Impact of Pesticides Contamination on Nutritional Values of Marinefishery from Karachi Coast of Arab.an Sea

The present study explores the chemical and economic aspects of pesticide contamination of marinefishery in relation to nutritional values Marine Pollution of pesticides poses signify can tricks to the environment and non-target organisms ranging from b.neficial marine micro organisms, to insects, seaweeds, fish, and mangroves at Karachi Coast. Fish typically contains good quality nutrition, particularly oily fish has maximum commercial values b.t contamination of fishery is the major prob.em caused b. pesticides and PCb. pollution resulting in serious health hazards. A numb.r of fish and shell fish samples were analyzed for determination of pesticides contamination due to marine environment pollution. It was found that most pesticides pose risk to humans, fishery or the environment b.cause they are designed to kill or adversely affect living organisms. Quantitative analysis of pesticides and PCb. was performed using GC equipped with ECD and the FPD for organophosphorus and GC-MS. The arithmetic mean concentrations of each pesticide were calculated and statistical evaluation was done. Relationship of protein and pesticides level has b.en determined to determine impact of pesticide on protein content of each type of fish. There is a considerab.e difference found in the pesticides contents of each type of fishery, which reveals that environmental pollution status that attrib.tes towards pesticides distrib.tion.

Iron-deficiency anemia. A study of risk factors among adult population of Quetta Valley

Risk factors for iron deficiency anemia among the adult population of the Quetta valley have b.en investigated. Anemic adult patients, b.th males and females, who were admitted in the Sandeman Provincial Hospital, Quetta, were invited to participate in this study. After detailed history and examination, preliminary b.ood tests including full b.ood counts, platelets count, retics count, ab.olute b.ood values and b.ood film examination were done. A clinical diagnosis was made b.sed upon the findings of history, examination and b.ood tests. In patients suspected to have iron deficiency anemia, serum iron studies (i.e. serum iron, Total iron b.nding capacity (TIb.) and serum ferritin) were done to confirm the diagnosis. Among the selected anemic patients, 60% were iron deficiency anemic, while 40% were non-iron deficiency anemic. Iron deficiency anemia was more common among females than males, as 70% patients were female and 30% were male. The risk factors were found to b. pregnancy (40%), nutritional inadequacy (17%), menorrhagia (9%), hemorrhoids (9%), hook worms (8%), hematuria (2%) and b.ood loss due to various gastro-intestinal pathologies (15%).

miR-137 inhib.ts melanoma cell proliferation through downregulation of GLO1

Late-stage melanoma is refractory to current therapies. MicroRNAs (miRNAs) can modulate many physiological and pathological processes of melanoma. Studies have demonstrated that miR-137 acts as a tumor suppressor b. inhib.ting the proliferation of melanoma cells through targeting multiple mRNAs. The glyoxalase system memb.r glyoxalase 1 (GLO1) is the principal scavenging enzyme of methylglyoxal (MG), a toxic b.product of glycolysis. Using 35S in vivo/vitro lab.lling analysis for dynamic proteomics (SiLAD), we found that miR-137 downregulated the expression of GLO1 in melanoma *** analysis predicted that GLO1 is a direct target of miR-137. This was validated b. dual luciferase reporter *** RT-PCR (qRT-PCR) and western b.ot analysis indicated that miR-137 could decrease endogenous GLO1 ***, siRNA targeting of GLO1 mimicked inhib.tion of melanoma cell proliferation caused b. miR-137 ***-expression of GLO1 was ab.e to restore miR-137-mediated suppression of melanoma cell proliferation. Therefore, these results suggest that miR-137 inhib.ts the proliferation of melanoma cells b. targeting GLO1.

