Single and double layer centrifugation improve the quality of cryopreserved bovine sperm from poor quality ejaculates

Background： Density gradient centrifugation was reported as a technique of semen preparation in assisted reproductive techniques in humans and animals. This technique was found to be efficient in improving semen quality after harmful techniques such as cryopreservation. Recently a modified technique, single layer centrifugation,was proposed as a technique providing a large amount of high quality spermatozoa, and this treatment was performed before conservation. Single layer centrifugation has been studied prevalently in stallions and in boars,but limited data were available for bulls. Occasionally bulls are known to experience a transient reduction in semen quality, thus techniques that allow improvement in semen quality could be applied in this context. The aim of this study was the evaluation of single layer and double layer centrifugation by the use of iodixanol, compared with conventional centrifugation and non-centrifuged semen, on the sperm characteristics during the cryopreservation process in bulls with normal and poor semen ***： Single layer centrifugation and double layer centrifugation both significantly increased the percentage of normal spermatozoa and decreased the percentage of non-sperm cells in poor quality samples, while both were ineffective in those of normal quality. Sperm characteristics in poor quality samples increased after single layer centrifugation and double layer centrifugation, reaching values similar to those recorded in normal samples, and this trend is maintained after equilibration and after cryopreservation. Pulse wave Doppler ultrasound of testicular arteries and their relationship with semen characteristics in healthy bulls

Background: Semen evaluation is used to estimate the testicular function. In bulls, the spermatozoa present in the ejaculate are the result of a process that begun more than 2 mo earlier, bequeathing a delayed depiction of the actual function of the testis. Since testis vascularization might be critical for the gonad function, selected pulse wave Doppler ultrasound parameters were assessed in this study, for instance the peak systolic velocity, the end diastolic velocity and the resistive index of the testicular artery along the spermatic cord, the marginal portion of the testicular artery and the intratesticular branches of the testicular artery both in healthy adult and young *** between these parameters and characteristics of semen that was collected numerous times, before and after the Doppler ultrasound ***: The peak systolic velocity and the end diastolic velocity measured in the testicular artery along the spermatic cord(supratesticular artery – SA) were variable among the bulls and within individual bulls, likely due to the convoluted course of the vessel. The resistive index was found highly repeatable in the same bull. A reduction in the resistive index was found between the supratesticular artery and the marginal portion of the testicular artery(P < 0.01), and between the marginal portion of the testicular artery and the intratesticular branches of the testicular artery(P < 0.05). No differences were recorded for the pulse wave Doppler ultrasound parameters in young bulls compared with adults. A significant correlation was found between the resistive index of the marginal portion of the testicular artery and total sperm in the ejaculate(r = 0.516, P < 0.05), the immature sperm(r = 0.462, P < 0.05), the teratoid sperm(r = 0.375, P < 0.05), and the "Dag defect" sperm(r = 0.389, P < 0.05). A Scalable Synthesis of Multiple Models of Geo Big Data Interpretation

The paper proposes a scalable fuzzy approach for mapping the status of the environment integrating several distinct models exploiting geo big data. The process is structured into two phases: the first one can exploit products yielded by distinct models of remote sensing image interpretation defined in the scientific literature, and knowledge of domain experts, possibly ill-defined, for computing partial evidence of a phenomenon. The second phase integrates the partial evidence maps through a learning mechanism exploiting ground truth to compute a synthetic Environmental Status Indicator (ESI) map. The proposal resembles an ensemble approach with the difference that the aggregation is not necessarily consensual but can model a distinct decision attitude in between pessimistic and optimistic. It is scalable and can be implemented in a distributed processing framework, so as to make feasible ESI mapping in near real time to support land monitoring. It is exemplified to map the presence of standing water areas, indicator of water resources, agro-practices or natural hazard from remote sensing by considering different models.

