c-Fos overexpression increases the proliferation of human hepatocytes by stabilizing nuclear cyclin D1

AIM： To investigate the effect of stable c-Fos over- expression on immortalized human hepatocyte （IHH） proliferation. METHODS： IHHs stably transfected with c-Fos （IHH- Fos） or an empty vector （IHH-c） were grown in me- dium supplemented with 1% serum or stimulated with 10% serum. cell proliferation was assessed by cell counts, 3H-thymidine uptake and flow cytometry analyses. The levels of cell cycle regulatory proteins （cyclin DI, E, A） cyclin dependent kinases （cdk） cdk2, cdk4, cdk6, and their inhibitors p15, p16, p21, p27, total and phosphorylated GSK-3β and epidermal growth factor receptor （EGF-R） were assayed by Western blotting. Analysis of O/c/in D1 mRNA levels was performed by reverse transcription-polymerase chain reaction and real-time polymerase chain reaction （PcR） analysis. Stability of cyclin DI was studied by cycloheximide blockade experiments. RESULTS： Stable c-Fos overexpression increased cell proliferation under low serum conditions and resulted in a two-fold increase in [3H]-thymidine incorpora- tion following serum addition. cell cycle analysis by flow cytometry showed that c-Fos accelerated the cell cycle kinetics. Following serum stimulation, cyclin D1 was more abundantly expressed in c-Fos overexpress- ing cells. cyclin D1 accumulation did not result from increased transcriptional activation, but from nuclear stabilization. Overexpression of c-Fos correlated with higher nuclear levels of inactive phosphorylated GSK- 3β, a kinase involved in cyclin D1 degradation and higher levels of EGF-R mRNA, and EGF-R protein com- pared to IHH-c both in serum starved, and in serum stimulated cells. Abrogation of EGF-R signalling in IHH- Fos by treatment with AG1478, a specific EGF-R tyro- sine kinase inhibitor, prevented the phosphorylation of GSK-3β induced by serum stimulation and decreased cyclin D1 stability in the nucleus. cONcLUSION： Our results clearly indicate a positive role for c-Fos in cell cycle regulation in hepatocytes. A Three-Dimensional Performance Measurement Model for cloud computing

New paradigms for processing and storing data such as cloud computing require new approaches for the measurement of cloud service performance. To establish a Service Level Agreement (SLA) between a cloud service provider and its customers, the cloud services and their service level objectives need to be identified. An additional challenge in the performance measurement of cloud services is the lack of models that integrate the different perspectives of providers, maintainers and customers within the same model in order to define the concepts commonly used in cloud SLA contracts. This work proposes a three-dimensional Performance Measurement Model for cloud computing (P2M2c-3D) which consolidates performance measurement from the perspectives of providers, maintainers and customers for the different types of cloud services.

cellulose nanocrystals （cNcs）, a unique and promising natural material extracted from native cellulose, have attracted considerable attention owing to their physical properties and special surface chemistry. This review focuses on chemical conjugation strategies that can be used for preparation of ？uorescent-molecule labeled cNcs and the development of biomaterials. Furthermore, their application in the detection of metal ions and future development prospects are discussed. We hope to provide a clear view of the strategies for surface fluorescent modifcation of cNcs and their application in detection of metal ions.

A Local Binary Pattern-Based Method for color and Multicomponent Texture Analysis

Local Binary Patterns (LBPs) have been highly used in texture classification for their robustness, their ease of implementation and their low computational cost. Initially designed to deal with gray level images, several methods based on them in the literature have been proposed for images having more than one spectral band. To achieve it, whether assumption using color information or combining spectral band two by two was done. Those methods use micro structures as texture features. In this paper, our goal was to design texture features which are relevant to color and multicomponent texture analysis without any assumption. Based on methods designed for gray scale images, we find the combination of micro and macro structures efficient for multispectral texture analysis. The experimentations were carried out on color images from Outex databases and multicomponent images from red blood cells captured using a multispectral microscope equipped with 13 LEDs ranging from 375 nm to 940 nm. In all achieved experimentations, our proposal presents the best classification scores compared to common multicomponent LBP methods. 99.81%, 100.00%, 99.07% and 97.67% are maximum scores obtained with our strategy respective

Intra-Diverticular Bladder Tumor about a New Observation in the Urology Department of chu cocody—Abidjan, côte d’Ivoire

This case report is about a 61 years old woman who consulted for hematuria, the cystoscopy performed revealed an intra-diverticular bladder tumor. After tumor resection, an anatomopathological exam revealed the urothelial carcinoma pT1. A few months later she presented with a local reccurence of bladder tumor. We proposed her anterior pelvectomy but she refused it and she was lost to follow-up. The aim of this study is to show the anatomopathological and evolutionary characteristics and the therapeutic difficulties of IDBT. These tumors are in the majority of cases infiltrating from the outset with a high potential for recurrence after transurethral resection of bladder (TURB). The standard treatments are TURB and total cystectomy.

