CagA EPIYA polymorphisms in Colombian Helicobacter pylori strains and their influence on disease-associated cellular responses

AIM: To investigate the influence of the CagA diversity in Helicobacter pylori (H. pylori ) strains from Colombia on the host cell biology. METHODs: Eighty-four H. pylori-cagA positive strains with different Glu-Pro-Ile-Tyr-Ala (EPIYA) motifs patterns, isolated from patients with gastritis (n=17), atrophic gastritis (n=17), duodenal ulcer (n=16), intestinal metaplasia (n=16) and gastric cancer (n=18), were included. To determine the integrity of the cag pathogenicity island (cag PAI) we evaluated the presence of cagA, cagT, cagE, and cag10 genes by polymerase chain reaction. AGs gastric epithelial cellswere infected with each strain and assayed for translo-cation and tyrosine phosphorylation of CagA by western blot, secretion of interleukin-8 (IL-8) by enzyme-linked immuno sorbent assay after taking supernatants from cocultures and cell elongation induction. For cell elongation quantification, coculture photographs were taken and the proportion of "hummingbird" cells (>15 μm) was determined. REsULTs: Overall 72% (60/84) of the strains were found to harbor a functional cag PAI. Levels of phos-phorylated CagA were significantly higher for isolates from duodenal ulcer than the ones in strains from gas-tritis, atrophic gastritis, intestinal metaplasia and gastric cancer (49.1% ± 23.1% vs 21.1% ± 19.5%, P s 26.2%±14.8%, Ps 21.5% ± 19.5%, Ps 29.5% ± 27.1%, P spectively). We observed variable IL-8 expression levels ranging from 0 to 810 pg/mL and from 8.8 to 1442 pg/mL at 6 h and 30 h post-infection, respectively. cagPAI-defective strains did not induce detectable levels of IL-8 at 6 h post-infection. Helicobacter pylori CagA protein polymorphisms and their lack of association with pathogenesis

AIM: To investigate Helicobacter pylori (H. pylori) CagA diversity and to evaluate the association between protein polymorphisms and the occurrence of gastric pathologies. METHODs: One hundred and twenty-two clinical isolates of H. pylori cultured from gastric biopsies obtained from Colombian patients with dyspepsia were included as study material. DNA extracted from isolates was used to determine cagA status, amplifying the C-terminal cagA gene region by polymerase chain reaction. One hundred and six strains with a single amplicon were sequenced and results were used to characterize the 3' variable region of the cagA gene. To establish the number and type of tyrosine phosphorylation motifs Glutamine acid-Proline-Isoleucine-Tyrosine-Alanine (EPI-YA) bioinformatic analysis using Amino Acid sequence Analyzer-Amino Acid sequence Analyzer software was conducted. Analysis of the association between the number of EPIYA motifs and the gastric pathology was performed using χ2 test and analysis of the presence of EPIYA-C motifs in relation to the pathology was made by logistic regression odds ratios. Comparisons among EPIYA types found and those reported in GenBank were performed using a proportion test in statistix Analytical software version 8.0. REsULTs: After amplification of the 3' of the cagA gene, 106 from 122 isolates presented a single amplicon and 16 showed multiple amplicons. As expected, diversity in the size of the cagA unique fragments among isolates was observed. The 106 strains that presented a single amplicon after 3' cagA amplification came from patients with gastritis (19 patients), atrophic gastritis (21), intestinal metaplasia (26), duodenal ulcer (22) and gastric cancer. DNA sequence analysis showed that the differences in size of 3' cagA unique fragments was attributable to the number of EPIYA motifs: 1.9% had two EPIYA motifs, 62.3% had three, 33.0% had four and 2.8% had five motifs. The Puna grasslands support grazing systems that produce meat and wool in multi-species herds,especially from llama(Lama glama)and ***,it is yet unknown whether grazing pressure can modify grassland structure and Plant Functional Types diversity and cover in Puna *** analyzed the relationship between grazing pressure and Plant Functional Types by comparing transects located near stockyards(high grazing pressure)and far from them(low pressure)and by evaluating the relationship between the Plant Functional Types cover to a Grazing Pressure Index(GPI).This index incorporates the heterogeneity of traditional pastoral *** two ecological sites(Ciénego and Tolar),we sampled paired transects(near-far).Plots near and far from corrals were similar in all the variables measured except for plots in Tolar,which tend to have higher total cover,possibly due to fecal fertilization near ***,we recorded an increase in total cover and grass cover at higher GPI in Ciénego,while in Tolar we found lower values of total cover and herbaceous dicotyledonous cover at intermediate intensities of *** only negative relationship found was the cover of clonal and non-clonal shrubs,possibly explained by the greater use of these shrubs as firewood near *** summary,our study does not show important changes in Plant Functional Types as a function of grazing pressure in the studied grasslands probably because grazing pressure is diluted or diminished when livestock rotates in different areas and grazing durations ***,the long grazing history with which the Puna has evolved could also contribute to the co-structuration between native vegetation and livestock farming.