Enterolithiasis or formation of gastrointestinal concretions is an uncommon medical condition that develops in the setting of intestinal stasis in the presence of the intestinal diverticula, surgical enteroanastomoses, blind pouches, afferent loops, incarcerated hernias, small intestinal tumors, intestinal kinking from intra-abdominal adhesions, and stenosing or stricturing Crohn’s disease and intestinal tuberculosis. Enterolithiasis is classified into primary and secondary types. Its prevalence ranges from 0.3% to 10% in selected populations. Proximal primary enteroliths are composed of choleic acid salts and distal enteroliths are calcified. Clinical presentation includes abdominal pains, distention, nausea, and vomiting of occasionally sudden but often fluctuating subacute nature which occurs as a result of the enterolith tumbling through the bowel lumen. Thorough history and physical exam coupled with radiologic imaging helps establish a diagnosis in a patient at risk. Complications include bowel obstruction, direct pressure injury to the intestinal mucosa, intestinal gangrene, intussusceptions, afferent loop syndrome, diverticulitis, iron deficiency anemia, gastrointestinal hemorrhage, and perforation. Mortality of primary enterolithiasis may reach 3% and secondary enterolithiasis 8%. Risk factors include poorly conditioned patients with significant obstruction and delay in diagnosis. Treatment relies on timely recognition of the disease and endoscopic or surgical intervention. With advents in new technology, improved outcome is expected for patients with enterolithiasis.

Asymmetric dimethylarginine as a mediator of vascular dysfunction in cirrhosis

Cirrhosis is associated with marked abnormalities in the circulatory function that involve a reduction in systemic vascular resistance. An important cause of this vasodilatation is the increased production or activity of nitric oxide(NO) in the splanchnic circulation. During portal hypertension and cirrhosis an increased endothelial NO synthase(e NOS) activity is demonstrated in splanchnic vessels. In contrast, the activity of e NOS in the cirrhotic liver is decreased, which suggests a different regulation of e NOS in the liver and in the splanchnic vessels. Asymmetric dimethylarginine(ADMA) is an endogenous NO inhibitor and higher plasma levels of ADMA are related to increased cardiovascular risk in both the general population and among patients with cirrhosis. It has been demonstrated that the liver is a key player in the metabolism of ADMA. This observation was further supported by investigations in human patients, showing a close correlation between ADMA plasma levels and the degree of hepatic dysfunction. ADMA is degraded to citrulline and dimethylamine by dimethylarginine dimethylaminohydrolases(DDAHs). DDAHs are expressed as type 1 and 2 isoforms and are widely distributed in various organs and tissues, including the liver. In this review, we discuss experimental and clinical data that document the effects of dimethylarginines on vascular function in cirrhosis. Our increasing understanding of the routes of synthesis and metabolism of methylarginines is beginning to provide insights into novel mechanisms of liver disease and allowing us to identify potential therapeutic opportunities.

Refining Linked Data with Games with a Purpose

With the rise of linked data and knowledge graphs,the need becomes compelling to find suitable solutions to increase the coverage and correctness of data sets,to add missing knowledge and to identify and remove *** approaches-mostly relying on machine learning and natural language processing techniques-have been proposed to address this refinement goal;they usually need a partial gold standard,i.e.,some“ground truth”to train automatic *** standards are manually constructed,either by involving domain experts or by adopting crowdsourcing and human computation *** this paper,we present an open source software framework to build Games with a Purpose for linked data refinement,i.e.,Web applications to crowdsource partial ground truth,by motivating user participation through fun *** detail the impact of this new resource by explaining the specific data linking“purposes”supported by the framework(creation,ranking and validation of links)and by defining the respective crowdsourcing tasks to achieve those *** also introduce our approach for incremental truth inference over the contributions provided by players of Games with a Purpose(also abbreviated as GWAP):we motivate the need for such a method with the specificity of GWAP *** crowdsourcing;we explain and formalize the proposed process,explain its positive consequences and illustrate the results of an experimental comparison with state-of-the-art *** show this resource’s versatility,we describe a set of diverse applications that we built on top of it;to demonstrate its reusability and extensibility potential,we provide references to detailed documentation,including an entire tutorial which in a few hours guides new adopters to customize and adapt the framework to a new use case.

