Antibiotic resistance and cagA gene correlation:A looming crisis of Helicobacter pylori

AIM:To determine antibiotic resistance of Helicobacter pylori(***) in Pakistan and its correlation with host and pathogen associated ***:A total of 178 strains of *** were isolated from gastric biopsies of dyspeptic *** patterns against first and second-line antibiotics were determined and trends of resistance were analyzed in relation to the sampling period,gastric conditions and cagA gene *** effect of cagA gene on the acquisition of resistance was investigated by mutant selection ***:The observations showed that monoresistant strains were prevalent with rates of 89% for metronidazole,36% for clarithromycin,37% for amoxicillin,18.5% for ofloxacin and 12% for ***,clarithromycin resistance was on the rise from 2005 to 2008(32% vs 38%,P = 0.004) and it is significantly observed in non ulcerative dyspeptic patients compared to gastritis,gastric ulcer and duodenal ulcer cases(53% vs 20%,18% and 19%,P = 0.000).On the contrary,metronidazole and ofloxacin resistance were more common in gastritis and gastric ulcer *** analysis and frequencies of resistant mutants in vitro correlated with the absence of cagA gene with metronidazole and ofloxacin ***:The study confirms the alarming levels of antibiotic resistance associated with the degree of gastric inflammation and cagA gene carriage in *** strains.

Therapeutic effect of hydrogen peroxide via altered expression of glutathione S-transferase and peroxiredoxin-2 in hepatocellular carcinoma

Background:Hepatocellular carcinoma(HCC)has a high incidence and mortality that epitomizes one of the prominent causes of cancer-related death *** therapeutic approaches are therefore *** oxygen species(ROS)are necessary for maintaining cell *** ROS is involved in HCC progression,hydrogen peroxide(H2O2)has anti-proliferative effect on ***:HCC Huh-7 cells were cultured and incubated with various concentrations of *** activity,levels of malondialdehyde,glutathione and protein oxidation were measured in treated and untreated Huh-7 ***,untreated and treated Huh-7 cells were subjected to two dimensional gel electrophoresis and identified protein spots which were differentially expressed by LC-MS/MS analysis.q RT-PCR was performed to validate the identified ***:H2O2 depleted glutathione(GSH)with the concomitant up-regulation of GSTP1 and Prx2.H2O2 also increased malondialdehyde and protein oxidation,decreased the activity of paraoxonase in Huh-7 ***:H2O2 could be used as a novel therapeutic agent that might be beneficial in inducing cell cytotoxicity and hence suppress HCC proliferation.

Eligibility of persons who inject drugs for treatment of hepatitis C virus infection

In this decade, an increase is expected in end-stageliver disease and hepatocellular carcinoma, most commonly caused by hepatitis C virus(HCV) *** people who inject drugs(PWID) are the ma-jor source for HCV infection, they were excluded fromantiviral treatments until recently. Nowadays there isincontrovertible evidence in favor of treating these patients, and substitution therapy and active substanceuse are no longer contraindications for antiviral treatment. The viral clearance in PWID after HCV antiviraltreatment with interferon or pegylated interferon combined with ribavirin is comparable to the viral clearancein non-substance users. Furthermore, multidisciplinary approaches to delivering treatment to PWID are advised, and their treatment should be considered on anindividualized basis. To prevent the spread of HCV inthe PWID community, recent active PWID are eligiblefor treatment in combination with needle exchange programs and substitution therapy. As the rate of HCV reinfection is low after HCV antiviral treatment, there is no need to withhold HCV treatment due to concerns about reinfection alone. Despite the advances in treatment efficacies and data supporting their success, HCV assessment of PWID and initiation of antiviral treatment remains low. However, the proportion of PWID assessed and treated for HCV is increasing, which can be further enhanced by understanding the barriers to and facilitators of HCV care. Removing stigmatization and implementing peer support and group treatment strategies, in conjunction with greater involvement by nurse educators/practitioners, will promote greater treatment seeking and adherence by PWID. Moreover, screening can be facilitated by noninvasive methods for detecting HCV antibodies and assessing liver fibrosis stages. Recently, HCV clearance has become a major endpoint in the war against drugs for the Global Commission on Drug Policy. This review highlights the most recent evidence concerning HCV infection

Assessing phylogenetic relationships of Lycium samples using RAPD and entropy theory

Aim: To evaluate the phylogenetic relationships among related species of Lycium samples. Methods: Random amplified polymorphic DNA (RAPD) fingerprinting and lab-on-a-chip electrophoresis techniques were used to analyze the characteristics of Lycium species. Seven species and 3 varieties of Lycium were studied. Based on RAPD fingerprint data obtained from 11 primers, we proposed a new index, called dispersivity, using entropy theory and projection methods to depict the diversity of the DNA fingerprints. Results: Using the proposed dispersivity, primers were sorted and the dendrograms of the 7 species and 3 varieties of Lycium were constructed synthetically by merging primer information. Conclusion:Phylogenetic relationships among Lycium samples were constructed synthetically based on RAPD fingerprint data generated from 11 primers.

