Anti-Candida and anti-Cryptococcus evaluation of 15 non-alkaloidal compounds from Pterogyne nitens

Objective: To evaluate anti-Candida and anti-Cryptococcus activities of 15 nonalkaloidal compounds from Pterogyne nitens Tulasne(Leguminosae), a South American medicinal ***: Compounds were submitted to antifungal assays, using microdilution method described by Clinical and Laboratory Standards Institute document, with minor modifications. Five species of Candida and two species of Cryptococcus, including clinical isolates were screened. Antifungal activity was expressed by minimum inhibitory concentration(MIC). Amphotericin B and fluconazole were used as standard antifungal ***: Among tested compounds, six substances presented fungal growth inhibition(MIC < 31.2 mg/m L) [three flavone derivatives(1–3), a glycosylated flavonol derivative(5)and two phenolic acids(10 and 12)]. Sorbifolin(1), exhibited potent antifungal activity,demonstrating MIC value of 3.90 mg/m L against Candida glabrata ATCC 90030, Cryptococcus gattii 118 and fluconazole-resistant clinical isolate of Cryptococcus neoformans var. grubii. Pedalin(2) and nitensoside B(3), two glycosylated flavone derivatives, were active against Cryptococcus neoformans ATCC 90012(MIC = 7.80 mg/m L).Conclusions: Flavone derivatives from Pterogyne nitens can serve as prototypes for the design and development of innovative anti-Candida and anti-Cryptococcus hits.

Technical Evaluation of Hybrid Clones of Corymbia spp. to Produce Market Pulp

This study presents hybrid clones of Corymbia *** by Aperam BioEnergia as potential substitutes for Eucalyptus wood in the pulp *** biomass of Eucalyptus *** compared with that of Corymbia *** performing modified kraft pulping and basic density *** were made by analyzing their respective mean annual increments of cellulose(MAI_(cel))and specific wood consumption(SWC),estimated using a kappa number of(19±1).The results showed that one of the hybrid clones(Corymbia citriodora×Corymbia torelliana,ID 4)had higher basic wood density,higher screened yield,better MAI_(cel#k19),and lower SWC than the other samples,reaching values of 0.608 t/m^(3),54.1%,24.6 ADt/ha/year(i.e.,air dry ton/hectare/year),and 2.74 m^(3)/ADt,***,clone ID 4 had the highest MAI_(cel)/SWC ratio score(8.98).Given its high forest productivity and low industrial cost compared with other samples,as well as its advantages over Eucalyptus ***,we concluded that clone ID 4 has great potential as an alternative biomass for pulp production.

Impact of infection in hospitalized cirrhotic patients in a university hospital

Background: Infection increases the morbidity and mortality in liver cirrhosis patients. The aim of this study was to investigate the impact of infection related to survival and risk factors for death in adult patients with liver cirrhosis in a university hospital. Methods: In a retrospective cohort study of Brazilian hospitalized cirrhotic patients, medical records data were analysed, and all patients who have had one or more confirmed bacterial infection during admission were se-ected for the study. Also, some data as biochemical investigation, Child score, MELD estimation, and evolution and death event were included. Statistical analysis: chi-square, Fisher and Mann-Whitney tests were used. Uni and multivariate analysis were performed, according to Cox regression model. The significant statistical level was p 2.5 mg/dl had increased the risk of death of 4.1, 3.2 and 3.2, respectively. Conclusion: Bacterial infections in hospitalized cirrhotic patients deserve special care, mainly spontaneous bacterial peritonitis, and also patients whose hiponatremia, upper gastrointestinal bleeding, high levels of cre-atinine and MELD high score are found.

Hemodynamic Fetal Response to Maternal Isometric Exercise in Pregnant Patients with Hypertension

