Altered Endothelin-1 Signaling in Production of Thromboxane A_(2) in Kupffer Cells from Bile Duct Ligated Rats

Kupffer cells(KCs),the liver resident macrophages accounting for 80-90%of the total population of fixed tissue macrophages in the body,not only play a key role in host defense via removing particulate materials from the portal circulation,but may also contribute to the pathogenesis of various liver *** have previously demonstrated that KCs play an important role in controlling portal hypertension and hepatocellular injury via releasing thromboxane A_(2)(TXA_(2))in early fibrosis induced by one-week bile duct ligation(BDL).Production of TXA_(2) is controlled by cytosolic phospholipase A_(2)(cPLA_(2))that is activated by the interaction of entothelin-1(ET-1)with its G-protein coupled ET receptor B(ETBR).However,the signaling pathways that contribute to the ET-1-induced activation of cPLA_(2) and production of TXA_(2) in KCs in the normal healthy or injured livers are not yet clear,which are investigated in the present study using isolated KCs from one-week BDL or sham *** pharmacological inhibition of cPLA_(2) or chelation of intracellular calcium abrogated the ET-1 induction of TXA_(2) from *** to those from sham rats,KCs from BDL animals displayed a significantly enhanced responsiveness of p38 mAPK to ET-1,increased ETBR and Gai subunit but decreased Gaq and Ga11 *** of ERK1/2 or Gq signaling abrogated significantly the ET-1 induction of TXA_(2) in sham KCs but only slightly in BDL *** contrast,inhibition of p38 mAPK and Gi signaling markedly attenuated the ET-1 induction of TXA_(2) in BDL KCs but had no effect in sham ***,inhibition of PLC or PKC abrogated ET-1 induction of TXA_(2) in KCs from both sham and BDL *** hepatic stress(such as BDL)induces significant modifications in the receptor and intermediates of ET-1 signaling in KC and subsequently alters ET-1 signaling mechanisms,particularly a shift from Gq induced signaling to Gi induced signaling,in the activation of cPLA_(2) and production of TXA_(2) in

ENSO-induced Latitudinal Variation of the Subtropical Jet modulates Extreme Winter Precipitation over the Western Himalaya

In this study,we investigate the complex relationship between western disturbances(WDs),the El Ni?o–Southern Oscillation(ENSO),and extreme precipitation events(EPEs) in the western Himalaya(WH) during the extended winter season(November–march).WDs west of WH coincide with 97% of recorded EPEs,contributing substantially(32% in winter,11% annually) to total precipitation within *** are 6% less frequent and 4% more intense during El Ni?o than La Ni?a to the west of *** El Ni?o(compared to La Ni?a) years,WDs co-occurring with EPEs are significantly more intense and associated with 17% higher moisture transport over “WH box”(the selected region where most of the winter precipitation over WH occurs).This results in twice the EPE frequency during El Ni?o periods than La Ni?a periods.A substantial southward shift(~180 km) of the subtropical jet(STJ) axis during El Ni?o brings WD tracks further south towards their primary moisture sources,especially the Arabian *** have shown that WDs that are both more intense and pass to the south of their typical latitudes have higher levels of vertically integrated moisture flux(VImF)within *** convergence in the most intense pentile of WDs is 5.7 times higher than in the weakest,and is 3.4 times higher in the second lowest latitude pentile than in the ***,this study demonstrates a direct link between changes in the latitudinal position and intensity of WDs associated with the winter STJ,and moisture convergence,which leads to the occurrence of EPEs over WH during ENSO phases.

The effect of adsorbent textural and functional properties on model naphthenic acid adsorption

A variety of Ca2+signals,in the form of intercellular pulses and waves,have beenreported to be associated with the various sequential stages of somitogenesis:from convergent extension and the formation of the paraxial mesoderm;during thepatterning of the paraxial mesoderm to establish segmental units;throughout theformation of the morphological boundaries that delineate the segmental units,andfinally from within the maturing somites as they undergo subsequent develop-ment and *** to both the technical challenges presented in imag-ing intact,developing embryos,and the subtle nature of the Ca2+transientsgenerated,they have proved to be difficult to ***,a combinationof cultured cell preparations and improvements in explant and whole embryo im-aging techniques has begun to reveal a new and exciting class of developmentalCa2+*** this chapter,we review the small,but expanding,number ofreports in the literature and attempt to identify common characteristics of thesomitogenic Ca2+transients,such as their mode of generation,as well as theirspatial and temporal *** may help to elucidate the significance andfunction of these intriguing Ca2+transients and thus integrate them into the com-plex signaling networks that orchestrate early developmental events.

A Scalable Testing Framework for Location-Based Services

A novel testing framework for location based services is introduced. In particular, the paper showcases a novel architecture for such a framework. The implementation of the framework illustrates both the functionality and the feasibility of the framework proposed and the utility of the architecture. The new framework is evaluated through comparison to several other methodologies currently available for the testing of location-based applications. A case study is presented in which the testing framework was applied to a typical mobile service tracking system. It is concluded that the proposed testing framework achieves the best coverage of the entire location based service testing problem of the currently available methodologies; being equipped to test the widest array of application attributes and allowing for the automation of testing activities.

Traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease are three distinct neurological disorders that share common pathophysiological mechanisms involving neuroinflammation. One sequela of neuroinflammation includes the pathologic hyperphosphorylation of tau protein, an endogenous microtubule-associated protein that protects the integrity of neuronal cytoskeletons. Tau hyperphosphorylation results in protein misfolding and subsequent accumulation of tau tangles forming neurotoxic aggregates. These misfolded proteins are characteristic of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease and can lead to downstream neuroinflammatory processes, including assembly and activation of the inflammasome complex. Inflammasomes refer to a family of multimeric protein units that, upon activation, release a cascade of signaling molecules resulting in caspase-induced cell death and inflammation mediated by the release of interleukin-1β cytokine. One specific inflammasome, the NOD-like receptor protein 3, has been proposed to be a key regulator of tau phosphorylation where it has been shown that prolonged NOD-like receptor protein 3 activation acts as a causal factor in pathological tau accumulation and spreading. This review begins by describing the epidemiology and pathophysiology of traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease. Next, we highlight neuroinflammation as an overriding theme and discuss the role of the NOD-like receptor protein 3 inflammasome in the formation of tau deposits and how such tauopathic entities spread throughout the brain. We then propose a novel framework linking traumatic brain injury, chronic traumatic encephalopathy, and Alzheimer's disease as inflammasomedependent pathologies that exist along a temporal continuum. Finally, we discuss potential therapeutic targets that may intercept this pathway and ultimately minimize long-term neurological decline.
