Combined influence of basal media and fibroblast growth factor on the expansion and differentiation capabilities of adipose-derived stem cells

Background: Interest in adipose-derived stem cells (ASCs) has increased in recent years due to their multi-linagedifferentiation capabilities. While much work has been done to optimize the differentiation media, few studies havefocused on examining the influence of different expansion media on cell behavior. In this study, three basal media(low glucose Dulbecco’s modified Eagle’s medium (DMEM), high glucose DMEM and DMEM-F12) supplementedwith or without fibroblast growth factor 2 (FGF) were examined to assess their suitability for expanding ***: Flow cytometry, colony-forming unit assays (CFU-Fs) and differentiation assays were utilized to study cellbehavior. High glucose media CFU-Fs produced fewest colonies while the addition of FGF increased colony *** passage 2, the majority of cells were positive for CD44, 45, 73, 90 and 105 and negative for CD14, 31 and 45,indicating a mesenchymal phenotype. A sub-population of CD34 positive cells was present among passage 2 cells;however, by passage 4 the cells were negative for CD34. FGF has a negative effective on passage 4 ASC adipogenesisand high glucose media plus FGF-enhanced osteogenic capacity of passage 4 ASCs. FGF supplemented basal mediawere most suitable for chondrogenesis. High glucose media plus FGF appeared to be the most beneficial for primingASCs to induce a keratocyte ***: These findings demonstrate the reciprocal effect FGF and basal media have on ASCs. This research hasimplications for those interested regenerating bone, cartilage, cornea or adipose tissues.

A Comparative Analysis of Quantitative Metrics of Root Architecture

High throughput phenotyping is important to bridge the gap between genotype and *** methods used to describe the phenotype therefore should be robust to measurement errors,relatively stable over time,and most importantly,provide a reliable estimate of elementary phenotypic *** this study,we use functional-structural modeling to evaluate quantitative phenotypic metrics used to describe root architecture to determine how they fit these *** results show that phenes such as root number,root diameter,and lateral root branching density are stable,reliable measures and are not affected by imaging method or *** aggregating multiple phenes such as total length,total volume,convex hull volume,and bushiness index estimate different subsets of the constituent phenes;they however do not provide any information regarding the underlying phene *** of phene aggregates are not unique representations of underlying constituent phenes:multiple phenotypes having phenes in different states could have similar aggregate *** growth angle is an important phene which is susceptible to measurement errors when 2D projection methods are *** that aggregate phenes which are complex functions of root growth angle and other phenes are also subject to measurement errors when 2D projection methods are *** results support the hypothesis that estimates of phenes are more useful than metrics aggregating multiple phenes for phenotyping root *** propose that these concepts are broadly applicable in phenotyping and phenomics.

Magnetic resonance imaging in rectal cancer:A surgeon's perspective

Case study approach to modeling historical disinfection by-product exposure in Iowa drinking waters

In the 1980 s, a case–control epidemiologic study was conducted in Iowa（USA） to analyze the association between exposure to disinfection by-products（DBps） and bladder cancer risk. Trihalomethanes（THMs）, the most commonly measured and dominant class of DBps in drinking water, served as a primary metric and surrogate for the full DBp *** THM exposure was calculated, based on rough estimates of past levels in Iowa. To reduce misclassification, a follow-up study was undertaken to improve estimates of past THM levels and to re-evaluate their association with cancer risk. In addition, the risk associated with haloacetic acids, another class of DBps, was examined. In the original analysis, surface water treatment plants were assigned one of two possible THM levels depending on the point of chlorination. The re-assessment considered each utility treating surface or groundwater on a case-by-case basis. Multiple treatment/disinfection scenarios and water quality parameters were considered with actual DBp measurements to develop estimates of past levels. The highest annual average THM level in the re-analysis was156 μg/L compared to 74 μg/L for the original analysis. This allowed the analysis of subjects exposed at higher levels（〉 96 μg/L）. The re-analysis established a new approach, based on case studies and an understanding of the water quality and operational parameters that impact DBp formation, for determining historical exposure.

