TLR2 and TLR4 polymorphisms influence m RNA and protein expression in colorectal cancer

AIM: To evaluate the effect of promoter region polymorphisms of toll-like receptor(TLR)2-196 to-174 del and TLR4-1607T/C(rs10759932) on m RNA and protein expression in tumor tissue and of TLR4+896A/G(rs4986790) on colorectal cancer(CRC) ***: The TLR2-196 to-174 del polymorphism was investigated using allele-specific polymerase chain reaction(PCR) and the TLR4-1607T/C and TLR4+896A/G by PCR-restriction fragment length p o l y m o r p h i s m( R F L P). W e g e n o t y p e d 4 3 4 D N A samples from 194 CRC patients and 240 healthy individuals. The m RNA relative quantification(RQ) was performed in 40 tumor tissue samples by quantitative PCR Taq Man assay, using specific probes for TLR2 and TLR4 genes, and ACTB and GAPDH reference geneswere used as endogenous controls. Protein expression was analyzed by immunohistochemistry with specific primary ***: No association was found for TLR4-1607T/C and TLR4+896A/G by three statistical models(logadditive, dominant and recessive). However, based on dominant and log-additive models, the polymorphic variant TLR2-196 to-174 del was associated with increased CRC risk [dominant: odds ratio(OR) = 1.72, 95%CI: 1.03-2.89; P = 0.038 and log-additive: OR =1.59, 95%CI: 1.02-2.48; P = 0.039]. TLR2 m RNA expression was increased in tumor tissue(RQ = 2.36) when compared to adjacent normal tissue(RQ = 1; P < 0.0001), whereas the TLR4 m RNA showed a basal expression(RQ = 0.74 vs RQ = 1, P = 0.452). Immunohistochemistry analysis of TLR2 and TLR4 protein expression was concordant with the findings of m RNA expression. In addition, the TLR2-196 to-174 del variant carriers showed m RNA relative expression 2.19 times higher than wild-genotype carriers. The TLR2 protein expression was also higher for the TLR2-196 to-174 del variant carriers [117 ± 10 arbitrary unit(a.u.) vs 95 ± 4 a.u., P = 0.03]. The Direct Use of Post-Processing Wood Dust in Gas Turbines

Woody biomass is a widely-used and favourable material for energy production due to its carbon neutral status. Energy is generally derived either through direct combustion or gasification. The Irish forestry sector is forecasted to expand significantly in coming years, and so the opportunity exists for the bioenergy sector to take advantage of the material for which there will be no demand from current markets. A by-product of wood processing, wood dust is the cheapest form of wood material available to the bioenergy sector. Currently wood dust is primarily processed into wood pellets for energy generation. Research was conducted on post-processing birch wood dust;the calorific value and the Wobbe Index were determined for a number of wood particle sizes and wood dust concentrations. The Wobbe Index determined for the upper explosive concentration (4000 g/m3) falls within range of that of hydrogen gas, and wood dust-air mixtures of this concentration could therefore behave in a similar manner in a gas turbine. Due to its slightly lower HHV and higher particle density, however, alterations to the gas turbine would be necessary to accommodate wood dust to prevent abrasive damage to the turbine. As an unwanted by-product of wood processing the direct use of wood dust in a gas turbine for energy generation could therefore have economic and environmental benefits.

Mesenchymal stem cells(MSC)have pushed the field of stem cell-based therapies by inducing tissue regeneration,immunosuppression,and angiogenesis mainly through vesicles and soluble factors release(paracrine signaling).MSC-extracellular vesicles(MSC-EV)adaptable secretome and homing to injured sites allowed researchers to unlock a new era of cell-free based *** parallel,nanoparticles(NP)have been explored in contributing to transport and drug delivery systems,giving drugs desired physical-chemical properties to exploit cell ***,NPs can be quickly recognized by immune cells and cleared from *** this viewpoint,we explore how combining both therapeutic strategies can improve efficacy and circumvent limitations of both *** benefit from the potent MSC membrane composition,guiding chemotaxis to tumor sites,a very restricted ***-EV has low immunogenicity,high stability,long half-life and can explore tissue targeting ligands as a precise drug carry,even across biological *** properties promote enhanced targeted drug delivery that can be combined with NP,exploring biological membrane production through:*** cell therapy with NP-infused MSC;***-containing MSC-EV generated by NP-infused MSC;*** coating NP in MSC membrane(“MSC NanoGhosts”),allowing precise cargo definition without losing ***,nanotechnology combined with cell-based therapeutic resources can greatly improve targeted drug delivery,improving efficacy and opening a new venue of therapeutic possibilities.

