Spray Characteristics of Non-Circular Nozzle in Air-Assisted Injection System

In order to analyze the spray characteristics of non-circular nozzle holes based on the air-assisted spray system, the spray characteristics of circular and non-circular nozzles were studied under the pressure of 0.2-0.6 MPa and the spray volume of 1000-5000 mL/h. Elliptical nozzle and triangular nozzle are classified as non-circular geometries. The spray cone angle was measured by processing the spray image captured by a CCD camera. The measured spray cone angles of the circular nozzles were analyzed, and the axis switching phenomenon of minor plane of elliptical nozzle was found during the test. Among the three shapes of nozzles, the elliptical nozzle had the largest spray cone angle, and the triangular nozzle had the smallest. The velocity field obtained depended on the PIV system. The results show that for axial velocity, elliptical orifice spray has greater kinetic energy and smaller droplet size under the same working parameters. Compared with the circular and elliptical nozzles, triangular orifice reached maximum spray velocity the fastest, but its velocity decay was the fastest. For radial velocity, away from the axis, the spray velocity of the elliptical orifice was less affected by the injection parameters, and the velocity was less than that of circular orifice and triangle orifice. Increasing air pressure will weaken radial propagation. The increase of liquid spraying rate had no remarkable effect on the increase of spraying rate. The results of particle size analysis show that the particle size of the non-circular orifice is reduced compared with that of the circular orifice, which promotes the breakup of droplets to a certain extent and enhances the atomization effect.

Determination of key soil characteristic parameters using angle of repose and direct shear stress test

Discrete element modelling(DEM)is a numerical method for examining the dynamic behavior of granular media.In order to build an accurate simulation model and provide more comprehensive soil characteristic parameters for the design and optimization of various soil contact machinery,in this paper,the discrete element simulation method(EDEM)combined with experimental approach is used to investigate the soil contact characteristic parameters in East Asia.In this study,Hertze-Mindlin(no slip)was used as a particle contact model by taking particle contact parameters and soil JKR(Johnson-Kendall-Roberts)surface energy as determinants,and repose angle,internal friction angle,and cohesive force as evaluation indexes.The method of Plackett-Burman,Stepest ascent,and Box-Behnken were used to gradually reduce the range of parameters needed for simulation until the most accurate value was determined.The results show that the restitution coefficient,static friction coefficient,and rolling friction coefficient between soil particles have significant effects on the DEM model,and their value of them are 0.596,0.725,and 0.16,respectively.Based on these parameters used for the repose angle test and direct shear stress test,the value of repose angle is 31.97°,the internal friction angle is 27.61°,and the cohesive force is 33.06 kPa.The relative errors with the actual measured values are 9.54%,1.87%,and 2.31%,respectively.In order to further test whether the simulation parameters of soil obtained by repose angle test and direct shear stress test are consistent with the real soil,comparison test between field test and discrete element simulation was used.The results show that the error in height of ridge between the simulated soil and the actual soil is 4.06%,which is within the acceptable range.It also indicates that the calibrated and optimized soil simulation model can accurately represent the real soil.The research provides theoretical basis and technical support for the study of soil contact parts by using the discrete element meth

Apoptosis of T-47D Cells Induced by Cinobufacini via a Caspase-3-dependent Manner

To investigate the mechanism of the anti-tumor activity of cinobufacini on the breast cancer cell line T-47D,the inhibitory effect of cinobufacini on the proliferation of T-47D was detected via MTT assay and the morphological changes of T-47D and HBL-100 cells caused by cinobufacini were observed with an inverted microscope.Cell apoptosis and cell cycle stages were detected by flow cytometry analysis.The effects of cinobufacini on the expression of active-form and pro-form of caspase-3 were assessed by Western blot analysis.Cinobufacini dramatically inhibited T-47D proliferation in a dose-and time-dependent manner.We found that more than 20％ of T-47D cells were killed after treatment with 20 mg/mL cinobufacini for 24 h in vitro.After 6 d of treatment with 20 mg/mL cinobufacini,the cell survival rate decreased by more than 40％.Flow cytometric analysis demonstrated that cinobufacini induced significant apoptosis and changes of the cell cycle distribution of T-47D cells.We used breast cell line HBL-100 as the control,the above experiments except cell cycle analysis showed that cinobufacini more obviously induced the apoptosis of T-47D cells than that of HBL-100 cells.Western blot analysis confirmed the protein expression of active caspase-3 increased with increasing the dose of cinobufacini.These results indicate that cinobufacini induces the apoptosis of T-47D cells via the up-regulation of caspase-3.

Calcium requirements for Chinese adults by cross-sectional statistical analyses of calcium balance studies： an individual participant data and aggregate data meta-regression

Background Chinese dietary reference intakes for calcium are largely based on foreign studies. We undertook meta- regression to estimate calcium requirements for Chinese adults derived from calcium balance data in Chinese adults. Methods We searched PubMed, Cochrane CENTRAL, and SinoMed from inception to March 5, 2014, by using a structured search strategy. The bibliographies of any relevant papers and journals were also screened for potentially eligible studies. We extracted a standardized data set from studies in Chinese adults that reported calcium balance data. The relationship between calcium intake and output was examined by an individual participant data （IPD） and aggregate data （AD） meta-regression. Results We identified 11 metabolic studies in Chinese adults within 18-60 years of age. One hundred and forty-one IPD （n=35） expressed as mg/d, 127 IPD （n=32） expressed as mg.kg body wt-l-d-1, and 44 AD （n=132） expressed as mg/d were collected. The models predicted a neutral calcium balance （defined as calcium output （Y） equal to calcium intake （C）） at intakes of 460 mg/d （Y=0.60C＋183.98） and 8.27 mg.kg body wt-l.d-1 （Y=0.60C＋3.33） for IPD, or 409 mg/d （Y=0.66C＋139.00） for AD. Calcium requirements at upper intakes were higher than that at lower intakes in all these models. Conclusion Calcium requirement for Chinese adults 18-60 years of age approximately ranges between 400 mg/d and 500 mg/d when consuming traditional plant-based Chinese diets.

