X-ray microscopy is an essential imaging method in many scientific fields,which can be extended to three-dimensional(3D)using tomography.Recently,metal halide perovskite(MHP)nanomaterials have become a promising candidate for X-ray scintillators,due to their high light yield,high spatial resolution,and easy fabrication.Tomography requires many projections and therefore scintillators with excellent stability.This is challenging for MHPs,which often suffer from fast degradation under X-ray irradiation and ambient conditions.Here,we demonstrate that MHP scintillators of CsPbBr3 nanowires(diameter:60 nm,length:5–9μm)grown in anodized aluminum oxide(CsPbBr3 NW/AAO)have sufficient stability for X-ray micro-tomography.A tomogram was taken with a Cu X-ray source over 41 h(dose 4.2 Gyair).During this period the scintillator brightness fluctuated less than 5%,which enabled a successful reconstruction.A long-term study with 2 weeks of continuous X-ray exposure(37.5 Gyair)showed less than 14%fluctuations in brightness and no long-term degradation,despite variations in the ambient relative humidity from 7.4%RH to 34.2%RH.The resolution was stable at(180±20)lp·mm−1,i.e.,about 2.8 micron.This demonstrates that CsPbBr3 NW/AAO scintillators are promising candidates for high resolution X-ray imaging detectors.

AIM:To investigate the expression and potential prognostic role of vascular endothelial growth factor(VEGF) and endoglin in gastroenteropancreatic neuroendocrine tumors(GEP-NETs) . METHODS:Microvessel density(MVD) in GEP-NETs was evaluated using endoglin and CD31 immunohistochemistry.In addition,tissue levels of endoglin and VEGF were determined in homogenates by ELISA. RESULTS:Endoglin was highly expressed on tumor endothelial cells.CD31 MVD in GEP-NETs was significantly higher compared to endoglin MVD(P<0.01) .Two-tofour-fold higher tissue levels of endoglin and VEGF were seen in tumors compared to associated normal tissue. This increased endoglin tissue expression in tumors was significantly related to tumor size(P<0.01) ,presence of metastases(P=0.04) ,and a more advanced tumor stage(P=0.02) ,whereas expression of VEGF was not. CONCLUSION:We suggest that endoglin is a potential marker to indicate and predict metastases,which might be useful in the post-resection therapeutic approach of patients with GEP-NETs.

High level HIV-1 DNA concentrations in brain tissues differentiate patients with post-HAART AIDS dementia complex or cardiovascular disease from those with AIDS

Highly active antiretroviral treatment(HAART) has had a significant impact on survival of individuals with acquired immunodeficiency syndrome(AIDS);however,with the longer life-span of patients with AIDS,there is increasing prevalence of AIDS dementia complex(ADC) and other non-AIDS-defining illness,and cardiovascular diseases(CVD) are also common.The influence of these varied disease processes on HIV-1 DNA concentration in brain tissues has not been thoroughly assessed in the post-HAART era.The purpose of the current study is to clarify the impacts of ADC and other complications of HIV disease on the viral load in the brains in AIDS patients with post-HARRT.We examined autopsy specimens from the brains of thirteen patients who died from complications of AIDS with quantitative polymerase chain reaction(QPCR).All but one patient had received HAART prior to death since 1995.Two patients died with severe CVD,multiple cerebrovascular atherosclerosis(CVA) throughout the brain and five patients died with ADC.Six patients had no ADC/CVA.A QPCR was used to measure the presence of HIV-1 DNA in six brain tissues(meninges,frontal grey matter,frontal white matter,temporal subcortex,cerebellum and basal ganglia).In the post-HARRT era,for non-ADC/CVA patients,HIV-1 DNA concentration in brain tissues was statistically higher than that in patients with ADC.In a new finding,two patients who suffered from severe CVD,especially CVA,also had high concentrations of HIV-1 in brain compartments not showing ADC related changes.To our knowledge,this is the first report of a relationship between the CVA and HIV-1 viral burden in brain.The current observations suggest that HAART-resistant HIV reservoirs may survive within ADC lesions of the brain as well as the macrophage rich atherosclerosis,which needs to be confirmed by more AIDS cases with CVA.

Objective: To investigate the expression of the receptors for vasoactive intestinal peptide (VIP) and secretin in colon cancer. Methods: This study visualized and characterized the receptors for VIP and secretin in the sequence of human tumor-free colon, adenoma, carcinoma, liver metastasis using storage phosphor autoradiography. Results: Receptors for VIP and secretin were demonstrated in tumor-free colon and colon tumors. A decrease in affinity of VIP receptors was shown in the colonic liver metastasis (Kd = 3.30 nmol) when compared with tumor-free colon (Kd = 0.82 nmol). An up-regulation of receptors for secretin was found in colonic liver metastases. Conclusions: VIP and secretin were both expressed on normal colon tissues. Binding of VIP decreased while secretin increased in colonic liver metastasis. A down-regulation of receptors for VIP in colonic liver metastases may helpful to understand the migration of colon cancer.