Objective: to evaluate the maternal-fetal hemodynamic response with Doppler in pregnant women with chronic arterial hypertension and preeclampsia submitted to controlled isometric activity. Methods: experimental study comparing 50 healthy and 26 hypertensive and 24 preeclamptic pregnant women, from 26 to 36 weeks of gestational age, submitted to isometric contraction with handgrip dynamometer. Maternal hemodynamic parameters (systolic and diastolic blood pressure;heart rate;uterine arteries Doppler) and fetal (heart rate;umbilical artery, middle cerebral and venous duct Doppler) were evaluated before, during and post-isometry. Results: in preeclampsia were observed higher values of blood pressure and uterine artery indexes in all times;middle cerebral artery indexes in the pre and post-isometry;and of maternal heart rate post-isometry. In hypertensive women, systolic blood pressure is increased all the times, with indexes of the right uterine and middle cerebral arteries higher in pre-isometry;middle cerebral and umbilical arteries greater during isometry;and maternal heart rate and umbilical artery indexes bigger after isometry. Conclusion: blood pressure is higher in preeclamptic and hypertension women. The right uterine artery has more resistance in preeclampsia, with a significant decrease in pre to isometry in hypertensive and preeclamptic women;and increased in contraction to post-isometry in healthy and hypertensive women. The left uterine artery increases resistance post-isometry in all groups. The fetal hemodynamic parameters did not show significant differences when comparing the before, during and post-isometry.

Background: Studies on physical activity during pregnancy and its impact on mother and fetus are still limited. International protocols consider only aerobic exercise and fail to provide information about other modalities such as isometric exercise. Isometric exercise promotes cardiorespiratory resistance and muscle strengthening, but it is rarely tested on pregnant women because it increases maternal blood pressure and can subsequently affect placental circulation. Objectives: To assess the fetal response by use of Doppler study in diabetes pregnant women submitted to isometric exercise. Methods: A cross-sectional experimental study was conducted on 25 diabetes pregnant women with gestational age between 26 and 36 weeks. The patients were submitted to isometric handgrip, and data were collected from the mother (blood pressure, heart rate and Doppler velocimetry for the uterine arteries) and from the fetus (heart rate, Doppler velocimetry for the umbilical artery, middle cerebral artery and ductus venosus). All variables were collected before, during and after the isometric handgrip. Poly(3-hydroxybutyrate)(PHB)is a biothechnological and biodegradable thermoplastic polymer from the polyhydroxyalkanoates(PHAs)family,whose chain regularity,high molecular weight,and physical and mechanical properties comparable to polypropylene(PP)are characteristics that have made PHB a prominent commercial ***,its susceptibility to thermal degradation and hydrolysis has limited many *** address the challenges associated with processing,a random copolymer of 95.86 mol%3-hydroxybutyrate and 4.14 mol%3-hydroxyvalerate(referred as PHB)was compounded without(neat PHB)and with 0.25,0.5,1,and 1.5 wt%of chain extender(Joncryl■ADR 4368),consisting of multifunctional epoxy groups,and melt mixed in an internal mixer at 170℃,50 rpm,and residence times of 5,10,and 15 *** effect of chain extender concentration and residence time on the effectiveness of Joncryl■ADR 4368 to recover the pristine PHB properties were investigated through torque rheometry,melt flow index(MFI),differential scanning calorimetry(DSC),tensile testing,and thermogravimetric *** results indicated that an increase in residence time strongly influenced the prevalence of thermomechanical degradation reactions over chain extension reactions of PHB during processing in the molten *** residence times favored chain extension,while longer times favored chain *** trend led to no enhancement of PHB mechanical properties processed for 10 and 15 min,regardless of additive content *** flow index,thermogravimetric analysis and tensile strength exhibited an asymptotic behavior with the increase in additive concentration,exhibiting a noteworthy improvement in tensile strength and temperature at the highest rate of thermal degradation(TMAX),and a decrease in melt flow index of neat PHB with only 0.25 wt%of additive mixed for 5 *** results found in this work allow additional data for processing PHB by manipulating the ratios of multifunctional chain

Feasibility and safety of autologous bone marrow mononuclear cell transplantation in patients with advanced chronic liver disease

