Microbial Degradation of Organic contaminants in Streambed/Floodplain Sediments in Passaic River—New Jersey Area

This paper is intended to explore soil organic matter and carbon isotope fractionation at three locations of the Passaic River to determine if microbial degradation of organic contaminants in soil is correlated to the surrounding physical environment. Microbial degradation of organic contaminants is important for the detoxification of toxic substances thereby minimizing stagnation in the environment and accumulating in the food chain. Since organic contaminants are not easily dissolved in water, they will penetrate sediment and end up enriching the adjacent soil. The hypothesis that we are testing is microbial activity and carbon isotope fractionation will be greater in preserved soils than urban soils. The reason why this is expected to be the case is the expectation of higher microbial activity in preserved environments due to less exposure to pollutants, better soil structure, higher organic matter content, and more favorable conditions for microbial growth. This is contrasted with urban soils, which are impacted by pollutants and disturbances, potentially inhibiting microbial activity. We wish to collect soil samples adjacent to the Passaic River at a pristine location, Great Swamp Wildlife Refuge, a suburban location, Goffle Brook Park, Hawthorne NJ, and an urban location, Paterson NJ. These soil samples will be weighed for soil organic matter (SOM) and weighed for isotope ratio mass spectrometry (IRMS) to test organic carbon isotopes. High SOM and δ13c depletion activity indicate microbial growth based on the characteristics of the soil horizon rather than the location of the soil sample which results in degradation of organic compounds.

Effect of PEG Incorporation on Physicochemical and in vitro Degradation of PLLA/PDLLA Blends:Application in Biodegradable Implants

Polyethylene glycol(PEG)was added at different concentrations to the blend of poly(L-lactic acid)(PLLA)and poly(D,L-lactic acid)(PDLLA)to tailor the *** differential scanning calorimetry(DSc)measurement showed that all blends were miscible due to shifting a single glass transition temperature into a lower temperature for increasing PEG *** DSc,FTIR,and XRD results implied the crystallinity enhancement for PEG content until 8 wt%,then decreased at 12 wt%*** XRD result indicated the homo crystalline phase formation in all blends and no stereocomplex *** in vitro degradation study indicated that PEG content is proportional to the degradation *** highest weight loss after 28 days was achieved at 12 wt%*** FTIR analysis showed a structural evolution overview during hydrolytic degradation,*** and decreasing crystallinity during 14 days for the blend without and with PEG,*** conclusion,the PEG addition increased crystallinity and degradation rate of the PLLA/PDLLA mixture,but PEG higher amounts led to a decrease in crystallinity,and the weight loss was *** can be useful for tuning PLA-based biomaterials with the desired physicochemical properties and appropriate degradation rates for applications in drug delivery/tissue engineering.

Daily genetic profiling indicates JAK/STAT signaling promotes early hepatic stellate cell transdifferentiation

AIM: To identify signaling pathways and genes that initiate and commit hepatic stellate cells (HScs) to transdifferentiation. METHODS: Primary HScs were isolated from male Sprague-Dawley rats and cultured on plastic for 0-10 d. Gene expression was assessed daily (quiescent to day 10 culture-activation) by real time polymerase chain reaction and data clustered using AMADA software. The significance of JAK/STAT signaling to HSc transdifferentiation was determined by treating cells with a JAK2 inhibitor. RESULTS: Genetic cluster analyses, based on expression of these 21 genes, showed similar expression profiles on days 1-3, days 5 and 6, and days 7-10, while freshly isolated cells (day Q) and day 4 cells were genotypically distinct from any of the other days. Additionally, gene expression clustering revealed strong upregulation of interleukin-6, JAK2 and STAT3 mRNA in the early stages of activation. Inhibition of the JAK/STAT signaling pathway impeded the morphological transdifferentiation of HScs which correlated with decreased mRNA expression of several profibrotic genes including collagens, α-SMA, PDGFR and TGFβR. cONcLUSION: These data demonstrate unique clustered genetic profiles during the daily progression of HSc transdifferentiation and that JAK/STAT signaling may be critical in the early stages of transdifferentiation.

A comparative Study of the Hydroxyl Value and Iodine Value in Polyoxyl Stearyl Ether in United States Pharmacopeia Specifications

The iodine value (iodine number) and hydroxyl value are important analytical characteristics of fats and oils. The iodine (I2) required saturating the fatty acids present in 100 grams of the oil or fat. Iodine value is a measure of the total number of double bonds (-c=c-) present in fats and oils. Unsaturated compounds contain molecules with double and triple bonds which are very reactive towards iodine. The iodine value has been determined according to Hanus with iodine monobromide in glacial acetic acid, and then the amount of iodine remaining unreacted is determined by titration using sodium thiosulfate volumetric standard solution. The hydroxyl value is the amount of potassium hydroxide in milligrams that is equivalent to the hydroxyl amount of 1 gram of the sample (mg KOH/g sample). Poloxyl Stearyl Ether is a mixture of the monostearyl ethers of mixed polyethylene glycols. It may contain various amounts of free stearyl alcohol and some free polyethylene glycol. In this study, the iodine value and hydroxyl value have been determined by titration in polyoxyl stearyl ether. Iodine value 1.84 g of I2 absorbed/100g sample, and hydroxyl value 162.65 mg KOH/g sample have been found in poloxyl stearyl ether. The iodine value and hydroxyl value results met the United States Pharmacopeia specifications for Polyoxyl Stearyl Ether.

Occult Hepatitis c Virus Infection:A Review

Detection and Recuperation of Mental Fatigue

Mental fatigue is a complex state that results from prolonged cognitive activity. Symptoms of mental fatigue can include change in mood, motivation, and temporary deterioration of various cognitive functions involved in goal-directed behavior. Extensive research has been done to develop methods for recognizing physiological and psychophysiological signs of mental fatigue. This has allowed the development of many AI-based models to classify different levels of fatigue, using data extracted from eye-tracking device, EEG, or EcG. In this paper, we present an experimental protocol which aims to both generate/measure mental fatigue and provide effective strategies for recuperation via VR sessions paired with EEG and eye tracking devices. This paper first provides a comprehensive state-of-the-art of mental fatigue predictive factors, measurement methods, and recuperation strategies. Then the paper presents an experimental protocol resulting from the state-of-the-art to 1) generate and measure mental fatigue and 2) evaluate the effectiveness of virtual therapy for fatigue recuperation, using a virtual reality (VR) simulated environment. In our work, we successfully generated mental fatigue through completion of cognitive tasks in a virtual simulated environment. Participants showed significant decline in pupil diameter and theta/alpha score during the various cognitive tasks. We trained an RBF SVM classifier from Electroencephalogram (EEG) data classifying mental fatigue with 95% accuracy on the test set. Finally, our results show that the time allocated for virtual therapy did not improve pupil diameter in post-relaxation period. Further research on the impact of relaxation therapy on relaxation therapy should allocate time closer to the standard recovery time of 60 min.