AIM:To study the (functional) relevance of single nucleotide polymorphisms (SNPs) in genes encoding matrix metalloproteinases (MMP)-1,-2,-3,-9,tissue inhibitors of metalloproteinases (TIMP)-1,-2 and tumor necrosis factor (TNF)-α in the etiopathogenesis of inflammatory bowel diseases (IBD),that may enhance susceptibility and/or disease severity. METHODS:Genomic DNA from 134 Crohn's disease (CD),111 ulcerative colitis (UC) patients and 248 control subjects was isolated from resected intestinal tissue or blood. Allelic composition at SNP loci was determined by PCR-RFLP or tetra primer ARMS PCR. RESULTS:The TIMP-1 genotype TT in women and T in men at SNP +372 T/C was found to increase CD susceptibility (39% vs 23.8%,P=0.018 and 67.9% vs 51.6%,P=0.055,respectively),while women with this genotype were less prone to development of fistulae during follow-up (41.4% vs 68.3%,P=0.025). Male IBD or CD patients carrying the TIMP-1 +372 T-allele expressed lower levels of TIMP-1 in surgically resected macroscopically inflamed tissue (0.065 < P < 0.01). The 5T5T genotype at MMP-3 SNP -1613 5T/6T increased the chance of stenotic complications in CD during follow-up (91.2% vs 71.8%,P = 0.022) but seemed to protect against colonic involvement of this disease at first endoscopic/radiologic examination (35.3% vs 59.5%,P=0.017). CONCLUSION:Allelic composition at the examinedSNPs in genes coding for TIMP-1 and MMP-3 affect CD susceptibility and/or phenotype,i.e.,fistulizing disease,stricture pathogenesis and first disease localisation. These findings reinforce the important role of these proteins in IBD.

Retinal Vascular Tortuosity in Hospitalized Patients with Type 2 Diabetes and Diabetic Retinopathy in China

Context: Diabetic patients are at high risk for microvascular complications of disease such as diabetic retinopathy (DR) and diabetic neuropathy. Imaging the retinal micro-vasculature offers a chance to measure quantitatively the microvascular changes in diabetic patients’ onset and progression. However, the relation between retinal biomarkers and diabetic risk factors is unclear in Chinese hospitalized type 2 diabetic patients. Aims: To examine the associations of retinal vascular tortuosity with risk factors of type 2 diabetes and diabetic retinopathy in Chinese hospitalized patients. Patients and Methods: Our cross-sectional study includes 504 participants with type 2 diabetes hospitalized in the department of endocrinology in Shengjing hospital, Shenyang, China. Patients’ socio-demographic, clinical and biological information was retrieved from their ID card, the interview, and the local hospital information system. Retinal photographs were taken by laser-scanning ophthalmoscopy of both eyes and checked if gradable for analysis. The weighted mean and standard deviation of tortuosity were calculated from the retinal photographs using a novel robust and fully automatic quantitative method. DR was assessed from the retinal photographs by the ophthalmologist according to the modified Airlie House classification into No DR and Any DR in our current study. Data were analyzed by SPSS Version 22 with Student’s t test, Mann-Whitney U test, Chi-square test, and linear regression. Results: For the total participants (504) in this study, the weighted mean and standard deviation (SD) of tortuosity was 12.05 × 10?3 (SD: 1.66 × 10?3) and 24.31 × 10?3 (SD: 3.69 × 10?3), respectively. 386 (76.6%) patients were diagnosed with DR who had older age, long duration of diabetes, higher brachial ankle pulse wave velocity (baPWV), higher systolic blood pressure (SBP), higher diastolic blood pressure (DBP), and higher retinal vascular tortuosity values. 高效抗逆转录病毒治疗(highly active antiretroviral treatment,HAART)在延长获得性免疫缺陷综合征(acquired immunodeficiency syndrome,AIDS)患者的生存方面具有重要作用,然而随着HIV感染者寿命的延长,AIDS痴呆综合征(AIDS dementiacomplex,ADC),其他非AIDS相关疾病,心血管病(cardiovascular disease,CVD)也比较常见.这些各种疾病对脑组织中HIV-1DNA的影响在HAART后还未完全搞清楚.本文的研究目的是澄清HAART后ADC以及其他HIV感染的并发症对AIDS病人脑组织中病毒载量的影响.本实验用定量PCR研究了13例AIDS病人脑组织尸检标本,这些病人死于AIDS并发症,除1例病人外,其他12例病人均从1995年开始接受HAART治疗,直到死亡.2例死于严重的CVD,脑组织中广泛的动脉粥样硬化(cerebrovascular atherosclerosis,CVA).5例AIDS病人死于ADC,6例AIDS人既无ADC,也无CVA.用定量PCR检测了病人脑组织6个部位(脑膜、前叶灰质、前叶白质、颞叶皮层下部、小脑和基底节)HIV-1DNA的含量并对其进行了比较.HAART后,与非ADC/CVA的AIDS病人相比,ADC的AIDS病人脑组织中HIV-1DNA含量明显升高.一个新的发现是,2例患有CVD,特别是CVA的病人,脑组织中,也同样检出高浓度的HIV-1DNA,这些病人并没有ADC的症状,这是首次报道CVA与脑组织中HIV-1病毒载量有关.研究表明,HAATR抗性的HIV病毒储存库可能存在于ACD患者脑组织的损伤部位及富含巨噬细胞的动脉粥样硬化斑块中,仍需要大量的CVA病例研究来证实后者.
