Elesclomol (ELC) is an anticancer drug inducing mitochondria cytotoxicity through reactive oxygen ***,for the first time,we encapsulate the poorly water soluble ELC in monoolein-based cubosomes stabilized with Pluronic *** uptake and nanocarrier accumulation close to the mitochondria with sub-micrometer distance is identified via three-dimensional (3D) confocal microscopy and edge-to-edge compartment *** monitor the therapeutic effect of the ELC nanocarrier,we apply for the first time,label-free time-lapse multi-photon fluorescence lifetime imaging microscopy (MP-FLIM) to track NAD(P)H cofactors with sub-cellular resolution on live cells exposed to an anticancer *** in vitro cytotoxicity is verified when loading the pre-complexed ELC with copper (ELC-Cu).Importantly,for equivalent copper concentration,cubosomes loaded with ELC-Cu show higher cytotoxicity compared to the free *** novel nanocarrier shows promising features for systemic ELC-Cu administration,and furthermore we establish the MP-FLIM technique for the assessment of anticancer drug delivery systems.

Landscape changes have caused the decline of biodiversity and affected the integrity of aquatic *** this study,we assessed the influence of industrialization associated with urban development on aquatic environments and the biodiversity of semiaquatic bugs(Gerromorpha)at different scales(water quality,habitat,and landscape).Using the simple buffer method,we evaluated nine streams assessing water quality variables,habitat structure,and linear features(radius¼300 m,length¼600 m)for the percentage of land use and land *** results found that species composition of Gerromorpha was mainly affected by primary forest cover,habitat integrity,leaves substrate,and water temperature(29%explanation),and the species richness was mostly influenced by the habitat integrity index(HII)and phosphorus *** model that best explained the variation in species richness showed a positive relationship with phosphorus concentrations and HII(43%variation).Therefore,our results show that those human activities are the main factors altering aquatic biodiversity from local to regional ***,the retention of riparian forests–even in urban and industrialized areas–can maintain the diversity of semiaquatic insect species by providing suitable environments for the habitat,feeding,and reproductive requirements of these aquatic ***,our results also show that the management of riparian vegetation in urban areas and industrial complexes promotes aquatic biodiversity conservation in regions under development in the Amazon.

AIM:To compare the natural history and course of acute diverticulitis in a younger age group with an older population and to evaluate whether younger patients should be managed differently. METHODS:This study was a retrospective review of 157 patients treated with acute diverticulitis between January 1,2004 and December 31,*** was stratified according to the Hinchey classification. Patients were divided into 2 populations:group A≤ 50 years(n=31) ;group B>50 years(n=126) .Mean patient follow-up was 15 mo. rESULTS:The median age was 60 years.A significantly higher proportion of patients in group B presented with complicated diverticulitis(36.5%vs 12.9%,P=0.01) .recurrence was more frequent in group A(25.8%vs 11.1%,P=0.03) and the mean time-torecurrence was shorter(12 mo vs 28 mo,P=0.26) . The most severe recurrent episodes of acute diverticulitis were classified as Hinchey stageⅠand none of the patients required emergency *** multivariate analysis,only age(P=0.024) was identified as an independent prognostic factor for recurrence. CONCLUSION:Based on the results of this study,we recommend that diverticulitis management should be based on the severity of the disease and not on the age of the patient.

目的分析社会隔离量表(Social Isolation Scale,SIS)在葡萄牙社区老年人中应用的信度和效度。方法选取250名社区老年人(≥65岁)进行横向描述性调查研究。调查工具包括社会人口学问卷、SIS、老年抑郁量表-4 (Geriatric Depression Scale-4,GDS-4)、6项卢本社会网络量表(6-item Lubben Social Network Scale,LSNS-6)、孤独自我评估问题和生活满意度量表(Satisfaction with Life Scale,SWLS)。分析结构效度(验证性因素分析)和收敛效度,评估ISI内部信度(综合信度)、外部信度(重测、组内相关系数)和评分者间信度(Cohen's kappa系数)。结果验证性因素分析显示双因素模型具有良好的拟合指数。SIS得分与LSNS-6(r=0.47)、SWLS(r=0.26)、孤独自我评估(r=0.35)和GDS-4(r=-0.16)得分相关。SIS组合信度良好(0.708)。组内相关系数0.84～0.98。Cohen's kappa系数为0.936～1。结论 SIS可作为评估葡萄牙社区老年人社会隔离的有效和可靠工具。医疗保健专业人员,特别是在社区工作的护士,可使用SIS评估老年人的社会隔离状况,以设计、实施和评估干预措施。

