Treatment of cancer with chemotherapy often faces drug resistance issues, which can stop after a period of remission. This resistance is due to several mechanisms;among them, the most important is the one in relation with the overexpression of the P glycoprotein (Pgp) produced by the mdr 1 gene. This gene confers to cells the multidrug resistance (MDR) phenotype in the setting of treatment for many cancers (leukemia-lymphoma...). The aim of this study was to investigate the binding site of P glycoprotein by affinity labeling in order to synthesize effective inhibitors on chemotherapeutic drugs efflux. The first objective was to increase P glycoprotein concentration in R7 cells by treating them with doxorubicin to obtain human Pgp rich fractions. The second consisted in characterizing the progesterone binding site on Pgp by affinity photolabeling with tritiated azidopine on the one hand, and by western-blot transfer with C219 antibody on the other hand. Our results indicate a significant increase in the level of expression Pgp in R7 cells treated with 1 and 2.5 nM doxorubicin, and clearly show enrichment in Pgp of doxorubicin treated R7 cells.

Many surgeons practice prophylactic drainage after cholecystectomy without reliable evidence,this study was conducted to answer the question whether to drain or not to drain after cholecystectomy for acute calculous cholecystitis.A retrospective review of all patients who had cholecystectomy for acute cholecystitis in Aseer Central Hospital,Abha,Saudi Arabia,was conducted from April 2010 to April *** were extracted from hospital case *** data included clinical presentation,routine investigations and liver function *** data included excessive adhesions,bleeding,bile leak,and drain *** cases such as pericholecystic collections,mucocele and empyema were also *** who needed therapeutic drainage were *** data included hospital stay,volume of drained fluid,time of drain removal,and drain site *** study included 103 patients allocated into two groups;group A（n = 38） for patients with operative drain insertion and group B（n = 65） for patients without drain *** number of patients with preoperative diagnosis of acute non-complicated cholecystitis was significantly greater in group B（80%） than group A（36.8%）（P 〈 0.001）.Operative time was significantly longer in group *** patients who were converted from laparoscopic to open cholecystectomy were in group *** analysis revealed that hospital stay was significantly（P 〈 0.001） longer in patients with preoperative *** was no added benefit for prophylactic drain insertion after cholecystectomy for acute calculous cholecystitis in non-complicated or in complicated cases.

AIM:To assess the effectiveness of hepatitis B virus(HBV) vaccination program among fully vaccinated ***:A national community based crosssectional study was carried out in 6 governorates representing Egypt. A total of 3600 children aged from 9 mo to 16 years who were fully vaccinated with HBV vaccine during infancy were recruited. Face to face interviews were carried out and sera were evaluated for hepatitis B surface antigen(HBsA g),anti-HBV core antibodies(total) and quantitative detection of hepatitis B surface antibody using enzyme linked immunoassays techniques. Samples positive to HBs Ag/anti-HBV core antibodies were subjected to quantitative HBV-DNA detection by real time polymerase chain reaction with 3.8 IU/L detection limit. RESULTS:Sero-protection was detected among 2059 children(57.2%) with geometric mean titers 75.4 ± 3.6 IU/L compared to 3.1 ± 2.1 IU/L among nonseroprotected children. Multivariate logistic analysis revealed that older age and female gender were the significant predicting variables for having non seroprotective level,with adjusted odds ratio 3.3,9.1and 14.2 among children aged 5 to ely compared to those ely. The prevalence of HBV infection was significantly higher among children aged ≥ 7 years(0.59%) compared to 0.07% among younger children with odds ratio equal to 8.4(95%CI:1.1-64.2) and P 0.05. C ON C LU S I ON :T he E gy pt ian c ompuls or y H B V vaccination program provides adequate protection. Occult HBV infection exists among apparently healthy vaccinated children. Adherence to infection control measures is mandatory.

Colorectal cancer (CRC) is significant publichealth problem in rich countries of the world, because of its frequency and mostly because of its severity. Our aim was to improve histologic diagnosis of malignancy with topographic coloration (HE) and cytokeratins 7, 20 immuno-labeling. The risk for colorectal cancer incease with age, but in our series, 10% of colorectal cancer exist in patients younger than 40 years old. In histological study, adenocarcinoma represented 100% and the evaluation for stage of disease showed that the most dominant isstage IV (32.6%). In our trial, we found that cytokeratins 7 and 20 immunoreactivity in colorectal adenocarcinoma are often positive for cytokeratin-20 and most of time negative for cytokeratin-7.