This study was designed to determine the neuroprotective potentials of aqueous and methanol extracts from Heinsia crinita leaves in *** total phenol and flavonoid contents of the extracts were determined using colorimetric method while phenolic characterization of the leaf was analyzed via high performance liquid chromatography-diode array detector(HPLC-DAD).The effects of the extracts on Fe^2+-induced lipid peroxidation in rats’brain homogenate,monoamine oxidase(MAO),Na^+/K^+-ATPase,acetylcholinesterase(AChE)and butyrylcholinesterase(BChE)activities were also *** aqueous extract had higher total phenol and flavonoid contents than the methanol ***-DAD revealed that quercetin ellagic,chlorogenic and caffeic acids were the most abundant phenolic compounds in the *** aqueous extract had higher inhibitory effects on MAO,AChE and BChE activities while there was no significant difference between their Fe^2+-induced lipid peroxidation inhibitory ***,both extracts stimulated Na^+/K^+-ATPase activity;however,methanol extract had higher stimulatory *** neuroprotective properties of *** leaves could be associated with its inhibitory effects on Fe2+-induced lipid peroxidation and modulation of MAO,Na^+/K^+-ATPase,AChE,and BChE ***,*** leaves could be used as a functional food and dietary intervention for the management of some neurodegenerative ***,the aqueous extracts exhibited better neuroprotective properties.

The role of the vascularized bone marrow component as a continuous source of donor-derived hematopoietic stem cells that facilitate tolerance induction of vascularized composite allografts is not completely *** this study,vascularized composite tissue allograft transplantation outcomes between recipients receiving either conventional bone marrow transplantation(CBMT)or vascularized bone marrow(VBM)transplantation from Balb/c(H2d)to C57BL/6(H2b)mice were *** high-or low-dose CBMT(1.5×10^(8)or 3×10^(7)bone marrow cells,respectively)was *** addition,recipients were treated with costimulation blockade(1 mg anti-CD154 and 0.5 mg CTLA4Ig on postoperative days 0 and 2,respectively)and short-term rapamycin(3 mg/kg/day for the first posttransplant week and then every other day for another 3 weeks).Similar to high-dose conventional bone marrow transplantation,5/6 animals in the vascularized bone marrow group demonstrated long-term allograft survival(>120 days).In contrast,significantly shorter median survival was noted in the low-dose CBMT group(~64 days).Consistently high chimerism levels were observed in the VBM transplantation ***,low levels of circulating CD4^(+)and CD8^(+)T cells and a higher ratio of Treg to Teff cells were maintained in VBM transplantation and high-dose CBMT recipients(>30 days)but not in low-dose VBM transplant ***-specific hyporesponsiveness was shown in tolerant recipients in *** of the vascularized bone marrow component after secondary donor-specific skin transplantation did not affect either primary allograft or secondary skin graft survival.

Acaricidal activity of Derris floribunda essential oil and its main constituent

Objective: To evaluate the acaricidal activity of the essential oil obtained from roots of Derris floribunda(D. floribunda)(Miq.) Benth, and its main constituent nerolidol against the Mexican mite Tetranychus mexicanus(T. mexicanus)(Mc Gregor).Methods: The essential oil from the roots of D. floribunda collected in the Amazon region(Brazil) was obtained by hydrodistillation. Its chemical composition was determined by GC–MS analysis. The acaricidal activities of this essential oil and nerolidol,were evaluated by recording the number of dead females(mortality) and eggs(fertility).Results: The essential oil showed sesquiterpenes as major volatile components. Nerolidol, the main component, represented 68.5% of the total composition of the essential oil. D. floribunda essential oil and nerolidol showed acaricidal activity, with LC_(50) of9.61 mg/m L air and 9.2 mg/m L air, respectively, over a 72 h period. In addition, both the essential oil and nerolidol significantly reduced the fecundity of T. ***: Due to the economic importance of T. mexicanus and the lack of new pesticides, our data are very promising in the search for efficient and safer acaricidal products. Furthermore, this is the first report about the chemical composition and bioactivity of the essential oil of the Amazon plant species D. floribunda.