Loss or gain of chromosomes,a condition known as aneuploidy,is a common feature of tumor cells and has therefore been proposed as the driving force for *** chromosomal instability can arise during mitosis as a result of mis-segregation of the duplicated sister chromatids to the two daughter *** normal cells,missegregation is usually prevented by the spindle assembly checkpoint(SAC),a sophisticated surveillance mechanism that inhibits mitotic exit until all chromosomes have successfully achieved bipolar attachment to spindle *** abrogation of SAC activity is lethal to normal as well as to tumor cells,as a consequence of massive chromosome ***,many human aneuploid tumor cells exhibit a weakened SAC activity that allows them to tolerate gains or losses of a small number of chromosomes;and interfering with this SAC residual activity may constitute a suitable strategy to kill cancer *** review focuses on the potential link between SAC and tumorigenesis,and the therapeutic strategy to target the SAC for cancer treatment.

Chronic pain, a multidimensional experience affecting individuals’ sensory, cognitive, and emotional aspects, significantly impacts their quality of life. Post-laminectomy syndrome, a condition characterized by persistent back pain following spinal surgery, often leads to disability and increased healthcare utilization. Methods: This randomized, controlled, blind clinical trial aimed to investigate the efficacy of Transcranial Direct Current Stimulation (tDCS) in managing pain from post-laminectomy syndrome in patients. Twenty-four participants were assigned to three groups: sham stimulation, active stimulation over primary motor cortex (M1), or stimulation over dorsolateral prefrontal cortex (DLPFC). Stimulation was administered for five consecutive days, 20 minutes per session, using a current of 1.5 mA through 25 cm2 electrodes. Pain intensity was assessed using Visual analog Scale (VAS) before, during, and after intervention. results: An ANOVA model demonstrates significant reduction in pain intensity compared to baseline in VAS, (F(7, 285) = 12.292;p 0.001;Power = 1.000;η2p = 0.534), in tDCS applied to M1, after five days of intervention. After stimulation, a significant improvement was observed in WHOQoL-Bref Quality of life item 1 (p = 0.04), considering statistical significant difference p 0.05. Correlation between the variables: quality of life, depression, anxiety and pain also demonstrates reduction in depression and anxiety according to Beck’s Depression and Anxiety Inventories (BDI and BAI), p 0.05. This effect was not observed in DLPFC stimulation group. Patients who believed they received active stimulation, in sham group, demonstrated potential for effective blinding. Conclusion: The tDCS applied to primary motor cortex effectively improved pain management and psychiatry symptoms in post-laminectomy syndrome patients. Objective: To evaluate the antioxidant activity of the essential oil obtained from rosmarinus officinalis(r. officinalis) in ethanol-induced gastric ulcer model in ***: The antioxidant properties of the essential oil obtained from r. officinalis were evaluated against gastric injury induced by absolute ethanol. Gastric tissues were prepared to enzymatic assays. The levels of glutathione, lipid peroxides, and the activities of glutathione peroxidase, superoxide dismutase and myeloperoxidase were ***: Ethanol produced severe hemorrhagic lesions in the stomach with ulcerative lesion of(140.2 ± 37.2) mm2. In animals pretreated with essential oil of r. officinalis(50 mg/kg, p.o.), a significant inhibition of mucosal injury of(21.2 ± 7.1) mm2(84%inhibition) was observed. The essential oil of r. officinalis protected the gastric mucosa probably by modulating the activities of the enzymes(superoxide dismutase and glutathione peroxidase) and increasing or maintaining the levels of glutathione. In addition,lipid peroxides levels were reduced. The essential oil of r. officinalis was analyzed by gas chromatography–mass spectrometer and the main constituents were cineole(28.5%),camphor(27.7%) and alpha-pinene(21.3%).Conclusions: We suggest that the monoterpenes present in the essential oil obtained from r. officinalis may be among the active principles responsible for the antioxidant activity shown by essential oil of r. officinalis.
