α,β-amyrin, a natural triterpenoid ameliorates L-arginine-induced acute pancreatitis in rats

AIM: To study the benef icial effects of triterpene α,β-amyrin and the underlying mechanisms in an experimental pancreatitis model. METHODS: Acute pancreatitis was induced in five groups of rats (n = 8) by L-arginine (2 × 2.5 g/kg, intraperitoneal, 1 h apart) and 1 h later, they received a single oral dose of α,β-amyrin (10, 30 and 100 mg/kg),methylprednisolone (30 mg/kg) and vehicle (3% Tween 80). A saline (0.9% NaCl) treated group served as a normal control. Efficacy was assessed at 24 h by determination of serum levels of amylase, lipase and proinflammatory cytokines [tumor necrosis factor (TNF)-α and interleukin (IL)-6], pancreatic myeloperoxidase (MPO) activity, lipid peroxidation [thiobarbituric acid reactive substances (TBARS)], nitrate/nitrite levels, and the wet weight/body weight ratio. Tissue histology and the immunoreactivity for TNF-α and inducible nitric oxide synthetase (iNOS) were performed. RESULTS: α,β-amyrin and methylprednisolone treatments significantly (P < 0.05) attenuated the L-arginine-induced increases in pancreatic wet weight/body weight ratio, and decreased the serum levels of amylase and lipase, and TNF-α and IL-6, as compared to the vehicle control. Also, pancreatic levels of MPO activity, TBARS, and nitrate/nitrite were signifi cantly lower. Histological f indings and TNF-α and iNOS immunostaining further confirmed the amelioration of pancreatic injury by α,β-amyrin. CONCLUSION: α,β-amyrin has the potential to combat acute pancreatitis by acting as an anti-in? ammatory and antioxidant agent.

Genetic Diversity of Uropathogenic Escherichia coli Isolated from Human T Lymphotropic Virus Type I (HTLV-I) Infected Individuals

Urinary tract infection (UTI) is a frequent pathology among HTLV-I+ individuals being capable of severely compromising the kidneys and bladder. Molecular characteristics of uropathogenic Escherichia coli (UPEC) from HTLV-I+ infected individuals are unknown. UPEC isolates from HTVL-I+ individuals, with and without clinical symptoms of myelopathy, were submitted to genetic typing seeking to infer bacterial diversity and potential virulence. 71 bacterial isolates were characterized according to random amplified polymorphic DNA and phylotypes. Phylotyping classified E. coli into four phylogenetic groups: A (18.3%), B1 (16.9%), B2 (39.4%), and D (25.3%) and 8 phylotypes according to the presence of the genetic sequences chuA, yjaA and the DNA fragment TSPE4.C2: ﹣﹣﹣ (5.6%) and ﹣+﹣ (12.6%) in phylogroup A, ﹣﹣+ (7.0%) and ﹣++ (9.8%) in B1, +++ (32.3%) and ++﹣ (7.0%) in B2, +﹣﹣ (15.4%) and +﹣+ (9.8%) in D. The B2 phylogroup was the most prevalent among HTLV﹣ associated myelopathy and asymptomatic individuals. RAPD-PCR typing revealed a high degree of bacterial polymorphism indicating a non-clonal origin. Genotypes were not found to be distributed according to clinical status or epidemiological features. Our results lead us to suggest that the neurological impairment in HTLV-I+ individuals can be a risk factor for urinary infections due E. coli which are caused by distinct bacterial lineages.

Combined hepatocholangiocarcinoma is a rare and unique form of primary hepatic neoplasm, expressing histopathological and phenotypic aspects of hepatocellularcarcinoma and cholangiocarcinoma in the same tumor. Diagnosis may be performed by imaging, showing typical features of both components. We present a case of a 55-year-old woman presenting with abdominal pain and a hepatic mass. The patient underwent surgery and combined hepatocholangiocarcinoma with stem cells features was confirmed on pathological analysis. There are no signs of recurrence to date. Combined hepatocholangiocarcinoma requires a preoperative diagnosis, since it is a unique entity with higher rates of local and lymph node recurrence, compared to isolated forms.