A modular-based Green Design Studio(GDS)platform has been developed in this study for fast and accurate performance analysis for early stage green building *** GDS platform aims to simplify the design and analysis process by embedding performance parameters into design elements in modules and employing near-real-time model for whole building performance simulation as well as by providing an easy-to-use and intuitive user interface to assist users without extensive knowledge on building *** platform consists of building modules as fundamental building blocks,performance predicting models,and a user interface for visualization and interactive *** the platform,a whole building is composed of modules organized in a hierarchical structure,including spaces,enclosures,service systems,sustainable resource systems and *** physics-based and data-driven models can be used to simulate the building performance and optimize building systems.A simplified physics-based model,the Resistance–Capacitance(RC)model,has been proposed as a generic simulation model for the flows of heat,air,moisture and pollutants,which is significantly faster than conventional simulation tools such as EnergyPlus,and hence more practical for use in real-time design interaction and optimization.A pilot case study is conducted to illustrate the modular-based design approach using a section of an office *** to conventional building performance analysis tools,the GDS platform can provide fast and reliable feedback on performance prediction for early *** modular approach makes it easier to modify the building design and evaluate the potentials and contributions of various green design features and technologies.

The Genetics of Speciation： Considering Early-Acting Isolation, Hybrid Evolution, and Epigenetic Mechanisms

Since Darwin＇s time, evolutionary biologists have sought to understand how species arise. The study of speciation remains a focus of evolutionary biology, and since the early 204 century, characterizing the genetics underlying this process has also become a central goal. Much research into the genetics of speciation has cen- tered on identifying the genes underlying reproductive isolating mechanisms, or ＇speciation genes＇（see Nosil and Schluter, 2011 for a discussion of this term）. With information about such genes available from many sys- tems, general questions about how speciation proceeds may be addressed： How many genes contribute to speciation？ Is reproductive isolation more commonly the result of divergence in protein-coding regions or regulatory regions？ Is reproductive isolation more fre- quently the result of natural selection or genetic drift？（Coyne and Orr, 2004; Nosil and Schluter, 2011）. Given recent advances in technology, such questions have be- come accessible in systems that were previously intrac- table for studying the genetics of speciation. Moreover, with this increase in the accessibility of genetic analyses for non-model systems, a broader set of questions about the genetics of speciation can now be addressed.

Alendronate and FTI-277 combination as a possible therapeutic approach for hepatocellular carcinoma:An in vitro study

Background： An important product of mevalonate pathway is downstream synthesis of isoprenoid unitsthat has long been implicated in development and progression of tumor. It has been speculated thatinhibition of protein prenylation might be therapeutically beneficial. The objective of current study wasto evaluate antitumor potential of a novel therapeutic combination of mevalonate pathway inhibitors, FTI-277 and alendronate. We also examined differentially expressed proteins in response to treatment usingproteomics approach.

Myelin repair and functional recovery mediated by neural cell transplantation in a mouse model of multiple sclerosis

Cellular therapies are becoming a major focus for the treatment of demyelinating diseases such as multiple sclerosis （MS）, therefore it is important to identify the most effective cell types that promote myelin repair. Several components contribute to the relative benefits of specific cell types including the overall efficacy of the cell therapy, the reproducibility of treatment, the mechanisms of action of distinct cell types and the ease of isolation and generation of therapeutic populations. A range of distinct cell populations promote functional recovery in animal models of MS including neural stem cells and mesenchymal stem cells derived from different tissues. Each of these cell populations has advantages and disadvantages and likely works through distinct mechanisms. The relevance of such mechanisms to myelin repair in the adult central nervous system is unclear since the therapeutic cells are generally derived from developing animals. Here we describe the isolation and characterization of a population of neural cells from the adult spinal cord that are characterized by the expression of the cell surface glycoprotein NG2. In functional studies, injection of adult NG2~ cells into mice with ongoing MOG3~_~5-induced experimental autoimmune encephalomyelitis （EAE） enhanced remyelination in the CNS while the number of CD3＋ T cells in areas of spinal cord demyelination was reduced approximately three-fold. In vivostudies indicated that in EAE, NG2＊ cells stimulated endogenous repair while in vitro they responded to signals in areas of induced inflammation by differentiating into oligodendrocytes. These results suggested that adult NG2~ cells represent a useful cell population for promoting neural repair in a variety of different conditions including demyelinating diseases such as MS.

Optimization of variables and assessment of in-vitro and in-vivo antihyperlipidemic activity of Eudragit RS nanoparticles containing simvastatin

Simvastatin, a BCS class II drug, is associated with poor aqueous solubility, first-pass metabolism and short half-life. In the present work, nanoparticles were prepared and evaluated. Optimization of formulation and process parameters was done through the use of independent and dependent variables. Preliminary studies were done to determine suitable range of the concentration of Eudragit polymer (10%–30%) and the ratio of drug to polymer (1:1 to 1:5) for the formation of nanoparticles by emulsification and a solvent evaporation technique. Results revealed that the mean size of nanoparticles was affected by stirring speed from 5000 RPM, 8000 RPM, and 12000 RPM. The results of increase in association efficiency and percent yield with increase in amount of drug from 100 mg, 150 mg, and 200 mg in selected range were observed. In-vitro release studies showed that two formulations possess highest initial burst and slow sustained drug release. There was 2.93-fold decrease in total cholesterol and 3.27-fold increases in triglyceride level during in-vivo study.