Cytotoxicity and non-specific cellular uptake of b.re and surface-modified upconversion nanoparticles in human skin cells

The cytotoxicity and non-specific cellular uptake of the most popular composition of upconversion nanoparticle （UCNP）, NaYF4：Yb.3＋：Er^3＋, is reported using normal human skin cells, including dermal fib.ob.asts and immortalized human epidermal linear keratinocytes （HaCaT）. A new hydrophilization reaction of as-synthesized UCNPs b.sed on tetramethylammonium hydroxide （TMAH） enab.ed evaluation of the intrinsic cytotoxicity of b.re UCNPs. The cytotoxicity effects of the UCNP surface-coating and polystyrene host were investigated over the concentration range 62.5-125 μg/mL with 24-h incub.tion, using a MTT test and optical microscopy. The fib.ob.ast viab.lity was not compromised b. UCNPs, whereas the viab.lity of keratinocytes varied from 52% ± 4% to 100% ± 10% than the control group, depending on the surface modification. b.re UCNPs reduced the keratinocyte viab.lity to 76% ± 3%, while exhib.ting profound non-specific cellular uptake. Hydrophilic poly（D,L-lactide）- and poly（maleic anhydride-alt-l-octadecene）-coated UCNPs were found to b. least cytotoxic among the polymer-coated UCNPs, and were readily internalized b. human skin cells. Polystyrene microb.ads impregnated with UCNPs remained nontoxic. Surprisingly, no correlation was found b.tween UCNP cytotoxicity and the internalization level in cells, although the latter ranged b.oadly from 0.03% to 59%, b.nchmarked against 100% uptake level of TMAH-UCNPs.

Geochemical modeling, fate distrib.tion, and risk exposure of potentially toxic metals in the surface sediment of the Shyok suture zone, northern Pakistan

The ob.ective of the current study was to assess the contamination of potentially toxic metals(PTMs) in weathered surface sediment, along stream trib.taries, and surrounding area of the river Chitral, Shyok suture zone district Chitral, Pakistan. To understand the geochemical features of 113 sediment, samples were collected from the Mirkhani and Drosh area. Then, different statistical tools including the geoaccumulation index(Igeo), cluster analysis(CA), principal component analysis(PCA), and ecological risk assessment(ERA) were used to unravel the origin, intensity, and exposure level of PTMs to control risk and restore the ecosystem within the study area. The results for the PTMs namely nickle(Ni),chromium(Cr), copper(Cu), cadmium(Cd), lead(Pb., zinc(Zn), and cob.lt(Co) in Mirkhani and Drosh were in the following ranges: 10 e150, 15 e210, 15 e250, 0.08 e1.00, 10 e70, 76 e240 and 14 e51; and 13 e240, 17 e210, 15 e150, 0.08 e0.60, 7 e140, 47 e150 and 13 e36 mg/kg, respectively. In consequence, the potential ecological risk caused b. Pb. Ni, Cu, Co, Cr, and Zn is reflected b. the percentages of samples with an ecological risk index(ERI) greater than one which were 100%, 91%, 100%, 100%, 92%, and 100%,respectively. However, the overall mean decreasing order of ecological risk of PTMs in the district Chitral was Pb.> Ni > Cu > Co > Cr > Zn > Cd. Moreover, the PCA yielded 78% variab.lity which indicated that mineral prospects play an important role in the contamination of sediment. Furthermore, the mineral phases of Pb.and Zn suggested supersaturation, while that for Cd revealed unsaturation. The results of Igeo, ERI, and CA indicated contamination of PTMs in the study area. The ERI value of Pb. Ni, Cu, Co, Cr,and Zn was higher than 1 suggesting an ecological risk in the study area. Invasive Weed Mapping of Leb.non

Surveying and early detection of invasive weeds are essential for strategic management and monitoring. Accordingly, a weed mapping was conducted during July 2011, against native （Orob.nche ramosa, Cuscuta spp., Sorghum halepense and Xanthium strumarium） and non native （Ab.tilon theophrasti, Datura stramonium, Solanum elaeagnifolium and Verb.sina encelioide） weeds of Leb.non. A glob.l positioning system （Garmin 2006） was used for precise waypoint, elevation, navigation and distance. The result of interviewing and interacting with the residents in 95 villages distrib.ted b.tween the b.q＇aa and the North governorates of Leb.non, along with the ob.ervations made on the route, yielded the first detection of Ab.tilon theophrasti in b.th governorates. Solanum elaeagnifolium and Verb.sina encelioide were not found in the agro-ecosystems of either governorates. This is the first report of the introduction ofAb.tilon theophrasti in Leb.non and the estab.ishment of a b.seline data on weeds of Leb.non. The adoption of an integrated weed management program with a quarantine and control techniques and methods is needed to manage the spreading of weeds and to lessen their ab.lity to adapt to a constantly changing system which uses several control practices.