AIM: To investigate the effect of microR NA on insulinlike growth factor binding protein-3(IGFBP-3) and hence on insulin-like growth factor-Ⅱ(IGF-Ⅱ) bioavailability in hepatocellular carcinoma(HCC).METHODS: Bioinformatic analysis was performed using ***, DIANA lab and Segal lab softwares. Total RNA was extracted from 23 HCC and 10 healthy liver tissues using mir Vana mi RNA Isolation Kit. microR NA-17-5p(miR-17-5p) expression was mimicked and antagonized in Hu H-7 cell lines using Hi Per Fect Transfection Reagent, then total RNA was extracted using Biozol reagent then reverse transcribed into cD NA followed by quantification of mi R-17-5p and IGFBP-3 expression using Taq Man real-time quantitative PCR. Luciferase reporter assay was performed to validate the binding of miR-17-5p to the 3'UTR of IGFBP-3. Free IGF-Ⅱ protein was measured in transfected Hu H-7 cells using IGF-Ⅱ elISA kit. RESULTS: Bioinformatic analysis revealed IGFBP-3 as a potential target for miR-17-5p. Screening of miR-17-5p and IGFBP-3 revealed a moderate negative correlation in HCC patients, where mi R-17-5p was extensively underexpressed in HCC tissues(P = 0.0012), while IGFBP-3 showed significant upregulation in the same set of patients(P = 0.0041) compared to healthy donors. Forcing mi R-17-5p expression in Hu H-7 cell lines showed a significant downregulation of IGFBP-3 mR NA expression(P = 0.0267) and a significant increase in free IGF-Ⅱ protein(P = 0.0339) compared to mock untransfected cells using unpaired t-test. Luciferase assay validated IGFBP-3 as a direct target of mi R-17-5p; luciferase activity was inhibited by 27.5% in cells co-transfected with miR-17-5p mimics and the construct harboring the wild-type binding region 2 of IGFBP-3 compared to cells transfected with this construct alone(P = 0.0474).CONCLUSION: These data suggest that regulating IGF-Ⅱ bioavailability and hence HCC progression can be achieved through targeting IGFBP-3 via manipulating the expression of miR NAs.

Evaluation of Marwit Rod el Leqah deposit as pegmatite high purity quartz source for advanced applications was conducted through the geological, mineralogical and geochemical studies. elemental analysis of a representative quartz sample showed that the quartz deposits were classified as medium purity quartz with 99.575% average SiO2 content and about 0.422% (4220 ppm) of accompanied mineral. Field study and petrography investigation using optical polarized transmitted microscope and reflected light microscope revealed that these impurities were mainly found as solid particulates of iron oxide, muscovite, rutile, orthoclase, granitic rafts, and as fluid inclusions as well. Moreover, structural undesirable trace elements were also presented in the quartz sample.

Appropriate knowledge of the parental cultivars is a pre-requisite for a successful breeding *** study characterized fruit yield,quality attributes,and molecular variations of ten tomato cultivars during three consecutive generations under greenhouse *** 86,Castle Rock,and Red Star cultivars showed the highest fruit yield(kg/plant),total phenolic compounds(TPC),and sap *** component analysis categorized the evaluated fruit yield into three groups based on their quality attributes.A robust positive correlation appeared among traits inside each group.A positive correlation was likewise noticed between the first and the second ***,a negative correlation was detected between the first,the second and the third *** profiling,using seven inter-simple sequence repeat(ISSR)primers,produced 60 loci,including 49 polymorphic *** molecular analysis also pinpointed the highest genetic similarity(0.92)between P73 and Moneymaker,while the lowest genetic similarity(0.46)was observed between Castle Rock and *** cultivars P73 and Moneymaker showed the lowest genetic distance(2.24),while the highest genetic distance(5.92)was observed between Super Marmand and Peto86,on the one hand,and between Castle Rock and Moneymaker,on the other *** chemical analysis of fruit sap indicated the highest levels of TPC,total flavonoids,anthocyanin,ascorbic acid and total soluble solids in Peto 86 and Castle Rock *** analysis of tomato cultivars based on morphological and molecular attributes indicated four distinct *** 86,Castle Rock,and Red star cultivars can be recommended for the tomato hybridization breeding programs in the future,with other tomato cultivars as potentially high-yielding parents.
