Within the last several decades,the scientific community has made substantial progress in elucidating the complex pathophysiology underlying spinal cord ***,despite the many advances using conventional mammalian models,both cellular and axonal regeneration following spinal cord injury have remained out of *** this sense,turning to non-mammalian,regenerative species presents a unique opportunity to identify pro-regenerative cues and chara cterize a spinal cord microenvironment permissive to *** the signaling pathways hypothesized to be dysregulated during spinal cord injury is the purinergic signaling *** addition to its well-known role as energy currency in cells,ATP and its metabolites are small molecule neurotransmitte rs that mediate many diverse cellular processes within the central nervous *** our unde rstanding of the roles of the purinergic system following spinal cord injury is limited,this signaling pathway has been implicated in all injury-induced secondary processes,including cellular death,inflammation,reactive gliosis,and neural *** that the purinergic system is also evolutionarily conserved between mammalian and non-mammalian species,comparisons of these roles may provide important insights into conditions responsible for recovery ***,we compare the secondary processes between key model species and the influence of purinergic signaling in each *** our understanding of this signaling system and pro-regenerative conditions continues to evolve,so does the potential for the development of novel therapeutic interventions for spinal cord injury.

Obesity is one of the diseases with severe health consequences and rapidly increasing worldwide *** the complex network of food intake and energy balance regulation is an essential prerequisite for pharmacological intervention with obesity.G protein-coupled receptors(GPCRs)are among the main modulators of metabolism and energy ***,for instance,regulate appetite and satiety in certain hypothalamic neurons,as well as glucose and lipid metabolism and hormone secretion from *** in some GPCRs,such as the melanocortin receptor type 4(MC4R),have been associated with early-onset ***,we identified the adhesion GPCR latrophilin 1(ADGRL1/LPHN1)as a member of the regulating network governing food intake and the maintenance of energy *** of the highly conserved receptor in mice results in increased food consumption and severe obesity,accompanied by dysregulation of glucose ***,we identified a partially inactivating mutation in human ADGRL1/LPHN1 in a patient suffering from ***,we propose that LPHN1 dysfunction is a risk factor for obesity development.

Local soil conditions can significantly modify the seismic motion expected on the soil *** most cases,the indications concerning the influence of the underlying soil provided by the in-force european and Italian Building Codes underestimate the real seismic amplification *** this reason,numerical analyses of the local seismic response(LSR)have been encouraged to estimate the soil filtering *** analyses are generally performed in free-field conditions,ignoring the presence of superstructures and,therefore,the effects of dynamic soil-structure interaction(DSSI).Moreover,many studies on DSSI are characterised by a sophisticated modelling of the structure and an approximate modelling of the soil(using springs and dashpots at the foundation level);while others are characterised by a sophisticated modelling of the soil and an approximate modelling of the structure(considered as a simple linear elastic structure or a single degree of freedom system).This paper presents a set of finite element method(FeM)analyses on a fully-coupled soil-structure system for a reinforced concrete building located in Fleri(Catania,Italy).The building,designed for gravity loads only,was severely damaged during the 26 December 2018 *** soil was modelled considering an equivalent visco-elastic behaviour,while the structure was modelled assuming both the visco-elastic and visco-inelastic *** comparison made between the results of the FeM analyses and the observed damage is valuable.

Dear editor,Gut microorganisms process food in animal guts and release many metabolic by-products,which are predicted to influence host physiological processes such as energy and lipid ***,we investigate how succi-nate,a TCA cycle intermediate that is a major predicted release product of gut bacteria in Drosophila,influences the nutritional physiology of its Drosophila *** ad-ministered succinate as a dietary supplement to microbe-free Drosophila,and quantified key nutritional *** succinate significantly reduced fly lipid levels by up to~50%.This response was not replicated in paral-lel experiments conducted with dietary fumarate supple-ment,indicating that it could not be attributed to a gen-eral effect of TCA *** hypothesize that microbe-derived succinate may contribute to the reduced lipid content of Drosophila bearing gut bacteria,relative to axenic *** generally,this study high-lights the importance of microbial-derived metabolites as regulators of host metabolism.

Understanding mosquito interaction with long-lasting insecticidal bednets is crucial in the development of more effective intervention methods to protect humans from malaria *** such,a 240×240×1000 mm laboratory setup for the in-line recording of digital holograms and subsequent in-focus reconstruction and 3D localisation of mosquitoes is *** bednet background removal methods are used to accurately localise a mosquito obscured by a bednet in 3D *** and physical data demonstrate that this method is suitable for mosquitoes positioned 3−1000 mm behind a bednet.A novel post-processing technique,involving a cascade-correlation of a Tamura of Intensity focus metric extracted from digitally reconstructed scenes,accurately localises mosquitoes positioned 35−100 mm behind a bednet from a single digital *** result of this study is a scalable digital holographic methodology to examine mosquito-bednet interaction in 3D at a level of accuracy previously only seen in 2D imaging of mosquitoes in a much smaller volume.

Importance:Reported coronavirus disease 2019(COVID-19)pandemic effects on pediatric trauma have been ***:We investigated the characteristics of pediatric trauma including alcohol use during the pandemic at our urban trauma ***:The trauma database of our adult level 1 trauma center was queried for all pediatric(age≤18 years)patients presenting between March 1,2020,and October 30,*** from 2017 to 2019 served as a *** analyzed included demographics,mechanisms,injury severity,hospitalization characteristics,and positive blood ***:Pandemic pediatric trauma volumes increased by 67.5%(330/year vs.197/year).Pandemic patients were younger(median age 13 vs.14 years,P=0.011),but similar in gender,ethnicity,severity,hospital length of stay,mortality,and rates of penetrating *** doubled(79/year vs.34/year)and shifted away from high falls>6 meters(0%vs.7.9%)to moderate falls 1–6 meters(58.2%vs.51.5%)(P=0.028).Transportation injury rates were similar however mechanisms shifted from motor vehicle crashes(−13.5%)towards recreational vehicles including motorcycles(+2.1%),all-terrain vehicles(+8.6%),and bicycles(+3.8%)(P=0.018).Pediatricpositive blood alcohol was significantly higher(11.2%vs.5.1%,Pespecially for ages 14–18 years(21.7%vs.9.5%,Perpretation:Pediatric trauma volumes during the COVID-19 pandemic *** patients had more recreational vehicle injuries and higher rates of positive blood *** suggests an increased need for alcohol assessment and targeted interventions in the pediatric population during pandemics or periods of school closures.