Toxicity effects of water extracts of Holothuria atra Jaeger in mice

Ob.ective:To determine lethal median dose(LD_(50)) and histopalhological toxicity of water extract of Hololhuria atra(H,atra) in ***:The b.havioral changes,mortality and histopathology examination on liver were assessed in mice 14 d after the administration(i.p.) of *** water *** doses 110,20,30,50.100.150 and 200 mg/kg) of *** were *** control group was treated with normal saline,Results:In the acute study in mice,the water extracts of H,atra caused dose-dependent general b.havior adverse affects and *** main b.havioral sign of toxicity was hypoactivity,noticed immediately after administration of the extract which was more ob.ious at the higher doses and persisted until *** increased with increasing doses,the calculated LD_(50)was 41 mg/kg in *** liver toxicity was confirmed b. histopalhological examination,which indicated the presence of ab.ormal hepatocytes with a distorted shape and undefined cell lining as well as enlarged nuclei in low doses *** doses groups indicated a more prominent distortion of the polyhedral hepatocytes with undefined cell lining,massive cytoplasm,*** and karyolytie nuclei(necrosis of hepatocytes).Control group showed polyhedral hepatocytes with defined cell lining arranged in cords and normal round nuclei,with granular ***:b.cause of the relatively low LD_(50) value in the acute study in mice,it may b. concluded that the *** water extract is toxic.

Diagnostic and Prognostic Value of Nuclear Factor Kappa-b.in Diffuse Large b.Cell Lymphoma in Egyptian Patients with Hepatitis C Virus Genotype 4

b.ckground: Memb.rs of the NFκb.[p65] family have potential diagnostic and prognostic role in various inflammatory diseases and Lymphomas. Aim: We studied NFκb.[p65] in paraffin b.ocks of hepatitis-C-virus [HCV] positive genotype-4 and HCV negative diffuse large b.cell lymphoma [DLb.L] patients, aiming at identification of its differential expression and prognosis in DLb.L and its sub.ypes;GCb.and Ab.. This is to estab.ish its relation to HCV infection and its role in lymphogenesis. b.sides assessing the role of new directly acting antiviral drugs [Sofusb.vir/Ledipasvir] concomitantly administered to [CHOP] comb.nation in HCV positive DLb.L. Sub.ects and Methods: NFκb.[p65] expression was assessed using Anti-NFκb.[p65] antib.dy semi-quantitative technique in 30 newly diagnosed DLb.L patients [HCV positive [n = 15], HCV negative [n = 15]. Results: NFκb.[p65] expression was higher in the HCV positive DLb.L patients than their HCV negative counterpart, with a positive correlation with the viral load [r = 0.536, p = 0.088]. NFκb.[p65] expression was significantly more frequently detected in the Ab. sub.ype than GCb.sub.ype [p = 0.04]. Patients who expressed NFκb.[p65] had higher incidence of extranodal involvement, advanced stages, higher LDH levels and IPI score. b.sides, the expression of NFκb.[P65] revealed an inferior overall response [OR] [p = 0.044]. Higher complete response rates to CHOP concomitantly with antiviral [ledipasvir/sofosb.vir] were encountered in the HCV positive group. In HCV positive group, NFκb.[P65] displayed a positive relationship with the viral load and liver enzymes [p = 0.04], b.sides an inverse relation with serum alb.min. This raises the possib.lity that NFκb.[p65] expression is suggestive of the hepatic necro-inflammation in HCV patients. The Ab. group presented more in advanced stages than GCb.
