Objective To assess the antimutagenicity of antioxidant vitamins （vitamins A, C, and E） as expressed by their efficacy to lower aflatoxin-induced mutations. Methods The Muller-5 method for mutation detection was used to assay the frequency of X-chromosome linked recessive lethal mutations （XRLMs） in Drosophila. Larvae were exposed to dietary concentration of aflatoxins and/or the human therapeutic doses of any of the three antioxidant vitamins. Absence of normal eyed males among M2 progeny gave an indication of mutation induction. Results Aflatoxin supplimentation significantly increased the incidence of XRLMs in Drosophila. Mutation frequency was also raised a little above the control level in case of vitamin treatment. However, notable mitigation in mutation frequency was registered when aflatoxin-treated larvae were concomitantly fed with any of the three antioxidant vitamins. Conclusion Aflatoxin exposure can enhance the frequency of gene mutation in Drosophila which is significantly lowered by each of the three antioxidant vitamins. The degree of amelioration produced by them is almost identical. This mitigation is based on the scavenging/trapping by antioxidant vitamins of DNA-reactive products （metabolites and radicals） emanating from aflatoxin metabolism.

Objective:To explore immunochemical characterization of antigens of Brucella canis(B. canis),and the use in seroprevalence study of canine ***:External hot phosphate buffer saline extract(HPBSE) and internal sonicated(SA) antigens were prepared from *** strain MEX 51 and imniunochemically *** antigens were used to test 527 serum samples of dogs by 2-mercaptoethanol-tubc agglutination test(2 ME-TAT), agar gel immunodiffusion test(AGID).dot-ELISA and indirect enzyme-linked immunosorbent assay(I-ELISA) to assess the seroprevalence of canine ***:The protein content of HPBSE and SA antigens was 0.387 mg/*** 0.195 mg/mL,respectively,whereas carbohydrate content was 0.174 mg/mL and 0.150 mg/mL,*** sodium dodecyl sulfate-polyacrylamide gel electrophoresis(12.5%) of HPBSE and SA,revealed 6 and 8 visible peptide bands ranging from 18-80 kDa and 12-45 kDa,*** blot analysis showed immunodominant bands of MW 12.28.39 and 45 kl)a for HPBSE and 20-24 kl)a for SA. The AGII) revealed HPBSE as more specific antigen than SA but both I-ELISA and dot-ELISA indicated SA antigen to be more specific and reliable than *** seroprevalence of canine brucellosis was 2.27%by 2ME-TAT.1.5%by AGID.3.03%by dot-ELISA and 16.12%by I-ELISA. Conclusions:On the basis of the results of present study,we concluded that HPBSE is suitable antigen for AGID,which is more specific:whereas SA antigen is suitable for I-ELISA,which is highly ***,initial screening of serum samples should be carried out by I-ELISA followed by confirmation with AGID.

Evaluation of retinal nerve fiber layer thickness profile in thyroid ophthalmopathy without optic nerve dysfunction

AIM： To evaluate retinal nerve fiber layer（RNFL）thickness profile in patients of thyroid ophthalmopathy with no clinical signs of optic nerve ***： A prospective, case-control, observational study conducted at a tertiary care centre. Inclusion criteria consisted of patients with eyelid retraction in association with any one of： biochemical thyroid dysfunction, exophthalmos, or extraocular muscle involvement; or thyroid dysfunction in association with either exophthalmos or extra-ocular muscle involvement;or a clinical activity score（CAS）〉3/7. Two measurements of RNFL thickness were done for each eye, by Cirrus HD-optical coherence tomography 6mo ***： Mean age of the sample was 38.75y（range13-70y） with 18 males and 22 females. Average RNFL thickness at first visit was 92.06 ±12.44 μm, significantly lower than control group（101.28±6.64 μm）（P =0.0001）.Thickness of inferior quadrant decreased from 118.2 ±21.27 μm to 115.0 ±22.27 μm after 6mo（P =0.02）. There was no correlation between the change in CAS and RNFL thickness. CONCLUSION： Decreased RNFL thickness is an important feature of thyroid orbitopathy, which is an inherent outcome of compressive optic neuropathy of any etiology. Subclinical RNFL damage continues in the absence of clinical activity of the disease. RNFL evaluation is essential in Grave＇s disease and active intervention may be warranted in the presence of significant damage.