Identification of highly efficacious PROTACs targeting BRD4 against acute myeloid leukemia:Design,synthesis,and biological evaluations

The abnormal activation of BRD4 accelerates the progression of acute myeloid leukemia(AML),developing more precise therapeutics to intervene BRD4 promise to be an excellent opportunity to avoid current limitations of chemotherapy in clinic.Herein,a range of small-molecule PROTACs with the privileged 8-methyl-pyrrolo[1,2-a]pyrazin-1(2H)-one scaffold were rationally designed,which harbored different carbon or ethylenedioxy chains to degrade BRD4 mediated by the E3 ubiquitin ligase CRBN.Among them,the most potential B24 exhibited remarkable BRD4 degradation and excellent anti-proliferative activities in MV4-11 cells,with values of DC_(50)and IC_(50)for 0.75 nmol/L and 0.4 nmol/L,respectively,which were better than the BRD4 inhibitor(+)-JQ-1.Notably,this compound could time-dependently degrade the target protein in the BRD4-,CRBN-,and proteasome-dependent manner.Besides,B24 dramatically decreased the level of proto-oncogene c-Myc,and induced cell apoptosis by arresting the cell cycle in G0/G1 phase,down-regulating Bcl-2 and up-regulating Bax to amplify apoptotic effectors.This proof-of-concept study also highlighted the feasibility of BRD4-based PROTACs as a more powerful strategy against AML.

Environmental deoxyribonucleic acid(eDNA),which includes antibiotic resistance genes,is ubiquitous in the environment.The interactions between eDNA and biochar,a promising material widely used in soil amendment and water treatment,greatly affect the environmental behavior of eDNA.Hitherto few experimental evidences are available yet,especially on the information of thermodynamics and energy distribution to explains the interactions between biochar and eDNA.This study investigated the adsorption of herring sperm DNA(hsDNA)on pine sawdust biochar,with a specific emphasis on the adsorption thermodynamics and site energy distribution.The adsorption of hsDNA on biochar was enhanced by an increase in the pyrolysis and adsorption temperatures.The higher surface area,strongerπ−πinteraction,and weaker electrostatic repulsion between hsDNA and biochars prepared at high pyrolysis temperatures facilitated the adsorption of hsDNA.The thermodynamics indicated that the adsorption of hsDNA on biochar was spontaneous and endothermic.Therefore,higher temperature was beneficial for the adsorption of hsDNA on biochar;this was well explained by the increase in E*and F(E*)with the adsorption temperature.These results are useful for evaluating the migration and transformation of eDNA in the presence of biochar.

Assessing the Dynamics of Grassland Net Primary Productivity in Response to Climate Change at the Global Scale

Understanding the net primary productivity(NPP) of grassland is crucial to evaluate the terrestrial carbon cycle. In this study, we investigated the spatial distribution and the area of global grassland across the globe. Then, we used the Carnegie-Ames-Stanford Approach(CASA) model to estimate global grassland NPP and explore the spatio-temporal variations of grassland NPP in response to climate change from 1982 to 2008. Results showed that the largest area of grassland distribution during the study period was in Asia(1737.23 × 104 km^2), while the grassland area in Europe was relatively small(202.83 × 10~4 km^2). Temporally, the total NPP increased with fluctuations from 1982 to 2008, with an annual increase rate of 0.03 Pg C/yr. The total NPP experienced a significant increasing trend from 1982 to 1995, while a decreasing trend was observed from 1996 to 2008. Spatially, the grassland NPP in South America and Africa were higher than the other regions, largely as a result of these regions are under warm and wet climatic conditions. The highest mean NPP was recorded for savannas(560.10 g C/(m^2·yr)), whereas the lowest was observed in open shrublands with an average NPP of 162.53 g C/(m^2·yr). The relationship between grassland NPP and annual mean temperature and annual precipitation(AMT, AP, respectively) varies with changes in AP, which indicates that, grassland NPP is more sensitive to precipitation than temperature.

An effective way to thermodynamically resolve the cathode irreversibility of Li-O2 battery is the usage of redox mediators（RM）. However, the sluggish reaction between RM and discharge products is still common, which inhibits the efficiency of RM. Here, with the reaction between LiOH and I2 as a model, we attempt to improve the reaction kinetics by unraveling the elementary reaction steps and the solvent effect via the first principles calculation and machine learning. Also, in terms of the debate on the reversibility of LiOH, we have experimentally clarified the particular experimental conditions to generate O2. The reaction processes were calculated for LiOH with various disordering degree, which is quantitatively defined based on the bond length deviation against the perfect crystalline structure. The solvation effects of 28 solvents on the lithium desorption（the rate determined step） were compared. The detailed theoretical and experimental exploration shows that an appropriate H2O content is the key to yielding O2. The discharge products having the higher disordering extent possesses faster reaction kinetics with RM. The functional group of the solvent molecule is proposed as the key to regulating reaction kinetics, as only the atoms involved in the functional group can predict the solvent effect well using the machine learning method. Moreover, the phosphate-based solvent is predicted to promote the LiOH decomposition kinetics most. These findings indicate that the rational discharge product structure regulation and appropriate solvent selection are prospect strategies for the pursuit of faster reaction kinetics between RM and the discharge products.
