The bone marrow contains a heterogeneous milieu of cells, including macrophages, which are key cellular mediators for resolving infection and inflammation. Macrophages are most well known for their ability to phagocytose foreign bodies or apoptotic cells to maintain homeostasis; however, little is known about their function in the bone microenvironment. In the current study, we investigated the in vitro interaction of murine macrophages and bone marrow stromal cells （BMSCs）, with focus on the juxtacrine induction of IL-6 signaling and the resultant effect on BMSC migration and growth. The juxtacrine interaction of primary mouse macrophages and BMSCs activated IL-6 signaling in the co-cultures, which subsequently enhanced BMSC migration and increased BMSC numbers. BMSCs and macrophages harvested from IL-6 knockout mice revealed that IL-6 signaling was essential for enhancement of BMSC migration and increased BMSC numbers via juxtacrine interactions. BMSCs were the main contributor of IL-6 signaling~ and hence activation of the IL-6/ gp130/STAT3 pathway. Meanwhile, macrophage derived IL-6 remained important for the overall production of IL-6 protein in the co-cultures. Taken together, these findings show the function of macmphages as co-inducers of migration and growth of BMSCs, which could directly influence bone formation and turnover.

Two stochastic models are derived for a susceptible-infectious-susceptible epidemic spreading through a metapopulation： a continuous time Markov chain （CTMC） model and an It6 stochastic differential equation （SDE） model. The stochastic models are numerically compared. Close agreement suggests that computationally intense CTMC simulations can be approximated by simpler SDE simulations. Differential equations for the moments of the SDE probability distribution are also derived, the steady states are solved numerically using a moment closure technique, and these results are compared to simulations. The moment closure technique only coarsely approximates simulation results. The effect of model parameters on stability of the disease-free equilibrium is also numerically investigated.

The role of androgen receptor （AR） in the initiation and progression of prostate cancer （PCa） is well established. Competitive inhibition of the AR ligand-binding domain （LBD） has been the staple of antiandrogen therapies employed to combat the disease in recent years. However, their efficacy has often been limited by the emergence of resistance, mediated through point mutations, and receptor truncations. As a result, the prognosis for patients with malignant castrate resistant disease remains poor. The amino-terminal domain （NTD） of the AR has been shown to be critical for AR function. Its modular activation function （AF-1） is important for both gene regulation and participation in protein-protein interactions. However, due to the intrinsically disordered structure of the domain, its potential as a candidate for therapeutic intervention has been dismissed in the past. The recent emergence of the small molecule EPI-O01 has provided evidence that AR-NTD can be targeted therapeutically, independent of the LBD. Targeting of AR-NTD has the potential to disrupt multiple intermolecular interactions between AR and its coregulatory binding partners, in addition to intramolecular cross-talk between the domains of the AR. Therapeutics targeting these protein-protein interactions or NTD directly should also have efficacy against emerging AR splice variants which may play a role in PCa progression. This review will discuss the role of intrinsic disorder in AR function and illustrate how emerging therapies might target NTD in PCa.

Patent documents are unique external sources of information that reveal the core technology underlying new inventions. Patents also serve as a strategic data source that can be mined to discover state-of-the-art technical development and subsequently help guide R＆D investments. This research incorporates an ontology schema to extract and represent patent concepts. A clustering algorithm with non-exhaustive overlaps is proposed to overcome deficiencies with exhaustive clustering methods used in patent mining and technology discovery. The non-exhaustive clustering approach allows for the clustering of patent documents with overlapping technical findings and claims, a feature that enables the grouping of patents that define related key innovations. Legal advisors can use this approach to study potential cases of patent infringement or devise strategies to avoid litigation. The case study demonstrates the use of non-exhaustive overlaps algorithm by clustering US and japan radio frequency identification （RFID） patents and by analyzing the legal implications of automated discovery of patent infringement.

The outage of a transformer in a power generation system will generally result in a shutdown of the entire system. Consequentially, large amounts of revenue are lost to the plant owner with a substantial negative impact on the civil community and industrial customers. This paper analyzes the current maintenance practices （i.e., an as-is process model） of a transformer manufacturing company, which provides maintenance services to power generation plants and reports the major weaknesses in current business practices. The maintenance weaknesses include isolated processes and paper based information communication. These inefficient processes result in service delays, cost increases, and inappropriate responses to customers＇ specific requests and complaints. To improve the efficiency and flexibility of the entire transformer maintenance service system, an e-integration approach is implemented with three key tasks. First, the current business processes are re-engineered to provide an improved （to-be） business model. Then, we replace paper based documentation methods with an electronic filing system for efficient and cost-effective information exchange. Afterward, the improved process model is implemented as an Internet based system to enhance the interactions among the transformer manufacturer, end-users, and service contractors for customized maintenance services. The case study demonstrates that implementing the e-integration approach increases the efficiency and effectiveness of the customized maintenance services.

Large sized power transformers are important parts of the power supply chain. These very critical networks of engineering assets are an essential base of a nation＇s energy resource infrastructure. This research identifies the key factors influencing transformer normal operating conditions and predicts the asset management lifespan. Engineering asset research has developed few lifespan forecasting methods combining real-time monitoring solutions for transformer maintenance and replacement. Utilizing the rich data source from a remote terminal unit （RTU） system for sensor-data driven analysis, this research develops an innovative real-time lifespan forecasting approach applying logistic regression based on the Weibull distribution. The methodology and the implementation prototype are verified using a data series from 161 kV transformers to evaluate the efficiency and accuracy for energy sector applications. The asset stakeholders and suppliers significantly benefit from the real-time power transformer lifespan evaluation for maintenance and replacement decision support.

Engineering and research teams often develop new products and technologies by referring to inventions described in patent databases. Efficient patent analysis builds R＆D knowledge, reduces new product development time, increases market success, and reduces potential patent infringement. Thus, it is beneficial to automatically and systematically extract information from patent documents in order to improve knowledge sharing and collaboration among R＆D team members. In this research, patents are summarized using a combined ontology based and TF-IDF concept clustering approach. The ontology captures the general knowledge and core meaning of patents in a given domain. Then, the proposed methodology extracts, clusters, and integrates the content of a patent to derive a summary and a cluster tree diagram of key terms. Patents from the International Patent Classification （IPC） codes B25C, B25D, B25F （categories for power hand tools） and B24B, C09G and H011 （categories for chemical mechanical polishing） are used as case studies to evaluate the compression ratio, retention ratio, and classification accuracy of the summarization results. The evaluation uses statistics to represent the summary generation and its compression ratio, the ontology based keyword extraction retention ratio, and the summary classification accuracy. The results show that the ontology based approach yields about the same compression ratio as previous non-ontology based research but yields on average an 11% improvement for the retention ratio and a 14% improvement for classification accuracy.