Common mechanisms plants use to translate the external stimuli into cellular responses are the activation of mitogen-activated protein kinase （MAPK） cascade. These MAPK cascades are highly conserved in eukaryotes and consist of three subsequently acting protein kinases, MAP kinase kinase kinase （MAPKKK）, MAP kinase kinase （MAPKK） and MAP kinase （MAPK） which are linked in various ways with upstream receptors and downstream targets. Plant MAPK cascades regulate numerous processes, including various environmental stresses, hormones, cell division and developmental processes. The number of MAPKKs in Arabidopsis and rice is almost half the number of MAPKs pointing important role of MAPKKs in integrating signals from several MAPKKKs and transducing signals to various MAPKs. The cross talks between different signal transduction pathways are concentrated at the level of MAPKK in the MAPK cascade. Here we discussed the insights into MAPKK mediated response to environmental stresses and in plant growth and development.

Log-normal flux distribution of bright Fermi blazars

We present the results of the γ-ray flux distribution study on the brightest blazars which were observed by Fermi-LAT. We selected 50 of the brightest blazars based on the maximum number of detections reported in the Third LAT AGN Catalog. We performed standard unbinned maximum likelihood analysis on the LAT data during the period between August 2008 and December 2016, in order to obtain the average monthly flux. After quality cuts, blazars for which at least 90% of the total flux had survived were selected for further study, and this included 19 FSRQs and 19 BL Lacs. The Anderson-Darling and χ2 tests suggest that the integrated monthly flux follows a log-normal distribution for all sources, except for three FSRQs for which neither a normal nor a log-normal distribution was preferred. A double log-normal flux distribution tendency was observed in these sources, though this has to be confirmed with improved statistics. We also found that the standard deviation of the log-normal flux distribution increases with the mean spectral index of the blazar, and can be fitted with a line of slope 0.24±0.04. We repeat our study on three additional brightest unclassified blazars to identify their flux distribution properties. Based on the features of their log-normal flux distribution, we infer these unclassified blazars may be closely associated with FSRQs. We also highlight that considering the lognormal behavior of the flux distribution of blazars, averaging their long term flux on a linear scale can largely underestimate the nominal flux and this discrepancy can propagate down to the estimation of source parameters through spectral modeling.

IFT20 is the smallest member of the intraflagellar transport protein（IFT） complex B. It is involved in cilia formation. Studies of IFT20 have been confined to ciliated cells. Recently, IFT20 was found to be also expressed in non-ciliated T cells and have functions in immune synapse formation and signaling in vitro. However, how IFT20 regulates T-cell development and activation in vivo is still unknown. We deleted the IFT20 gene in early and later stages of T-cell development by crossing IFT20flox/flox（IFT20f/f） mice with Lck-Cre and CD4-Cre transgenic mice, and investigated the role of IFT20 in T-cell maturation and in the development of T cell-mediated collagen-induced arthritis（CIA）. We found that both Lck-Cre/IFT20f/f and CD4-Cre/IFT20f/f mice were indistinguishable from their wild-type littermates in body size, as well as in the morphology and weight of the spleen and thymus. However, the number of CD4- and CD8-positive cells was significantly lower in thymus and spleen in Lck-Cre/IFT20f/f mice. Meanwhile, the incidence and severity of CIA symptoms were significantly decreased, and inflammation in the paw was significantly inhibited in Lck-Cre/IFT20f/f mice compared to Lck-Cre/IFT201/1littermates. Deletion IFT20 in more mature T cells of CD4-Cre/IFT20f/f mice had only mild effects on the development of T cells and CIA. The expression of IL-1b, IL-6 and TGF-b1 were significantly downregulated in the paw of Lck-Cre/IFT20f/f mice, but just slight decreased in CD4-Cre/IFT20f/f mice. These results demonstrate that deletion of IFT20 in the early stage of T-cell development inhibited CIA development through regulating T-cell development and the expression of critical cytokines.