Assessment of the Risk of Violence in Portuguese Psychiatric Settings Using the Broset Violence Checklist

The risk of violence in psychiatric settings implies the assessment of dynamic predictors to adjust nursing interventions. So as to identify the pattern of aggressive behaviors, assess the dynamic predictors of violence in hospitalized patients, and analyze the predictive qualities of the Brcset Violence Checklist （BVC）, an exploratory/descriptive study was conducted in psychiatric wards in Coimbra, Portugal. The instruments used were： the staff observation aggression scale-revised （SOAS-R）, visual analogue scale （VAS）, and the BVC. For the period of a month, 64 patients with a mean age of 29 years, unemployed, and with psychotic disorders were observed. In this group, 13 people displayed 15 aggressive behaviors of moderate severity, which had consequences for nurses; they were triggered by the denial of something through verbal aggression and controlled by non-restrictive measures. The most common predictors of violence were irritability and boisterousness. It was also concluded that the BVC shows good predictive characteristics （sensitivity and specificity） of violence, thus, it may be considered as a useful and effective instrument to assess the risk of violence and, consequently, to adjust nursing interventions to prevent this phenomenon.

Effect of Environmental Factors on Rock-inhabiting Fungal Communities from Brazilian Soapstone Samples

The present study aims to evaluate the environmental effect on fungal community composition associated with biodeterioration occurring in stones （soapstone） at two distinct locations in Minas Gerais State, Brazil： Congonhas city and Sanctuary of Cara^a. Four collections of fungal communities over one year were obtained from both research sites from the soapstone block surfaces exposed for over two decades. The molecular diversity profile of the fungal community at the two localities was obtained by DGGE （Denaturing Gradient Gel Electrophoresis）, and the genomes of the most representative population were sequenced. DGGE showed the formation of two clusters with filamentous fungal communities. Sequencing of the most representative bands revealed the presence of fungi associated with the biodeterioration of soapstone. In addition, many of the identified species were associated with photobionts that could generate lichens, indicating that environmental characteristics affect the occurrence of filamentous fungi, which leads to biodeterioration of stones. Authors＇ study focused on an environmental variation of an extreme habitat for fungi associated with soapstone in the state of Minas Gerais, Brazil and identified the presence of interesting rock-inhabiting fungal communities including species related to lichens, which can accelerate the deterioration of stones by the production of organic acids.

Estimation of Pollutant Loads in Ardila Watershed Using the SWAT Model

Abstract： Excess of organic matter and nutrients in water promotes eutrophication process observed in the Ardila River. It was classified as much polluted being critical for Alqueva-Pedrogāo System. The aim of this study was to estimate the transported nutrients loads in a transboundary watershed using the SWAT （soil and water assessment tool） model and to determine the contribution of nutrients load in the entire watershed. Ardila watershed is about 3,711 km^2 extended from Spain （78%） to the eastern part of Portugal （22%）. It was discretized into 32 sub-basins using automated delineation routine, and 174 hydrologic response units. Monthly average meteorological data （from 1947 to 1998） were used to generate daily values through the weather generator Model incorporated in SWAT. Real daily precipitation （from 1931 to 2003） was introduced. The model was calibrated and verified for flow （from 1950 to 2000） and nutrients （from 1981 to 1999）. Model performance was evaluated using statistical parameters, such as NSE （Nash-Sutcliffe efficiency） and root mean square error （R2）. Calibration and verification flow results showed a satisfactory agreement between simulated and measured monthly date from 1962 to 1972 （NSE = 0.8; R^2 = 0.9）. The results showed that the most important diffuse pollution comes from the two the main tributary （Spain）. The estimated nitrogen and phosphorous loads contribution per year was respectively 72% and 59% in Spain and 28% and 41% in Portugal. The SWAT model was revealed to be a useful tool for an integrated water management approach that might be improved taking into count the WFD （water framework directive）.

