Doxycycline induces bone repair and changes in Wnt signalling

Doxycycline （DOX） exhibits anti-inflammatory and MMP inhibitory properties. The objectives of this study were to evaluate the effects of DOX on alveolar bone repair. Controls （CTL） and DOX-treated （10 and 25 mg. kg- 1） molars were extracted, and rats were killed 7 or 14 days later. The maxillae were processed and subjected to histological.and immunohistochemical assays. Hematoxylin-eosin staining （7th day） revealed inflammation in the CTL group that was partly reversed after DOX treatment. On the 14th day, the CTL group exhibited bone neoformation, conjunctive tissue, re-epithelization and the absence of inflammatory infiltrate. DOX-treated groups exhibited complete re-epithelization, tissue remodelling and almost no inflammation. Picrosirius red staining in the DOXlO group （7th and 14th days） revealed an increased percentage of type I and III collagen fibres compared with the CTL and DOX25 groups. The DOXlO and DOX25 groups exhibited increases in osteoblasts on the 7th and 14th days. However, there were fewer osteoclasts in the DOXlO and DOX25 groups on the 7th and 14th days. Wnt-lOb- immunopositive cells increased by 130% and 150% on the 7th and 14th days, respectively, in DOX-treated groups compared with the CTL group. On the 7th day, Dickkopf （Dkk）-I immunostaining was decreased by 63% and 46% in the DOXlO and DOX25 groups, respectively. On the 14th day, 69% and 42% decreases in immunopositive cells were observed in the DOXlO and DOX25 groups, respectively, compared with the CTL group. By increasing osteoblasts, decreasing osteoclasts, activating Wnt lOb and neutralising Dkk, DOX is a potential candidate for bone repair in periodontal diseases.

Pseudomelanosis duodeni (PD) is a rare dark speckled appearance of the duodenum associated with gastrointestinal bleeding,hypertension,chronic heart failure,chronic renal failure and consumption of different *** report four cases of PD associated with chronic renal failure admitted to the gastroenterology outpatient unit due to epigastric pain,nausea,melena and progressive reduction of hemoglobin *** endoscopy revealed erosions in the esophagus and stomach,with no active bleeding at the *** addition,the duodenal mucosa presented marked signs of melanosis;later confirmed by histopathological *** though PD is usually regarded as a benign condition,its pathogenesis and clinical significance is yet to be defined.

Aims Natural vegetation plays an important role in global carbon cycling and ***,the Cerrado(Brazilian savannah)is considered a carbon sink because of its intrinsic *** aim was to evaluate how the aboveground biomass and biodiversity relationship change between three Cerrado remnants with different protection status:a‘control area’(Legal Reserve area),a protected area(PA)and a non-protected area(Non-PA).Methods all three studied fragments are situated in northern Minas Gerais state,*** estimated the aboveground carbon stocks based on the forest *** also measured three dimensions of biodiversity metrics for each plot:functional trait dominance,taxonomic diversity and functional *** following functional traits were evaluated for the species:wood density,maximum diameter and seed *** carried out generalized linear models seeking to evaluate how carbon stocks,community-weighted mean(CWM)trait val.es,species richness and diversity,and functional diversity indices differ among the *** Findings The Cerrado areas without protection status had lower carbon stocks,species richness,species diversity,functional richness and functional dispersion,whereas both PA and Non-PA had lower CWM maximum diameter and seed size compared with the Legal Reserve control *** linear models showed that carbon stocks,species and functional richness metrics were correlated within and across sites,and thus,species richness could serve as a good proxy for functional richness and carbon *** carbon stocks were positively driven by species richness and CWM maximum diameter,while they were negatively driven by functional *** richness,species diversity and CWM seed size appeared in the set of best models,but with no significant direct effect on carbon ***,we concluded that absence of protection in the Cerrado areas decreases both species richness and carbon stocks.

