Pharmacological drug screening to inhibit uveal melanoma metastatic cells either via EGF-R, MAPK, mtor or PI3K

AIM: To screen five potential pharmacological substances specifically targeting EGF-R, MAPK, mtor, or PI3K for their antiproliferative effects, possible impact on cell viability, as well as cell death rates on three different uveal melanoma metastasis cell lines in vitro. METHODS: Three different uveal melanoma metastasis cell lines(OMM2.5, OMM2.3, and OMM1), that originated from human hepatic and subcutaneous metastasis, were exposed to inhibitors of different targets: erlotinib(EGF-R), everolimus(mtor), selumetinib(MAPK), trametinib(MAPK) or the alkylphosphocholine erufosine(PI3K). Cell viability was assessed with a 2,3-bis-(2-methoxy-4-nitro-5-sulfophenyl)-2H-tetrazolium-5-carboxanilide(XTT) dye reduction assay after 24 h of treatment. Antiproliferative effects were evaluated separately after a 72-hour incubation of the cells with the pharmacological substance. Subsequently, the IC_(50) was calculated. Tumor cell death was investigated using a double stain apoptosis detection assay. RESULTS: Selumetinib, trametinib, and erufosine significantly decreased cell viability of all OMM cell lines(Pwed a significant inhibition of cell proliferation(Pwere ***: The preliminary drug screening demonstrates five new candidates, successfully targeting the canonical MAPK/ERK and PI3K/AKT/m tor pathways in uveal melanoma metastasis cells in vitro. Hence, these findings provide an experimental basis to explore future single or combined therapy strategies for metastatic uveal melanoma.

The surface water quality was investigated in the Córrego Sujo watershed, located in the municipality of Teresópolis, a mountainous region in the state of Rio de Janeiro, Brazil. The emphasis focused on inorganic parameters of water, since there are applications of pesticides in agricultural cultivations. The natural and anthropogenic contributions of the study area were monitored monthly in eight sites in the river channels of the drainage basin. The results were analyzed using an ICP-OES spectrophotometer and subsequently compared to Conama Resolution 357/05. In order to support the interpretation of the results of water quality, flow and precipitation data were measured in the watershed, along with the land use and vegetation cover map produced from a GeoEye satellite image. The study revealed that the surface waters were in high concentrations and above those recommended by Conama Resolution. It was noted also that the lithological and pedological variables influenced the results of water quality. Regarding seasonality, it has not been possible to establish a common standard to all elements in the rainy season or dry season, however, in the analysis of each parameter separately, there was evidence of a concentration distribution in temporal variation. Therefore, the results of this study indicate that it is also necessary to highlight these issues in the territor.al planning of the municipality and in the management of the Committee of Rio Piabanha watershed, to avoid the increasing of uncontrolled expansion of irrigated crops and intensive use of agrochemical inputs.

Twin-wire Networks for Zero Interconnect,High-Density 4-wire Electrical Characterizations of Materials

Four-wire measurements have been introduced by Lord Kelvin in 1861 and have since become the standard technique for characterizing small resistances and ***,high-density 4-wire measurements are generally complex,time-consuming,and inefficient because of constraints on interconnects,pads,external wires,and mechanical contacts,thus reducing reproducibility,statistical significance,and throu ***,we introduce,systematically design,analyze,and experimentally validate zero interconnect networks interfaced to external instrumentation by couples of twin wire.3D-printed holders with magnets,interconnects,nonadhesive layers,and spacers can effortlessly establish excellent electrical connections with tunable or minimum contact forces and enable accurate measurements even for delicate devices,such as thin metals on soft *** an example,we measured all the resistances of a twin-wire 29-resistor network made of silver-nanoparticle ink printed on polyimide,paper,or photo paper,including during sintering or temperature calibration,resulting in anunprecedentedly easy and accurate characterization of both resistivity and its temperature *** theoretical framework and experimental strategies reported here represent a breakthrough toward zero interconnect,simple,and efficient high-density 4-wire characterizations,can be generalized to other 4-wire measurements(impedances,sensors)and can open the way to more statistically meaningful and reproducible analyses of materials,high-throughput measurements,and minimally invasive characterizations of biomaterials.

This paper summarizes hydraulic studies carried out as part of the feasibility study for a new bridge over the Parana River in Argentina. The bridge will physically connect the provinces of Santa Fe and Corrientes, currently separated over the length a 200-km river reach. In addition to the hydraulic studies summarized here, the feasibility study includes analyses of the geology and geomorphology of the sites, land surveying, geophysical and hydrologic studies, studies of the navigation channel and fluvial transportation, transportation studies, and environmental impact studies. The main components of the present study were a geomorphologic analysis of the middle reach of the Parana River, and a quantitative hydraulic study. Qualitative and quantitative analyses provided a description of the hydraulic system, and served as a basis for a comparison of alternative bridge locations and the analysis of their vulnerability. The quantitative hydraulic study included the construction, calibration, and operation of one-dimensional (1D) and depth-averaged two-dimensional (2D) flow models. In order to tie the models to the most reliable data the 1D models covered a 450-km long reach, and included the main channel (about 2 km wide) and the flood plain (about 30 km wide). The results of the 1D models played a major role in the selection of the bridge location, and were subsequently used as boundary conditions for 2D modeling. The 2D models were constructed for a 70 km x 30 km modeling domain. This paper presents results of 1D modeling for large floods, both in the natural state and with the proposed bridge, and results of 2D modeling in the natural state. Modeling at this scale requires sound judgment to strike a balance between the desirable level of model detail and the cost of acquiring data for model construction and calibration.
