Rock1 Inhibitor Abrogates Chronic Rejection in Rat Cardiac Model System

Background: We have shown previously that the abrogation of acute and chronic rejection of rat cardiac allografts occurs through the down-regulation of RhoA pathway and involves the changes in RhoA kinase (ROCK)-dependent actin cytoskeleton and T cell motility. Here we studied the ability of the Y-27632, a highly selective inhibitor of Rho-associated protein kinase p160ROCK (ROCK1), to abrogate chronic rejection of the allograft and influence T cell infiltration. Methods: Heterotopic cardiac transplants were performed between donor Wistar Furth (WF) and ACI recipient rats. Controls received sub-therapeutic dose of cyclosporine (CsA, 10 mg/kg) for 3 days or 7 days therapeutic dose of cyclosporine. The experimental groups of ACI recipient received one preoperative dose of the Y-27632 inhibitor (2 mg/kg, gavage feed) in conjunction with the sub-therapeutic dose of CsA for 3 days or inhibitor alone for 7 days. The cardiac grafts were harvested at 100 days of post-transplantation for histological and immunohistochemical assessment of chronic rejection, vascular sclerosis, and infiltration by different T cell subtypes. Results: Cardiac allografts from recipients exposed to Y-27632 inhibitor in conjunction with sub-therapeutic dose of CsA showed drastically reduced vascular sclerosis, minimal myocardial total cellular infiltration, and were selectively infiltrated with Foxp3+ T regulatory (Treg) cells. Conclusions: Our novel finding that a single dose of the ROCK1 inhibitor Y-27632 attenuates chronic rejection in rat cardiac model system by promoting development of Treg cells warrants its potential as a novel therapeutic agent specific for the inhibition of chronic rejection.

In malaria-endemic regions,people often get exposed to various pathogens simultaneously,generating coinfection *** such scenarios,overlapping symptoms pose serious diagnostic *** delayed diagnosis may lead to an increase in disease severity and catastrophic *** coronavirus disease 2019(COVID-19)pandemic caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has affected various areas globally,including malaria-endemic *** Plasmodium and SARS-CoV-2 co-infection and its effect on health are yet *** present a case report of a previously healthy,middle-aged individual from the malaria-endemic area who suffered SARS-CoV-2 and Plasmodium falciparum *** patient developed severe disease indications in a short time *** patient showed neurological symptoms,altered hematological as well as liver-test parameters,and subsequent death in a narrow time *** hereby discuss the various aspects of this case regarding treatment and hematological ***,we have put forward perspectives related to the mechanism behind severity and neurological symptoms in this fatal parasite-virus co-infection *** malaria-endemic regions,due to overlapping symptoms,suspected COVID-19 patients should also be monitored for diagnosis of malaria without any *** SARS-CoV-2 and Plasmodium co-infection could increase the disease severity in a short time *** treatment,dexamethasone may not help in severe cases having malaria as well as COVID-19 positive status and needs further exploration.

The different investigation has been carried out on the biological activities of titanium dioxide nanoparticle but the effect of this nano product on the antibacterial activity of different antibiotics has not been yet demonstrated. In this study the nano size TiO2 is synthesized using citric acid and alpha dextrose and the enhancement effect of TiO2 nanoparticle on the antibacterial activity of different antibiotics was evaluated against Methicillin-resistant Staphylococcus aureus (MRSA). During the present study, different concentrations of nano-scale TiO2 were tested to find out the best concentration that can have the most effective antibacterial property against the MRSA culture. Disk diffusion method was used to determine the antibacterial activity of these antibiotics in the absence and presence of sub inhibitory concentration of TiO2 nano particle. A clinical isolate of MRSA, isolated from Intensive Care Unit (ICU) was used as test strain. In the presence of sub-inhibitory concentration of TiO2 nanoparticle (20 μg/disc) the antibacterial activities of all antibiotics have been increased against test strain with minimum 2 mm to maximum 10mm. The highest increase in inhibitory zone for MRSA was observed against pencillin G and amikacin (each 10 mm). Conversely, in case of nalidixic acid, TiO2 nanoparticle showed a Synergic effect on the antibacterial activity of this antibiotic against test strain. These results signify that the TiO2 nanoparticle potentate the antimicrobial action of beta lactums, cephalosporins, aminoglycosides, glycopeptides, macrolids and lincosamides, tetracycline a possible utilization of nano compound in combination effect against MRSA.

