Suppression of monosodium urate crystal-induced inflammation by inhibiting TGF-β-activated kinase 1-dependent signaling:role of the ubiquitin proteasome system

Monosodium urate(MSU)crystals activate inflammatory pathways that overlap with interleukin-1β(IL-1β)***,the post-translational mechanisms involved and the role of signaling proteins in this activation are *** the present study,we investigated the intracellular signaling mechanisms involved in MSU-induced activation of THP-1 macrophages and human nondiseased synovial fibroblasts(NLSFs)and the in vivo efficacy of an inhibitor of tumor grovArth factor-β(TGF-β)-activated kinase 1(TAK1),5Z-7-oxozeaenoI,in MSU-induced paw inflammation in C57BL76 ***-1 macrophage activation with MSU crystals(25-200 ng/ml)resulted in the rapid and sustained phosphorylation of interleukin-1 receptor-activated kinase 1(IRAK1 Thr^(209))and TAK1(Thr^(184/187))and their association with the E3 ubiquitin ligase *** the cellular level,MSU inhibited the deubiquitinases A20 and UCHL2 and increased 20s proteasomal activity,leading to a global decrease in K^(63)-linked ubiquitination and increase in K^(48)-linked ubiquitination in THP-1 *** MSU did not stimulate cytokine produaion in NLSFs,it significantly amplified IL-1β-induced IL-6,IL-8,and ENA-78/CXCL5 *** studies and MD simulations followed by TAK1 in vitro kinase assays revealed that uric acid molecules are capable of arresting TAK1 in an active-state conformation,resulting in sustained TAK1 kinase ***,MSU-induced proinflammatory cytokine production was completely inhibited by 5Z-7-oxozeaenol but not IRAK1/4 or TRAF6 *** of 5Z-7-oxozeaenol(5 or 15 mg/kg;orally)significantly inhibited MSU-induced paw inflammation in C57BL/6 *** study identifies a novel post-translational mechanism of TAK1 activation by MSU and suggests the therapeutic potential of TAK1 in regulating MSU-induced Inflammation.

Recovery of some valuable elements from lamprophyre dykes in the Abu Rusheid area, Southeastern Desert, Egypt

In the Abu Rusheid area, Southeastern Desert of Egypt, polymineralized (HREEs, U, Zn and Pb) lam-prophyre dykes are recently recorded. The lamprophyre dykes cut cataclastic rocks along share zones in NNW-SSE trends. The secondary U-minerals are essentially represented by uranophane, kasolite, torbernite and autunite. Ex-perimental work was carried out on the lamprophyre dykes for the recovery of uranium and other valuable elements (e.g. Zn, Pb and Al) from 50 kg of technological sample. The optimum conditions were: 100 g/L sulfuric acid con-centration, 1:5 S/L (solid/liquid) and soaking for 15 days at room temperature. The leaching efficiencies for the re-covery of uranium and other elements like Al, Zn and Pb were 40, 33.6, 98.8 and 78, respectively.

Distribution of Different organotin and organolead Compounds in Sediment of Suez Gulf

organotin and organolead compounds were determined in sediments of the Suez Gulf The concentrations of Tributyltin (TBT) ranged from 0.27 to 2.77 with an average value of 1.37 μgg-1;dry wt. However, the concentrations of dibutyltin (DBT) ranged from 0.07 to 2.27 with an average value of 0.58 μgg-1;dry wt. A significant correlation was found between TBT and DBT with r = 0.82, (p = 0.05) indicating that the occurrence of DBT is mainly related to the degradation of TBT. Generally, the high concentration of TBT was attributed to shipping activity in harbours. In addition, Diphenyltin (DPhT) concentrations ranged from not detected to 2.09 with an average of 1.10 μgg-1 dry wt. Antifouling agents, industrial discharge and the influence of sewage discharge are the main sources of pollution by DPhT compounds in Suez Gulf. on the other side, organolead (oLC) concentrations ranged from 10.88 - 440.2 with an average of 168.7 ngg-1;dry wt. A significant setting of oLC recorded in sediments of Suez Gulf was mainly attributed to cars exhaust and/or spelling and direct evaporation of fuels.