The green environmental laws and regulations are legislated, implemented, and enforced in many countries and economic regions. The provision of green products and services are the fast growing trend in global consumer markets. Therefore, introducing new products with environmental considerations becomes critical for global brand manufacturers. This research depicts an integrated and intelligent eco- and inno-product design methodology to support environmental friendly green product development. The methodology adopts approaches, such as life cycle assessment （LCA）, quality function deploymnet for environement （QFDE）, theory of inventive problem solving （TRIZ） and back-propagation network （BpN） to achieve eco- and inno-design objectives. LCA evaluates and compares the environmental impacts of production. QFDE transforms high-level concerns of environment into design requirements. When there are many historical QFDE data, the BpN prediction model is trained and deployed to automate the specifications of green design improvement. TRIZ is to support the creation of innovative product design ideas effectively and efficiently during the concept design stage. Finally, this paper presents two eco-design cases of power adaptor to demonstrate the proposed methodology.

GABA Attenuates L-DOpA-Induced Striatal and Nigral ERK1/2 Signaling in a Rat Model of parkinson’s Disease

L-DOpA is the primary drug used to treat parkinson’s disease (pD) symptoms, but motor side effects limit its long term use. previous experimental studies show that L-DOpA acts on supersensitive D1 receptors in the basal ganglia to induce extracellular signal-regulated kinases 1 and 2 (ERK1/2), a pair of MAp-kinase proteins that may be involved in induction of motor side effects. Since GABA is known to be intimately involved in basal ganglia function, we investigated whether elevating GABA levels via a GABA-transaminase (GABA-T) inhibitor affects the L-DOpA-induced ERK1/2 phosphorylation in the striatum and substantia nigra (SN) using a rat model of pD. Unilateral dopaminergic lesions of median forebrain bundle neurons were done using the neurotoxin 6-hydroxydopamine. Rats were prescreened for the extent of the lesion by apomorphine-induced rotation test. Lesioned rats were treated with aminooxyacetic acid (AOAA, a GABA-T inhibitor), L-DOpA, or in combination. Immunohistochemistry of tyrosine hydroxylase (TH, a direct indicator of dopaminergic lesion), substance p (Sp, an indirect marker that decreases after lesion), and phospho-ERK1/2 was done using slices at the level of striatum and SN. Unilateral dopaminergic lesioned rats, as expected, exhibited >90% TH loss and a modest Sp loss in the striatum and SN. L-DOpA alone induced a 343% and 330% increase in phospho-ERK1/2 in the striatum and SN, respectively. We report here a novel finding that pretreatment with AOAA attenuated the L-DOpA induced increase in phospho-ERK1/2 by 62% and 68% in the striatum and SN, respectively, suggesting a DA-GABA-ERK1/2 link in the therapeutic and/or side effects of L-DOpA.

Dopamine and GABA Interaction in Basal Ganglia: GABA-A or GABA-B Receptor Stimulation Attenuates L-DOpA-Induced Striatal and Nigral ERK1/2 Signaling in a Rat Model of parkinson’s Disease

parkinson’s disease (pD) is characterized by degeneration of nigrostriatal dopamine (DA) neurons. The primary drug used to treat pD symptoms is L-DOpA, but side effects such as dyskinesias limit its use. previous findings show that L-DOpA treatment induces extracellular signal-regulated kinase (ERK1/2), a MAp-kinase protein. γ-aminobutyric acid (GABA) is intimately involved in basal ganglia function. Our previous study using a unilaterally lesioned rat model of pD indicated that elevating GABA levels by GABA transaminase inhibitor, aminooxyacetic acid significantly attenuated L-DOpA-induced ERK phosphorylation in the striatum and substantia nigra (SN). The aim of the present study was to assess the role of GABA-A and GABA-B receptor by using a selective agonists, muscimol and baclofen respectively, on L-DOpA-induced ERK phosphorylation in the striatum and SN. Unilaterally 6-OHDA-lesioned rats were prescreened by apomorphine induced rotation test for the extent of DA loss. Lesioned rats were treated with L-DOpA alone or after muscimol or baclofen pretreatment. Appropriate control groups were used. phospho-ERK levels, tyrosine hydroxylase (to ascertain DA loss) and substance p (an indirect marker for DA loss) levels were assessed by immunohistochemistry using coronal slices at the level of striatum and SN. L-DOpA administration induced a robust increase (>300%) in phospho-ERK1/2 levels in the striatum and SN. Muscimol as well as baclofen pretreatment attenuated the L-DOpA-induced increase in phospho-ERK1/2 levels by >60% in the striatum and SN. Muscimol and baclofen pretreatment also greatly reduced the number of L-DOpA induced phospho-ERK1/2 stained cells in the striatum as well as the contralateral rotational behavior. The present data taken together with our previous study indicate that the L-DOpA induced increase in ERK1/2 is attenuated by GABA via a GABA-A and GABA-B receptor linked mechanism.