From the evolution of the mankind, Turmeric has been used in conventional medication. India is in lead for producing, marketing and exporting the Turmeric and its value added products. Curcuma longa (Turmeric) is an Indian rhizomatous medicinal herb from the Zingiberaceae family that is common and widely available across the globe. The components of Turmeric are curcumin, demethoxycurcumin and bisdemethoxycurcumin and these are collectively known as curcuminoids. Curcumin, the active ingredient of Turmeric is generally investigated by the scientific community for its wide range of antioxidant activity, anti-Inflammatory properties and anti-cancer activity, anti-metabolic syndrome activities, neuroprotective activity, antimicrobial effects, anti-arthritis effects, anti-viral effects, anti-asthma and anti-diabetic effects, anti-obesity, cardio and liver toxicity protection activity, anti-depression and anxiety activities. Turmeric has been widely used as a typical household treatment for cough, sore throat, respiratory ailments and could be an effective immunity booster against SARS-CoV-2 therapy during the ongoing pandemic situation. Safety evaluation studies indicate that both turmeric and curcumin are well tolerated at a very high dose without any toxic effects. Thus, turmeric and its constituents have the potential for the development of modern medicine for the treatment of various diseases. So in this review, we describe the various metabolic roles of curcumin and activities for the benefit of human health.

The Epidemiology and Spatial Analysis of Stroke in Trinidad and Tobago in the First Decade of the 21<sup>st</sup>Century (2000-2009)

Objective: To investigate the pattern and distribution of stroke in Trinidad and Tobago from 2000-2009. To identify the prevalence of co-morbid conditions among new stroke patients during the period under surveillance. Methods: Data were collected from May 2010 to July 2010 from the clinic of 728 new persons treated stroke at one of the main treatment centers. Variables measured included age, gender ethnicity, smoking status and co-morbid conditions. SPSS (Version 17) for Windows and ARC GIS version 9.3 were used to facilitate both descriptive and inferential data analysis. Results: Of the 728 new hospital admissions for the period January 2000-December 2009 for stroke, 369 (50.7%) were males and 359 (49.3%) were females. 59.8% were South-East Asian;30.5% were African and 9.7% were mixed ethnicity. The predominant age group was 60 - 69 years (n = 215, 29.5%) while less than 1% were under 30. Ischemic stroke accounted for 352 (48.4%) of all new cases for the period;Hemorrhagic stroke accounted for 14.6% (n = 107), with 37% (n = 269) classified as other unspecified condition (including unknown). Of the 728 cases examined, 171 patients died before being discharged and 552 were treated and discharged. Information of 5 cases was not available. Using this data, the overall case fatality ratio was calculated as 23.5%, with the case fatality ratio for males being 23.2% and the equivalent ratio for females being 22.9%. Using a standard classification, the majority, (n = 389, 53.4%), of cases were classified as mild;246 (33.8%) were deemed moderate, and 93 (12.8%) were severe cases. Hypertension was clinically diagnosed in 80.9% of the cases;56.3% were diabetic, and 21.7% were classified as smokers having been either past or current smokers. Other lifestyle risk factors such as obesity and exercise were not examined due to the lack of the relevant data. Objective:To evaluate the survival outcomes for a cohort of nasopharyngeal cancer with intracranial extension(ICE)treated with induction chemotherapy(ICT)followed by chemo-intensity-modulated radiotherapy(CTRT)at a tertiary cancer ***:We retrospectively analyzed 45 patients with histologically proven,non-metastatic NPC with ICE treated at our institute between October 2008 and October *** were classified as minor ICE or major ICE,based on the extent of *** the patients received 2-3 cycles of a taxane-based ICT regimen followed by *** was delivered with"riskadapted"intensity-modulated radiotherapy(IMRT)technique in all ***:After a median follow up of 45 months(range:8-113 months),the estimated 5-year DFS,LRFS,DMFS,and OS of the entire cohort was 58%,82%,67%and 74%*** multivariate analysis,histological subtype was an independent predictor of LRFS,and age was an independent predictor of *** extent of ICE showed only a trend towards worse DFS(P=0.06).None of the factors significantly predicted for DMFS or ***,N-stage,and response to ICT did not significantly affect any of the *** 2 or worse subcutaneous fibrosis was seen in 22%of patients and grade 2 or worse xerostomia was seen in 24%of patients at last follow ***-three percent of the patients developed clinical hypothyroidism at last follow *** of the patients experienced any neurological or vascular ***:Taxane-based induction chemotherapy followed by chemo-intensity modulated radiotherapy resulted in excellent locoregional control and survival with acceptable toxicities in patients of nasopharyngeal cancer with intracranial *** metastasis continues to be the predominant problem in these patients.