Desorption of Methylene Blue Adsorbed on Activated carbon from cocoa Pod Shell

Environmental protection has become a concern for the world. For this reason, the objective of this work is to remove methylene blue adsorbed on activated carbon. The coal used comes from cocoa pod shells. Before pyrolysis, the shells were ground, sieved and impregnated with orthophosphoric acid. Before desorption, the activated carbons were initially saturated with MB. These saturated coals were brought into contact with a sodium chloride (Nacl) solution and then stirred. The evolution of the resorbed MB concentration was monitored by spectrophotometry. The desorption tests showed a remarkable elimination from the first 10 minutes. The desorption kinetics comprises two phases: a rapid kinetics between 0 and 30 minutes and a slow kinetics between 30 and 60 minutes. The desorption of the dye reaches a concentration aqual to 0.84 mg/l at pH = 4 at temperature = 80°c. For modeling, the coefficient of the Langmuir II model is greater than or equal to O.9893. The model of Langmuir III is less than or equal to 0.9373. The Freundlich model coefficient is 0.9842 or less. The desorption is thefore carried out on energy-homogeneous adsorption sites and without any interaction between the adsorbed cations of the dye. Experimental parameters such as pH, temperature and concentration of sodium chloride (Nacl) solution influence the desorption of MB. And the model of Langmuir II describes well the process of desorption of the MB.

A Semi-Vectorial Hybrid Morphological Segmentation of Multicomponent Images Based on Multithreshold Analysis of Multidimensional compact Histogram

In this work, we propose an original approach of semi-vectorial hybrid morphological segmentation for multicomponent images or multidimensional data by analyzing compact multidimensional histograms based on different orders. Its principle consists first of segment marginally each component of the multicomponent image into different numbers of classes fixed at K. The segmentation of each component of the image uses a scalar segmentation strategy by histogram analysis;we mainly count the methods by searching for peaks or modes of the histogram and those based on a multi-thresholding of the histogram. It is the latter that we have used in this paper, it relies particularly on the multi-thresholding method of OTSU. Then, in the case where i) each component of the image admits exactly K classes, K vector thresholds are constructed by an optimal pairing of which each component of the vector thresholds are those resulting from the marginal segmentations. In addition, the multidimensional compact histogram of the multicomponent image is computed and the attribute tuples or ‘colors’ of the histogram are ordered relative to the threshold vectors to produce (K + 1) intervals in the partial order giving rise to a segmentation of the multidimensional histogram into K classes. The remaining colors of the histogram are assigned to the closest class relative to their center of gravity. ii) In the contrary case, a vectorial spatial matching between the classes of the scalar components of the image is produced to obtain an over-segmentation, then an interclass fusion is performed to obtain a maximum of K classes. Indeed, the relevance of our segmentation method has been highlighted in relation to other methods, such as K-means, using unsupervised and supervised quantitative segmentation evaluation criteria. So the robustness of our method relatively to noise has been tested.

Evaluation of Dosimetric Performance and Global Uncertainty of the Harshaw 6600 Plus System Used to Staff Monitoring in côte d’Ivoire

The objective of this work is to check the dosimetric performances of the TLD-100 as stated by the manufacturer as well as the technical standards of radiation protection. The purpose of the performance audit is to assess the inhomogeneity of TLD sensitivity, repeatability and reproducibility, linearity, energy dependence, angular dependence, and fading. All tests were performed under the conditions of ambient temperature and relative humidity recommended by the manufacturer. We began the study by calibrating the Harshaw 6600 Plus, and checking its performance. The TLD-100 performance verification results were all acceptable and in accordance with the manufacturer’s advertised values and the radiation protection technical standards. However the performance of the TLD-100 that we have evaluated may have some limitations;these limits, which are sources of uncertainty, have been taken into account in this work by evaluating the overall uncertainty of the Hp (10) dose in the uncertainty range 9.45% to 15.80% by simple formulas. The TLD-100 personal dosimeters and the 6600 Plus reader system indicate that the calculated values of the overall uncertainty Hp (10) are well below the allowable values of 21% to 42% suggested for personal dosimetry services. The obtained data encourage the use of the system for the routine evaluation of the external exposure of workers under ionizing radiation in our laboratory.

Growth and Freeze-Drying Optimization of Bifidobacterium crudilactis

Bifidobacterium crudilactis FR62/b/3 belongs to a new population of bifidobacteria isolated from raw milk and raw milk cheese. The objective of this work was to study the large scale culture of the strain and its stability in a dry formulation. Growth rate of Bifidobacterium crudilactis FR62/b/3 was optimal at a pH of 5.0 and a temperature of 37°c. At a temperature growth of 33°c and a pH of 5.0, the stationary phase was reached after 22 h, the viable cell number and the mean dry biomass concentration were respectively of 8.3 × 109 cFU/mL and of 2.1 g/L. Resistance of Bifidobacterium crudilactis FR62/b/3 to freeze-drying and effect of a variety of cryoprotectants to maintain the viability were also evaluated. Sorbitol was the most suitable cryoprotectant for freeze-drying as well as storage whereas sucrose and monosodium glutamate were only efficient during storage.
