thermal cups at regular time *** provided extensive experimental information for checking the possibility of simulating solidification of compacted graphite irons by means of a microstructure modelling *** solidification,compacted graphite develops very much as lamellar graphite but with much less *** this basis,a simulation of the thermal analysis records was developed which considers solidification proceeding in a pseudo binary Fe-C *** simulated curves were compared with the experimental ones obtained from three representative alloys that cover the whole microstructure change during the holding of the *** most relevant result is that the parameter describing branching capability of graphite is the most important for reproducing the minimum eutectic temperature and the recalescence which are so characteristic of the solidification of compacted graphite cast irons.

Background: To study novel treatment modalities for pancreatic ductal adenocarcinoma(PDAC), we need to transfer the knowledge from in vitro to in vivo. It is important to mirror the clinical characteristics of the typically local invasive growth of pancreatic cancer and the distant spread resulting in liver metastasis. notably, for xenotransplant studies using human specimen, two models, i.e. subcutaneous(s.c.) and orthotopic(o.t.) transplantation are widely used. Methods: The subcutaneously and orthotopically inoculated Colo357 Bcl-x L cell-derived tumors were directly compared with and without TnF-related apoptosis inducing ligand(TRAIL) treatment. The size of primary tumors, number of liver metastasis and the histologic markers Ki67, M30, TnF-α and CD31 were assessed. Results: Upon TRAIL treatment, the primary tumors did not change their size, neither in the s.c. nor in the o.t. approaches. But when s.c. was compared to o.t., the size of the s.c. tumors was more than twofold bigger than that of the o.t. tumors( Pnoculated PDAC cells developed liver metastasis upon TRAIL treatment much more frequently( n=13/17) than mice with subcutaneously inoculated PDAC cells( n=1/11)( Png force for this increased metastasis, a higher TnF-α staining intensity in the o.t. tumors was observed by immunohistochemistry. Conclusions: These data from a direct side-by-side comparison underline the importance of the proper inoculation site of the PDAC cells. Local invasion and liver metastases are a hallmark of PDAC in the clinic;the o.t. model is clearly superior in reflecting this setting. Moreover, a serious side-effect of a possible new therapeutic compound became obvious only in the o.t. model.

As ecosystem engineers,earthworms play a key role in the soil ***,due to increasing anthropogenic pressure,soil organisms,including earthworms,are being threatened by habitat *** this study,we undertook a qualitative and quantitative investigation of earthworms of the family Lumbricidae in four types of Carpathian beech woodland(Fagetum carpaticum),characterized by their understory vegetation(I,*** drymejae;Ⅱ,***;Ⅲ,***;and IV,***),in the Bieszczady national Park(Eastern Carpathians,Southeast Poland).At each investigated site,soil monoliths(25 cm×25 cm×25 cm)were examined by hand *** were expelled from deep soil layers using a weak formalin solution(0.4%).Depending on the phytocoenosis,7 species of Lumbricidae were identified at each of the sites I,Ⅱ,andⅢand 10 at site *** IV(***)differed significantly(Pnd the Shannon-Wiener diversity index(0.52±0.12).Although present three decades ago,the deep-burrowing species Octodrilus transpadanus and the litter-dwelling species Dendrobaena octaedra were not found at site IV in the present *** suspect that these two species may have been displaced by the invasive Lumbricus terrestris,which was not found at the same site in the *** observations warrant further investigation to verify the predicted effects of an invasive earthworm,including the potential effects on soils and other fauna and flora,which have been documented in numerous countries.

Deterioration in overall health,hormonal disturbances,and erectile dysfunction(ED)contributes to limitations in sexual activity in the elderly,which is further limited by incorrect beliefs about the hazards of sexual activity in cardiac *** aimed to analyze the occurrence of ED in elderly men,their perception of the relevance of good sexual function,and their expectations of physicians.A cross-sectional study encompassed 731 patients with coronary artery disease(CAD)subjected to cardiac *** data and data on modifiable risk factors and patient expectations were *** was assessed using the IIEF-5 *** among the risk factors for ED,occurrence of ED,and patient expectations,as well as the changes in the indicators between 2012 and 2016,were *** mean age of men was 70.7±5.1 *** prevalence of ED was 93.0%.The IIEF-5 score was significantly associated with age,tobacco smoking,exercise tolerance,time to diagnosis of CAD,and treatment with calcium channel blockers and *** declared that sexual activity was overall important(47.9%)or very important(25.6%).Three hundred and sixty(49.3%)patients expected their physician to show interest in their sexual health,but the topic was addressed in only 12.5%.Over the past few years,we have observed an increase in the awareness and importance of sexual health as well as a significant increase in patients’expectations of physicians to show interest in their sexual ***’expectations of discussing and receiving treatment for ED remain an unmet medical need.