The vaquejada race is a sport with animals which has the greatest socio-economic impact in Brazil and is responsible for generating an intense cultural and economic movement. However, this activity has been the subject of questions about well-being involving the environmental and mental comfort conditions offered to equine and cattle interactions. It is necessary to perform technical evaluations of the cattle involved on the basis of scientific study to better understand the processes of sport and, with this, promote their evolution on good welfare practices in the 21st century. Therefore, to test the hypothesis that the use of the official rules of the vaquejada where cattle use a protective harness on the tail protects the cattle’s tail from injuries in this region that compromise health. Before, during and after races, a study was developed that aimed to perform a general and radiological clinical evaluation in the proximal coccygeal region in cattle that ran in a modern cattle stall. Forty male bovines were evaluated (average: 2 year-old and 420 kg). Oxen were subjected to clinical and radiological examination before and after racing with official rules. Results of the clinical and radiological evaluations performed by veterinarians, did not observe clinical occurrences before and after the races in the 40 cattle evaluated. The increase in heart and respiratory rates was a reflection of temporary management stressors, as the animals returned to the feeding/rumination/interaction pattern after running in the rest pen. The radiographic findings in the tail, represented by the reduction of the intervertebral space and areas of subcutaneous radiolucency, suggest technical artifacts derived from

Galectin-3 expression favors metastasis in murine melanoma

Galectin-3 is a member of the lectin family that binds β-galactosides and plays an important role in several types of tumors. Melanoma is an invasive cancer responsible for 80% of deaths associated to skin cancers. There are some evidences that galectin-3 interacts with β-catenin, a molecule involved with Wnt signaling pathway. Here, we evaluate the role of galectin-3 intumor growth and metastasis, as well as its interaction with β-catenin. Murine melanoma cells (B16F10) were injected subcutaneously and intravenously in male C57BL/6 wild-type (WT) and galectin-3 knock-out (KO) mice. Tumor growth and lung melanoma colonies were assessed. The expression of galectin-3 and β-catenin was evaluated by immuno-histochemistry. We observed that tumor growth did not differ between the groups. However, to metastasis, the number of lung colonies in WT mice was significantly increased in comparison to that observed in KO mice. The cytoplasm expression of galectin-3 was observed in subcutaneous and metastatic tumors,w in both groups. We observed its nuclear expression in some of subcutaneous tumors of KO mice. The expression of β-catenin was detected in cell membrane of all subcutaneous tumors analyzed, whereas in the metastatic tumors we observed both cytoplasm and cell membrane staining. Altogether, our data suggest that galectin-3 favors the metastasis of melanoma cells and this process is not associated with β-catenin.

Detection of Contamination and Analysis of Vertical Transmission of BmNPV in Eggs and Moths of Bombyx mori

This study reports the molecular detection of Bombyx mori nucleopolyhedrovirus (BmNPV) in silkworm strains of the Universidade Estadual de Maringá Brazilian Germplasm Bank (UBGB). DNA extraction was carried out by using six Bombyx mori female moths of each strain, followed by PCR amplification. A pair of primers was designed based on a specific sequence of the baculovirus genome related to the BmNPV ORF 14. Another pair of primers was used to amplify the silkworm Actin A3 gene segment, which was used as positive control. Twenty gene pools were analyzed, and fifteen revealed a fragment of 443 base pairs (bp), which indicated the presence of the BmNPV. The frequency of contaminated moths was as following: 100% for silkworm strains M18-2, M12-2 and J1;83% for C25, C75 and C24 strains;66% for KR01;50% for M11-A;33% for AS3, B106, M8 and M11 and 16% for C211, E8 and Hindu strains. These are promising results for the identification of contaminated B. mori moths by BmNPV, which may prevent virus proliferation in subsequent generations. We also analyzed DNA samples extracted from B. mori eggs, but the results were not conclusive regarding the detection of the fragments of the expected size (443 bp). The difficulty in detecting BmNPV contamination in B. mori eggs may be due to the low concentration of virus in samples.
