Spine biomechanical testing method. in the past few d.cad.s have not evolved.beyond.employing either.cad.ver.c stud.es or.finite element mod.ling ***,both these appr.aches may have inher.nt cost and.time *** stud.es ar. the pr.sent gold.stand.r. for.spinal implant d.sign and.r.gulator. appr.val,but they intr.d.ce significant var.ability in measur.ments acr.ss patients,often r.quir.ng lar.e sample *** element mod.ling d.mand. consid.r.ble exper.ise and.can be computationally expensive when complex geometr. and.mater.al nonlinear.ty ar. *** analogue spine mod.ls could.complement these tr.d.tional method. as a low-cost and.high-fid.lity alter.ative.A fully 3d.pr.ntable L-S1 analogue spine mod.l with ligaments is d.veloped.and.valid.ted.in this *** stiffness of the mod.l und.r.pur. bend.ng load.ng in flexion-extension,Later.l Bend.ng(LB)and.Axial r.tation(Ar.is evaluated.and.compar.d.against histor.cal ex vivo and.in silico ***,the effect of inter.pinous,inter.r.nsver.e ligaments and.the Thor.columbar.Fascia(TLF)on spinal stiffness is evaluated.by systematic constr.ction of the *** flexion,mod.l r.nge of Motion(r.M)was 12.92±0.11°(ex vivo:16.58°,in silico:12.96°)at *** LB,aver.ge r.M was 13.67±0.12°at 7.5 Nm(ex vivo:15.21±1.89°,in silico:15.49±0.23°).Similar.y,in Ar.aver.ge r.M was 17.69±2.12°at 7.5Nm(ex vivo:14.12±0.31°,in silico:15.91±0.28°).The ad.ition of inter.pinous and.inter.r.nsver.e ligaments incr.ased.both flexion and.LB stiffnesses by appr.ximately 5%.Ad.ition of TLF showed.incr.ase in flexion and.Ar.stiffnesses by 29%and.24%,*** novel mod.l can r.pr.d.ce physiological r.Ms with high r.peatability and.could.be a useful open-sour.e tool in spine biomechanics.

The sequence of Gid. （a gene for.a gibber.llin （GA） r.ceptor.fr.m r.ce） was used.to id.ntify a putative or.hologue fr.m bar.ey. This was expr.ssed.in E. coil, and.pr.d.ced.a pr.tein that was able to bind.GA in vitr. with both str.ctur.l specificity and.satur.bility. Its potential r.le in GA r.sponses was investigated.using bar.ey mutants with r.d.ced.GA sensitivity （gsel mutants）. Sixteen d.ffer.nt gsel mutants each car.ied.a unique nucleotid. substitution in this sequence. In all but one case, these changes r.sulted.in single amino acid.substitutions, and. for.the r.maining mutant, a substitution in the 5＇ untr.nslated.r.gion of the mr.A is pr.posed.to inter.er. with tr.nslation initiation. Ther. was per.ect linkage in segr.gating populations between new mutant alleles and.the gsel phenotype, lead.ng to the conclusion that the putative GId. GA r.ceptor.sequence in bar.ey cor.espond. to the Gsel locus. d.ter.ination of end.genous GA contents in one of the mutants r.vealed.enhanced.accumulation of bioactive GA1, and.a d.ficit of C20 GA pr.cur.or.. All of the gsel mutants had.r.d.ced.sensitivity to exogenous GA3, and.to AC94377 （a GA analogue） at concentr.tions that ar. nor.ally ＇satur.ting＇, but, at much higher.concentr.tions, ther. was often a consid.r.ble r.sponse.
