Angiogenesis Factors Associated with New Breast Cancer Cell Line AMJ13 Cultured <i>in Vitro</i>

Background: AMJ13 is a new breast cancer cell line that has been established from a 70-year-old Iraqi woman with a histological diagnosis of infiltrating ductal carcinoma. It is the first for an Iraqi population. In breast cancer, angiogenesis provides the tumor tissue, which is rapidly proliferated with oxygen and nutrients, removes wastes and increases the opportunity of cancer cells to invade other organs. Methods: The AMJ13 breast cancer cell line was represented at three different passages and incubated for interval times. Microarray panel of 43 different angiogenesis markers was used to scan the supernatant for the factors. ELISA was used to quantify some of the important angiogenesis factors released in the culture medium and to confirm absence of those who was not detected by the antibody array. RT-PCR was used to confirm the gene expression (mRNA) of studied factors. Results: Microarray analysis showed that TIMP1 and two secreted at highest levels compared to the rest of the factors with low presence of endostatin. Other non-detectable factors by microarray examined by ELISA assay that showed highest expression level of VEGF-A were obtained at earliest passage, while the highest levels of FGF-b were obtained at late passage. The VEGF-D secretion was shown low concentrations at all studied passages. There is no detectable level of EGF protein in different passages and times interval tested. There are no significant differences in secretion of sICAM between different passages and incubation periods. Conclusion is that AMJ13 cell line depends on VEGF-A as main angiogenesis factor to induce micro-vessels supported by low levels of VEGF-D for lymphatic vessels formation. AMJ13 cell line depends on FGF as growth factors as in late passages it was shifted to depend mainly on FGF completely. all of this process may be regulated by TGF-β. Value-Based Test Case Prioritization for Regression Testing Using Genetic algorithms

Test Case Prioritization(TCP)techniques perform better than other regression test optimization techniques including Test Suite Reduction(TSR)and Test Case Selection(TCS).Many TCP techniques are available,and their performance is usually measured through a metric Average Percentage of Fault Detection(APFD).This metric is value-neutral because it only works well when all test cases have the same cost,and all faults have the same *** APFD for performance evaluation of test case orders where test cases cost or faults severity varies is prone to produce false ***,using the right metric for performance evaluation of TCP techniques is very important to get reliable and correct *** this paper,two value-based TCP techniques have been introduced using Genetic algorithm(GA)including Value-Cognizant Fault Detection-Based TCP(VCFDB-TCP)and Value-Cognizant Requirements Coverage-Based TCP(VCRCB-TCP).Two novel value-based performance evaluation metrics are also introduced for value-based TCP including Average Percentage of Fault Detection per value(APFDv)and Average Percentage of Requirements Coverage per value(APRCv).Two case studies are performed to validate proposed techniques and performance evaluation *** proposed GA-based techniques outperformed the existing state-of-the-art TCP techniques including Original Order(OO),Reverse Order(REV-O),Random Order(RO),and Greedy algorithm.

BM-MSCs Differentiated Insulin-Producing Cells Produce More Insulin in Presence of EGF than of FGF

Adult stem cells have the ability to differentiate into islet like cells for the hope of treating diabetes mellitus (DM). The most important part is the differentiation process from na?ve stem cells to fully differentiated fully functional islet cells. For this purpose, we wanted to optimize the current proposed differentiation media by replacing the FGF with EGF and measure insulin production. Bone marrow-derived MSCs were from mice long bones and expanded in cell culture before induction of differentiation. Stem cells surface markers were analyzed by immunocytochemistry. Cultured stem cells were negative for CD34 while they expressed high levels of CD90. Differentiated cells morphology was studied by using H & E stain. Differentiated cells were detected by studying protein expression of insulin as specific marker for IPC differentiation. Cells function was studied by measuring the insulin production in tissue culture supernatant in vitro and also insulin release in response to glucose challenge. Ditizone staining were both positive. Insulin was secreted by these cells in response to different concentrations of glucose stimulation in a regulated manner. Cells induced with formula contain EGF produced more insulin in the same formula but contain FGF instead, this prove that EGF is the best to use during differentiation process.

Comparison of Shisha Smoking Behavior among Medical and Pharmacy Students

Background: Health care workers are respected in every society and have a strong influence on the behaviour of the community. In order to reduce the shisha dependence in the Pakistani population, it is necessary to evaluate the perspective of future doctors and pharmacists regarding this addiction as they become role models for the future generations. Objective: The aim is to compare the knowledge, attitudes, and practices of shisha use among medical students and pharmacy students in order to assess their support in the anti-shisha campaigns of Pakistan. Methodology: Cross sectional study was performed in four major medical universities of Karachi, Pakistan through a modification of the WHO global health survey questionnaire. Result: 398 medical students and 470 pharmacy students were included in the study which showed that currently 9.6% pharmacy students and 4.8% medical students used shisha daily (p = 0.000). Pharmacy students started shisha use in less than 15 years of age while this number was 29.5% among medical students (P = 0.000). Moreover, 44.6% pharmacy students and 50% medical students were not planning to quit this addiction within the next six months. 14% of pharmacy students and 20.4% in medical students (P = 0.002) were unaware that passive smoking caused lung cancer, while 23.4% pharmacy students and 33.5% medical students rejected it as cause of heart diseases (p = 0.006). Conclusion: A significant number of medical and pharmacy students were unaware about some of the serious consequences of shisha smoking. Therefore, both groups of students need to be educated about the health hazards of shisha use, in order to control this growing menace.

Sensitivity and Specificity of Various Serological Tests for Detection of Brucella spp. Infection in Male Goats and Sheep

Brucellosis is a zoonosis found throughout the world with major implications both in the field of public health and farming economy. It is most often caused by Brucella abortus and Brucella melitensis. This study aimed for evaluation of the sensitivity and specificity of immunochromatographic assay (ICA) and comparing the results with those obtained from Rose Bengal Plate Test (RBPT) and in direct Enzyme Linked Immunsorbent Assay (ELISA) for serodiagnosis of sheep and goat brucellosis. Out of 41 sera samples (21 ram and 20 male goats) which were examined, antibodies against Brucella were detected in ram 8 (38.09%), 6 (28.57%) and 16 (76.19%) in ram samples, whereas they were detected in 3 (15%), 1 (5%) and 12 (60%) serum sample of goats males by RBPT, (ICA) and iELISA respectively. The kappa value between iELISA and ICA was -0.08 and that between RBPT and ELISA was -0.01 in ram and the kappa value between iELISA and ICA was -0.06 and that between RBPT and ELISA was -0.14 in male goats. The sensitivity for RBPT and ICA were 37.5% and 25% while specificity was 60% and 60% in ram;whereas, in male goats, sensitivity for RBPT was 8.33% and ICA was 8.33% while specificity was 75% and 100% respectively. It can be concluded that the developed ICA is immunodiagnostic assay, and it is rapid, non-expensive, economical and suitable for large-scale screening in developing countries and rural areas.

Combined anterior and posterior ring fixation versus posterior ring fixation alone in the management of unstable Tile B and C pelvic ring injuries:A randomized controlled trial