AIM： To evaluate the safety and feasibility of bone marrow cell （BMC） transplantation in patients with chronic liver disease on the waiting list for liver transplantation. METHODS： Ten patients （eight males） with chronic liver disease were enrolled to receive infusion of autologous bone marrow-derived cells. Seven patients were classified as Child-Pugh B and three as Child-Pugh C. Baseline assessment included complete clinical and laboratory evaluation and abdominal MRI. Approximately 50 mL of bone marrow aspirate was prepared by centrifugation in a ficoll-hypaque gradient. At least of 100 millions of mononuclear-enriched BMCs were infused into the hepatic artery using the routine technique for arterial chemoembolization for liver tumors. Patients were followed up for adverse events up to 4 mo. RESULTS： The median age of the patients was 52 years （range 24-70 years）. All patients were discharged 48 h after BMC infusion. Two patients complained ofmild pain at the bone marrow needle puncture site. No other complications or specific side effects related to the procedure were observed. Bilirubin levels were lower at 1 （2.19 ± 0.9） and 4 mo （2.10 ± 1.0） after cell transplantation that baseline levels （238 ± 1.2）. Albumin levels 4 mo after BMC infusion （3.73 ± 0.5） were higher than baseline levels （3.47 ± 0.5）. International normalized ratio （INR） decreased from 1.48 （SD = 0.23） to 1.43 （SD = 0.23） one month after cell transplantation. CONCLUSION： BMC infusion into hepatic artery of patients with advanced chronic liver disease is safe and feasible. In addition, a decrease in mean serum bilirubin and INR levels and an increase in albumin levels are observed. Our data warrant further studies in order to evaluate the effect of BMC transplantation in patients with advanced chronic liver disease.

Clinical Analysis of a Coaxial Dilator Set Attached to Needle Puncture for Percutaneous Nephrolithotomy

Introduction: Percutaneous approach to the kidney is a very useful alternative in renal surgery which minimizes the morbidity of a variety of procedures. Objectives: To evaluate the efficacy, practicity of a reusable system of metallic coaxial dilator coupled to a special puncture needle to perform lumbar puncture and dilation of the percutaneous tract. Methods: A randomized experimental study was carried out involving 50 individuals. These patients were randomized as follows: Group 1 had the procedure performed with the conventional disposable Amplatz set traditionally used for this procedure. Patients in Group 2 were operated utilizing the new reusable coaxial set specially designed for this purpose. The following parameters were measured to compare the 2 groups: sex Corporeal Mass Index (CMI), Pre and postoperative hematocrit and hemoglobin. Time interval between the puncture, dilation and access to the renal pelvis and insertion of the nephroscope were also analyzed. Results: There was no statistical difference between the 2 groups concerning hematocrit and hemoglobin changes when comparing prewith postoperative period. Patients in Group 2 required a significant lower time between puncture and final access to the upper collecting system. Conclusion: The new coaxial dilator set showed to be as safe as the conventional Amplatz set with the advantage of reusability and decrease of tract dilation time.

Background: Dengue is a Neglected tropical disease (NTDs) with high incidence in Brazil. This disease is caused by Dengue virus and is transmitted by Aedes aegypti mosquito. The search for new approaches for controlling of this disease is the subject of numerous studies. The aaNAT is a key enzyme in the metabolism of A. aegypti and is crucial in the sclerotization process, as well as regulation of circadian rhythm and inactivation of neurotransmitters. Computational techniques applied to studies of biological systems become an effective weapon in the mapping and management of 3D data structures, giving direction and guidance of potential ligands that can form stable complexes with targets of interest, using a Molecular Docking approach. The present study was conducted by a virtual screening, followed by docking calculations, in order to find molecules that could inhibit aaNAT. In this study, we used available compounds in SAM database (Bioinformatics and Medicinal Chemistry Laboratory—Southwest Bahia State University, Jequié-Bahia, Brazil), PubChem and ZINC. Results: The result of dockings with selected ligands showed good energy affinities, presenting potential inhibitory interactions with the enzyme active site. Conclusions: The Coa-S-acetyl-tryptamine and 3-indoleacriloil-coenzyme-A showed the same binding energies -8.9 Kcal/Mol and were described as possible inhibitors of aaNAT.

Protective Effect of Taurine in the Induction of Genotoxicity by Mutagenic Drugs

Genotoxicity is a toxic action of several drugs, such as anti-neoplastic and nitro-related compounds. TAU （Taurine, 2-aminoetano sulfonic acid） is an important and potent antioxidant amino acid involved in the maintenance of homeostasis in many systems like leukocytes, skeletal muscle, the heart, the kidney, the liver, the bowel, the brain and the retina. The aim of this work was evaluated by the protective effect of TAU co-administered with mutagenic drugs such as benznidazole, cyclophosphamide, hydroxyurea, metronidazole, and nitrofurazone in an in vivo model using a micronucleus assay, evaluatation for the ability of different substances to promote DNA injury. The results showed that TAU decreased genotoxic activity of all drugs suggesting that the protective action of TAU supplementation can be beneficial to reduce genotoxicity of drugs. In this context, we propose one possible mechanism for how TAU blocks the chromosomal aberrations caused by the action of genotoxic substances.