Human action recognition from skeletal data is an important and active area of research in which the state of the art has not yet achieved near-perfect accuracy on many well- known datasets. In this paper, we introduce the Distribution of Action Movements Descriptor, a novel action descriptor based on the distribution of the directions of the motions of the joints between frames, over the set of all possible mo- tions in the dataset. The descriptor is computed as a normal- ized histogram over a set of representative directions of the joints, which are in turn obtained via clustering. While the descriptor is global in the sense that it represents the overall distribution of movement directions of an action, it is able to partially retain its temporal structure by applying a window- ing scheme. Fouling phenomenon is considered among the major reasons that cause significant increase of operating cost of desalination plants equipped with reverse osmosis(RO)*** phenomenon is studied in the present work in the case of RO polyamide aromatic membranes using model seawater containing inorganic salts and colloidal *** solubility conditions of CaCO3 and CasO4 were applied to study RO performances with and without colloid *** experiments,the membrane permeate fluxes were continuously ***,studies of chemical composition,structure,and morphology of the materials deposited on the membrane surface were conducted using energy dispersive microanalysis(EDs)X-ray diffraction and scanning electronic microscopy(sEM).Results show that in conditions of calcium carbonate oversaturation there is a reduction in the permeate flow of 11.2%due to fouling of the membrane by the precipitation of this *** in the same conditions of calcium sulphate oversaturation the reduction of the flow is 5%,so we can conclude that in conditions of oversaturation of both salts,calcium carbonate produces a greater fouling of the membrane that in its view causes greater decrease in the flow of *** this based on the results of the test with both salts in oversaturated *** in the formation of calcite and gypsum crystals onto the membranes as XRD analyses *** presence of colloidal silica in those conditions intensifies strongly the fouling,leading until to 24.1%of permeate flux decrease.

Antifungal activity against Fusarium oxysporum of quinolizidines isolated from three controlled-growth Genisteae plants:structure-activity relationship implications

The Genisteae tribe belongs to the Fabaceae *** wide occurrence of secondary metabolites,explicitly high-lighting the quinolizidine alkaloids(QAs),characterizes this *** the present study,twenty QAs(1-20),including lupanine(1-7),sparteine(8-10),lupanine(11),cytisine and tetrahydrocytisine(12-17),and matrine(18-20)-type QAs were extracted and isolated from leaves of three species(i.e.,Lupinus polyphyllus(’rusell’hybrid),Lupinus muta-bilis,and Genista monspessulana)belonging to the Genisteae *** plant sources were propagated under greenhouse *** isolated compounds were elucidated by analyzing their spectroscopical data(Ms,NMR).The antifungal effect on the mycelial growth of Fusarium oxysporum(Fox)of each isolated QA was then evaluated through the amended medium *** best antifungal activity was found to be for compounds 8(IC_(50)=16.5μM),9(IC_(50)=7.2μM),12(IC_(50)=11.3μM),and 18(IC_(50)=12.3μM).The inhibitory data suggest that some QAs could effi-ciently inhibit Fox mycelium growth depending on particular structural requirements deduced from structure-activity relationship *** identified quinolizidine-related moieties can be involved in lead structures to develop further antifungal bioactives against Fox.

Wereport on the colloidal and rheological characterization of suspensions of commercial single-walled carbon nanotubes(sWCNT)and oxidized single walled carbon nanotubes(sWCNT-oxi24h),in water and fetal bovine serum(FBs).The effects of two surfactants,dodecyl-benzene sodium sulfonate(sBDs)and sodium dodecyl sulfate(sDs)at different concentrations(0.5-2.0%)on the zeta potential and the rheology of the suspensions is also *** suspensions were characterized by Transmission Electron Microscopy(TEM)and Atomic Force Microscopy(AFM).The colloidal stability was determined by measurements of zeta potential in the physiological pH range 6.0surements were implemented at concentrations of 1 and 1.5 mg/ml,and temperatures of 25℃,30℃,and 37℃.The results obtained show that sWCNT suspensions in FBs,with and without addition of surfactants,depict a high tendency to aggregate with the bovine serum albumin(BsA),a processwe could further characterize using Molecular Dynamics(MD)*** suspensions in aqueous media show a negative zeta potential between−60 and−80 mV,whereas in FBs the stability is lower,with values of-20 *** observed no significant changes in the zeta potential with the different *** viscosity values obtained are smaller in water than in FBs,demonstrating the higher stability in water.

AIM:The pathogenesis of hypogonadism in liver cirrhosis is not well *** results from our laboratory showed that IGF-1 deficiency might play a pathogenetic role in hypogonadism of *** administration of IGF-1 for a short period of time reverted the testicular atrophy associated with advanced experimental cirrhosis. The aim of this study was to establish the historical progression of the described alterations in the testes, explore testicular morphology,histopathology,cellular proliferation,integrity of testicular barrier and hypophyso- gonadal axis in rats with no ascitic cirrhosis. METHODs:Male Wistar rats with histologically-proven cirrhosis induced with carbon tetrachloride(CCI_4)for 11 wk, were allocated into two groups(n=12,each)to receive recombinant IGF-1(2 μg/100 gd,sc)for two weeks or *** rats receiving vehicle were used as control group(n=12). REsULTs:Compared to controls,rats with compensated cirrhosis showed a normal testicular size and weight and very few histopathological testicular abnormalities. However,these animals showed a significant diminution of cellular proliferation and a reduction of testicular transferrin *** addition,pituitary-gonadal axis was altered,with significant higher levels of FsH(Pscontrols)and increased levels of LH in untreated cirrhotic ***,IGF-1 treatment normalized testicular transferrin expression and cellular proliferation and reduced serum levels of LH(P=ns vs controls,and Ps untreated cirrhotic group). CONCLUsION:The testicular barrier is altered from an early stage of cirrhosis,shown by a reduction of transferrin expression in sertoli cells,a diminished cellular proliferation and an altered gonadal *** treatment with IGF-1 could be also useful in this initial stage of testicular disorder associated with compensated cirrhosis.