Background:In Cerrado humid areas,veredas are considered as hygrophilous communities,marked by the presence of the Mauritia flexuosa palm tree emerging in the wetter *** in veredas ecosystems due to drought and changes in the water table,which represent important areas for biodiversity maintenance in the Brazilian cerrado,can lead to the emergence of new landscapes and *** that the veredas are changed in species composition due to anthropogenic factors,in this study,we evaluated the flora composition and described the current vegetation profile of two veredas under different disturbances levels that are located in the north of Minas Gerais State,***:altogether,2,268 individuals of 91 species belonging to 36 families were *** results demonstrated that the studied veredas were floristically different in terms of the arboreous-shrubby species composition,richness,and *** the hygrophilous forest formations and open areas in the two veredas present typical species from these environments(same species with high importance val.es),the drought in the water table caused an expansion of Cerrado species in the open areas and in the hygrophilous ***,in the hygrophilous forest there was an increase in the abundance of species that occur in less saturated ***:Our results demonstrate that the studied veredas are changing in species composition due to anthropogenic factors which leads to the drought of these formations(water balance effect).These anthropogenic effects cause a modification with the expansion and increase in the abundance of typical species of other areas of Cerrado and resistant to less saturated soils,leading to the veredas ***,future studies should investigate other veredas on a broader scale to detect changes in the water table that lead to a consequent change in vegetation *** this way,we will have support for the conscientious management and conservatio

Excessive Moderator Band in a Female Cat’s Left Ventricle

This report describes a case of cardiomyopathy associated with excessive moderator bands in a 7 years old cat. In the cat echocardiography examination was observed cardiomegaly, decreased systolic function and thinning of the left ventricle wall. The electrocardiogram showed the occurrence of supraventricular tachycardia. The treatment was initiated with digoxin, furosemide and taurine supplementation, with an improvement of the clinical signs, however the animal died in a fall. The diagnosis of cardiomyopathy associated with excessive moderator band, was based on the multiple cardiac bunds which connect the septum to the free wall of the left ventricle observed in the cat necropsy.

Recent data indicate that IGF1R/IRS signaling is a potential therapeutic target in BCR-ABL1-negative myeloproliferative neoplasms(MPN);in this pathway,IRS2 is involved in the malignant transformation induced by JAK2^(V617F),and upregulation of IGF1R signaling induces the MPN ***157,a synthetic compound designed as an IGF1R-IRS1/2 inhibitor,has been shown to induce antineoplastic effects in solid ***,we aimed to characterize the molecular and cellular effects of NT157 in JAK2^(V617F)positive MPN cell lines(HEL and SET2)and primary patient hematopoietic *** JAK2^(V617F)cell lines,NT157 decreased cell viability,clonogenicity,and cell proliferation,resulting in increases in apoptosis and cell cycle arrest in the G2/M phase(paling,and it activated the AP-1 complex,downregulated four oncogenes(CCND1,MYB,WT1,and NFKB1),and upregulated three apoptotic-related genes(CDKN1A,FOS,and JUN)(pess in a JAK2/STAT-independent ***157 inhibited erythropoietin-independent colony formation in cells from polycythemia vera patients(pese findings further elucidate the mechanism of NT157 action in a MPN context and suggest that targeting IRS1/2 proteins may represent a promising therapeutic strategy for MPN.

<i>Saccharomyces cerevisiae</i>as a Model to Confirm the Ability of FTIR to Evaluate the Presence of Protein Aggregates

It is known that the presence of protein aggregates in biological samples is associated with natural aging processes and age-related diseases. The objective of this technical study was to evaluate the potential of Fourier Transform Infrared Spectroscopy to identify the presence of protein aggregates in Saccharomyces cerevisiae containing high levels of protein aggregates. We acquired ATR-FTIR spectra at mid-infrared range (between 4000 and 600 cm-1) and used multivariate analysis to analyze the data. Significant differences between spectra of wild type and mutant strains in the spectral range assigned to proteins were observed. In particular, an increase in β-sheet structures in mutant strains (spectral signals at 1683 and 1628 cm-1) was observed, indicating the putative presence of protein aggregates. These results prove the capacity of FTIR to evaluate changes in protein conformation, mainly protein aggregation, in a fast, simple and non-expensive way, producing insights on the possible application of this technique to the detection of protein aggregates in human biological samples.

Solidification of Be-free Ni-based dental alloy

A novel, Nb- and Si-rich and Be-free Ni-based alloy was cast by two methods of investment casting and continuouscasting to study the microstructure evolution during solidification and its mechanical properties. The solidification of the alloy startedwith the primary crystallization of FCC-γ, followed by a binary eutectic reaction, with the formation of a heterogeneous constituent：FCC-γ＋G-phase, which replaced the low-melting eutectic （FCC-γ＋NiBe） in the Be-bearing alloys. alNi6Si3 and γ′ formed during theterminal stages of solidification by investment casting, while the formation of alNi6Si3 was suppressed by continuous casting. Thescheil solidification model agreed very well with the experimental results.