Chemodynamic therapy(CDT)based on cascade catalytic nanomedicine has emerged as a promising cancer treatment ***,most of the reported cascade catalytic systems are designed based on symmetric-or co-assembly of multiple catalytic active sites,in which their functions are difficult to perform independently and may interfere with each *** in cascade catalytic system that involves fragile natural-enzymes,the strong oxidation of free-radicals toward natural-enzymes should be carefully considered,and the spatial distribution of the multiple catalytic active sites should be carefully organized to avoid the degradation of the enzyme catalytic ***,a spatially-asymmetric cascade nanocatalyst is developed for enhanced CDT,which is composed by a Fe_(3)o_(4)head and a closely connected mesoporous silica nanorod immobilized with glucose oxidase(mSio_(2)-Gox).The mSio_(2)-Gox subunit could effectively deplete glucose in tumor cells,and meanwhile produce a considerable amount of H_(2)o_(2)for subsequent Fenton reaction under the catalysis of Fe_(3)o_(4)subunit in the tumor *** the advantage of the spatial isolation of mSio_(2)-Gox and Fe_(3)o_(4)subunits,the catalysis of Gox and freeradicals generation occur at different domains of the asymmetric nanocomposite,minimizing the strong oxidation of free-radicals toward the activity of Gox at the other *** addition,direct exposure of Fe_(3)o_(4)subunit without any shelter could further enhance the strong oxidation of free-radicals toward ***,compared with traditional core@shell structure,the long-term stability and efficiency of the asymmetric cascade catalytic for CDT is greatly increased by 138%,thus realizing improved cancer cell killing and tumor restrain efficiency.

A genetic algorithm-determined deep feedforward neural network architecture(GA-DFNN)is proposed for both day-ahead hourly and week-ahead daily electricity consumption of a real-world campus building in the United *** to the comprehensive relationship between affecting factors and real-world building electricity consumption,the adoption of multiple hidden layers in the deep neural network(DFNN)algorithm would improve its prediction *** architecture of a DFNN model mainly refers to its quantity of hidden layers,quantity of neurons in the hidden layers,activation function in each layer and learning process to obtain the connecting *** optimal architecture of DFNN model was generally determined through a trial-and-error process,which is an exponential combinatorial problem and a tedious *** address this problem,genetic algorithm(GA)is adopted to automatically design an optimal architecture with improved generalization *** year and six months of measurement data from a campus building is used for training and testing the proposed GA-DFNN model,*** demonstrate the effectiveness of the proposed GA-DFNN prediction model,its prediction performance,including mean absolute percentage error,coefficient of determination,root mean square error and mean absolute error,was compared to the reference feedforward neural network models with single hidden layer,DFNN models with other architecture,random search determined DFNN model,long-short-term-memory model and temporal convolutional network *** comparison results show that the proposed GA-DFNN predictive model has superior performance than all the reference prediction models,demonstrating the optimization effectiveness of GA and the prediction effectiveness of DFNN model with multiple hidden layers and optimal architecture.

Microscopic model for chemical etchability along radiation damage paths in solids

It would be very interesting to develop a picture about removal of atoms from the radiation damaged paths or latent nuclear tracks and undamaged bulk material in track detectors. Here, theory of chemical etching is described briefly and a new model for chemical etching along radiation damaged paths in solids is developed based on basic scientific facts and valid assumptions. Dependence of chemical etching on radiation damage intensity and etching conditions is discussed. A new parameter for etching along radiation damaged paths is introduced, which is useful for investigation of relationship between chemical etchability and radiation damage in a solid. Results and discussion presented here are also useful for further development of nuclear waste immobilization.

Prognostic value of pre-treatment F-18-FDG PET-CT in patients with hepatocellular carcinoma undergoing radioembolization

AIM To evaluate the value of pre-treatment 18F-FDG PET/CT in patients with HCC following liver *** We identified 34 patients with HCC who underwent an FDG PET/CT scan prior to hepatic radioembolization at our institution between 2009 and 2013. Patients wereseen in clinic one month after radioembolization and then at 2-3 mo intervals. We assessed the influence of FDG tumor uptake on outcomes including local liver control(LLC), distant liver control(DLC), time to distant metastases(DM), progression free survival(PFS) and overall survival(oS).RESULTS The majority of patients were males(n = 25, 74%), and had Child Pugh Class A(n = 31, 91%), with a median age of 68 years(46-84 years). FDG-avid disease was found in 19(56%) patients with SUVmax ranging from 3 to 20. Female patients were more likely to have an FDG-avid HCC(P = 0.02). Median follow up of patients following radioembolization was 12 months(1.2-62.8 mo). FDG-avid disease was associated with a decreased 1 year LLC, DLC, DM and PFS(P or LLC, DLC, and PFS(all P oNCLUSIoN In this retrospective study, pre-treatment HCC FDGavidity was found to be associated with worse LLC, DLC, and PFS following radioembolization. Larger studies are needed to validate our initial findings to assess the role of F-18-FDG PET/CT scans as biomarker for patients with HCC following radioembolization.