SIRENE: A Spatial Data Infrastructure to Enhance Communities' Resilience to Disaster-Related Emergency

Planning in advance to prepare for and respond to a natural hazard-induced disaster-related emergency is a key action that allows decision makers to mitigate unexpected impacts and potential damage. To further this aim, a collaborative, modular, and information and communications technology-based Spatial Data Infrastructure(SDI)called SIRENE—Sistema Informativo per la Preparazione e la Risposta alle Emergenze(Information System for Emergency Preparedness and Response) is designed and implemented to access and share, over the Internet, relevant multisource and distributed geospatial data to support decision makers in reducing disaster risks. SIRENE flexibly searches and retrieves strategic information from local and/or remote repositories to cope with different emergency phases. The system collects, queries, and analyzes geographic information provided voluntarily by observers directly in the field(volunteered geographic information(VGI) reports) to identify potentially critical environmental conditions. SIRENE can visualize and cross-validate institutional and research-based data against VGI reports,as well as provide disaster managers with a decision support system able to suggest the mode and timing of intervention, before and in the aftermath of different types of emergencies, on the basis of the available information and in agreement with the laws in force at the national andregional levels. Testing installations of SIRENE have been deployed in 18 hilly or mountain municipalities(12 located in the Italian Central Alps of northern Italy, and six in the Umbria region of central Italy), which have been affected by natural hazard-induced disasters over the past years(landslides, debris flows, floods, and wildfire) and experienced significant social and economic losses.

Effects of elevated atmospheric CO2 and nitrogen fertilization on nitrogen cycling in experimental riparian wetlands

Studies on the relationship between plant nitrogen content and soil nitrogen reduction under elevated CO2 conditions and with different nitrogen additions in wetland ecosystems are lacking. This study was meant to assess the effects of elevated CO2 concentrations and inorganic nitrogen additions on soil and plant nitrogen cycling. A cultured riparian wetland, alligator weeds, and two duplicated open top chambers （OTCs） with ambient （380μmol/mol） and elevated （700 μmol/mol） CO2 concentrations at low （4 mg/L） and high （6 mg/L） nitrogen fertilization levels were used. The total plant biomass increased by 30.77% and 31.37% at low and high nitrogen fertilization levels, respectively, under elevated CO2 conditions. Plant nitrogen content decreased by 6.54% and 8.86% at low and high nitrogen fertilization levels, respectively. The coefficient of determination （R2） of soil nitrogen contents ranged from 0.81 to 0.96. Under elevated CO2 conditions, plants utilized the assimilated inorganic nitrogen （from the soil） for growth and other internal physiological transformations, which might explain the reduction in plant nitrogen content. A reduction in soil dissolved inorganic nitrogen （DIN） under elevated CO2 conditions might have also caused the reduction in plant nitrogen content. Reduced plant and soil nitrogen contents are to be expected due to the potential exhaustive use of inorganic nitrogen by soil microorganisms even before it can be made available to the soil and plants. The results from this study provide important information to help policy makers make informed decisions on sustainable management of wetlands. Larger-scale field work is recommended in future research.

In the context of escalating global energy demands,urban areas,specifically the building sector,contribute to the largest energy consumption,with urban overheating exacerbating this *** urban modelling for heat-mitigation and reduction of energy demand is crucial steps towards a sustainable built-environment,complementing onsite energy generation in the design and development of Net-zero Energy(NZE)Settlement,especially in the context of Australian weather *** a significant gap in existing literature,this study offers empirical analysis on the climate and energy efficacy of integrated heat mitigation strategies applied in 14 neighbourhood typologies located in Sydney,*** the application of cool materials on roads,pavements,and rooftops,alongside urban vegetation enhancement,the analysis demonstrates scenario effectiveness on heat mitigation that leads to reduce ambient temperature and energy demands along with CO_(2)emissions within the *** building arrangement,built-area ratio,building height,and locations,ENVI-met and CitySim are utilized to assess the heat-mitigation and the energy demand of neighbourhoods,*** indicate that mitigation measures can lead up to a 2.71℃reduction in ambient temperature and over 25%reduction in Cooling Degree Hours,with a 34.34%reduction in cooling energy demand and overall energy savings of up to 12.49%.In addition,the annual energy-saving yields a CO_(2)reduction of approximately 141.12 tonnes,where additional vegetation further amplifies these reductions by enhancing CO_(2)*** study showcases the pathway towards achieving NZE goals in climates similar to that of Australia,highlighting significant benefits in heat-mitigation,environmental impact,and energy-savings.
